Abstract
The invention relates to the process for production of recombinant human c-peptide, comprising cultivation of strain producer Escherichia coli, destruction of bacterial cells by disintegration, separation of inclusion bodies containing hybrid protein, theirs solubilisation in buffer solution containing carbamide and dithiothreitol, renaturation and purification of hybrid protein, its degradation with trypsin and carboxypeptidase B, which is carried out simultaneously, wherein tryptic cleavage products are separated with chromatography on SP- sepharose, balanced with 0.03-0.1 M of ammonium-acetate buffer pH 3.6, containing 3 M of carbamide, eluated with linear gradient of potassium chloride from 0 to 0.5 M in initial buffer, with subsequent purifying and receiving of desired product.
