Abstract

L'invention concerne une méthode permettant de faciliter la recombinaison homologue dirigée vers un site dans un organisme en vue de produire des mutants, et consistant à: 1) construire une bibliothèque de grand vecteur d'insertion comprenant au moins un grand vecteur d'insertion, 2) construire un second vecteur présentant un élément transposable, 3) incuber le produit de l'étape 1) avec le produit de l'étape 2) en présence d'une transposase spécifique des séquences nucléotidiques répétées inversées sur le vecteur de plasmide, 4) amplifier le grand vecteur d'insertion dissocié résultant de l'étape 3, 5) introduire le grand vecteur d'insertion dissocié produit à l'étape 4) dans une cellule hôte cible, et 6) sélectionner systématiquement une recombinaison homologue réussie produite par à l'étape 5) avec le second marqueur sélectionnable.
