The useful model refers to pharmacy, in particular to the methods of obtaining complexes of biologically active 2 substances from medicinal raw materials, namely the extract from the leaves of cultivated grapes, preferably the Isabella variety, with pronounced hepatoprotective activity and can be used as an active active substance in the preparation of medicinal products in various medicinal forms.
A known method of obtaining a complex of biologically active substances that has a hepatoprotective effect (Patent 30879A, Ukraine, Ab1KZ38/43, Ab1KOU/20, application 06.16.1998, publ. 12.15.2000, Bull. Mo7)|, which consists of 70 extraction of pea grass with ethanol, evaporation of the extract to an aqueous residue, followed by purification with methanol chloride.
Obtaining the product by the specified method requires the use of such a toxic solvent as methanol chloride, and, as a result, additional equipment and the creation of special conditions for the implementation of the method. 12 The method of obtaining the polyphenolic complex "Flavitin" with anti-inflammatory, analgesic, anti-ulcer and antioxidant activity is the closest to the claimed one. (Patent 59681A, Ukraine, Ab1KZ5/78, application 11/15/2002, published 09/15/2003, Byul.Mo9|, which consists in the extraction of crushed grape leaves (preferably the Dabugi variety) 5095 with ethyl alcohol at a raw material:extractant ratio of 1 :10, the obtained extract is evaporated to remove ethyl alcohol, the aqueous residue is filtered from the sediment, the sediment is treated with water for resorption of phenolic compounds, the resulting aqueous solution of the latter is combined with the filtrate, evaporated and dried.
The disadvantages of the method according to the prototype include the need to use additional resorption of the sediment after filtering the extract, the low yield of the finished product and the use of mainly grape leaves of the variety
оDabugi, which is uncommon on the territory of Ukraine. 29 The task of the useful model is to create a new method of obtaining a means with hepatoprotective activity, pt"), in which by using grape leaves of domestic varieties as raw materials, carrying out exhaustive extraction of biologically active substances (BAR) with the given parameters of the method, a means with pronounced hepatoprotective activity is obtained, which can be used as an active substance in the manufacture of medicines in various dosage forms, and the method itself is economical and easy to perform. at
Поставлене завдання вирішується таким чином, ідо у способі одержання засобу з гепатопротекторною «І активністю, який включає екстракцію подрібненого листя винограду етанолом, фільтрування та упарювання, корисною моделлю передбачено, що в якості сировини використовують листя винограду, переважно сорту -The task is solved in this way, i.e. in the method of obtaining a product with hepatoprotective activity, which includes the extraction of crushed grape leaves with ethanol, filtering and evaporation, a useful model provides that grape leaves are used as raw materials, mainly of the variety -
--Isabella, the extraction is carried out with 39-4195 ethanol at a ratio of raw materials to the extractant as 1:12 and successively three times for 12 hours, and the obtained extracts are combined, filtered and evaporated to the state of a thick extract. --
The predominant use of grape leaves of the Isabella variety as raw materials is caused by the fact that the research conducted by the authors showed that among various cultivated grape varieties, the Isabella variety is characterized by a high concentration of BAR in the leaves with hepatotector activity, which makes their "extraction" industrially expedient and ensures a high yield of the final product . At the same time, the domestic raw material base of this Z 70 grape variety is sufficient for industrial production. c In the course of the experiments, it was determined that the optimal extractant for obtaining the agent c
39-41905.Ethanol in a concentration of 39-41905 has hepatoprotective activity in accordance with the claimed method.
When the concentration of ethanol is reduced below 3995, the maximum extraction of the BAR complex from the raw material is not achieved. Increasing the concentration of ethanol above 4195 is impractical, because it does not lead to a significant increase in BAR output. - The expediency of using the raw material:extractant ratio of 1:12 in the claimed method has been proven experimentally and takes into account the water absorption characteristics of the raw material.
When the ratio of raw materials: extractant was reduced, a decrease in the yield of the main groups of BAR 7 was observed due to incomplete wetting of the raw materials. "iz" 20 When the raw material:extractant ratio increased, no significant changes were observed in the qualitative composition of the final product, however, the costs of the extractant and energy costs for evaporating the obtained liquid extract to a thick state increased.
In order to ensure the exhaustive extraction of BAR from the raw material by the claimed method, it is provided for the extraction to be carried out sequentially in the C state for 12 hours each, with the subsequent combination of the obtained extracts. 29 The finished product is obtained in the form of a thick extract with a dry residue content of at least 7095. Obtaining from a dry extract is impractical because it is not stable during storage due to high hygroscopicity.
The product obtained by the claimed method is a thick extract of a dark brown color, acidic in taste, with a specific pleasant smell, containing at least 1.595 oxycinnamic acids (calculated per chlorogenic acid), 2.295 flavonoids (calculated per hyperoside), 10.595 tannins substances 60 The claimed method is carried out as follows: crushed grape leaves, preferably of the Isabella variety, are subjected to extraction with 39-4195 ethanol at a ratio of raw materials: extractant of 1:12, the extraction is carried out sequentially in three stages of 12 hours, each time with new portions of the extractant, the obtained extracts are combined , filtered and evaporated to obtain a thick extract.

Determination of the hepatoprotective activity of grape leaf extract, obtained by the claimed method, was carried out in experiments on rats on the model of acute tetrachloromethane hepatitis. Acute toxic hepatitis was reproduced by a single intragastric administration of 5095 oily solution of tetrachloromethane in a 7/0 dose of T1ml/100g of body weight. The animals were divided into 6 experimental groups: an intact control group; control pathology; animals of the third - fifth groups against the background of tetrachloromethane damage to the liver were injected with extract from grape leaves in doses of 25, 50 and 10 Ω/kg, respectively, animals of the sixth group, along with tetrachloromethane, received silibor in a therapeutic dose of 25 mg/kg as a reference drug. Grape leaf extract and the comparator drug were administered one hour before and two hours after tetrachloromethane administration. The conclusion about /5 The expression of hepatoprotective activity was made according to the degree of reduction of the liver mass coefficient (MCF), the content of products of lipid peroxidation: malondialdehyde (MDA) and diene conjugates (DC) in the liver tissue, as well as a decrease in the activity of markers in the blood serum cytolysis - alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and cholestatic syndrome marker - alkaline phosphatase (AL). Biochemical indicators of the liver and blood serum of experimental animals were determined 18 hours after the introduction of 5075 oil solution of tetrachloromethane. The results of the conducted research are shown in Table 1. compared with silyboron.
Indicator Intact control / Control for pathology Silibor 25mg/kg ssh yu yu y yu yu 3o " - the difference is reliable in relation to the intact (p 20.05); -- их - the difference is reliable in relation to the control pathology (p 0.05) . and - the difference is significant in relation to the refereno-preparation (p -0.05). " 70 As can be seen from Table 1, the introduction of grape leaf extract against the background of acute toxic hepatitis did not lead to regressive changes in the development of pathology. In the conditions of the model pathology, grape leaf extract showed pronounced antioxidant properties and reliably reduced the content of MDA and diene conjugates in liver tissue at all tested doses. At a dose of 10 Ωg/kg, grape leaf extract exerted a significantly more pronounced inhibitory effect on the accumulation of MDA in liver tissue than the comparison drug and reduced the DC content almost to -75% of the level of the intact control.
The introduction of grape leaf extract led to a decrease in cytolysis, which appeared to be reliable with a decrease in the activity of ALT and AST in blood serum when using grape leaf extract in doses of 50 -1 and 1 bOmg. The introduction of grape leaf extract led to a significant decrease in the activity of LF in blood serum, which is evidence of a decrease in cholestatic syndrome.
50 In all studied doses, the extract from grape leaves, obtained according to the stated method, showed hepatoprotective activity no worse, and at a dose of 10 Омг/kg it was higher than the activity of the comparison drug - silibor.
