Abstract

La présente invention concerne le diagnostic sur ADN et ARN basé sur la spectrométrie de masse, par exemple, la spectrométrie de masse à désorption-ionisation par impact laser assistée par matrice et saisie en vol (MALDI-TOF), la méthode d'ES-MS, la spectrométrie de masse à résonance ionique cyclotronique (ICR), la spectrométrie de masse par transformation de Fourier, ou des combinaisons de celles-ci, dans lesquelles des sondes d'ADN modifiées partiellement par des analogues nucléosidiques bloqués sont utilisées afin d'augmenter la stabilité et la résolution. L'invention concerne notamment un procédé de détection d'une séquence d'acide nucléique cible d'une molécule d'acide nucléique ou de détection d'une mutation dans une séquence d'acide nucléique d'une molécule d'acide nucléique, dans lequel (a) la molécule d'acide nucléique ou (b) une partie de la molécule d'acide nucléique ou (c) un oligonucléotide complémentaire de la séquence ou au moins une sous-séquence de la molécule d'acide nucléique est analysée par spectrométrie de masse afin d'obtenir des informations directes ou indirectes concernant ladite séquence ou mutation d'acide nucléique, le procédé consistant en l'hybridation d'un oligonucléotide modifié par LNA à la molécule d'acide nucléique.
