Abstract

Die Erfindung betrifft ein Verfahren zur Synthese und Amplifikation von Nukleinsäuren mittels einer Enzym-katalysierten Reaktion, bei der eine Nukleinsäure ausgehend von einer einzelsträngigen Initiatornukleinsäure kopiert wird, ohne daß eine Hybridisierung der Initiatornukleinsäure an die zu kopierende Nukleinsäure stattfindet. Die Erfindung betrifft somit ein Verfahren, bei dem eine lineare Nukleinsäure ausgehend von einer zumindest am 3'-Ende einen einzelsträngigen Bereich aufweisenden Initiatornukleinsäure durch die Aktivität einer Polymerase kopiert wird, wobei die Initiatornukleinsäure keine Homologie mit der zu kopierenden Nukleinsäure aufweisen muß.
