Abstract
本發明公開了HLA-A11限制性乙型肝炎病毒HBc141-151
表位肽的T細胞受體及其應用。該T細胞受體包含α鏈和β鏈;α鏈包含三個互補決定區,氨基酸序列分別如SEQ ID No. 2的第48-53位、第71-77位和第112-121位所示;β鏈包含三個互補決定區,氨基酸序列分別如SEQ ID No. 4的第46-50位、第68-73位和第111-122位所示。實驗證明,該T細胞受體不僅具有HBV多肽表位依賴性的活化和增殖能力,而且體內和體外均具有良好的殺傷靶細胞的活性,還能夠有效清除體內HBV的慢性感染。此發明具有很重要的應用價值。

本發明係關於一種HLA-A11限制性乙型肝炎病毒HBc141-151
表位肽的T細胞受體及其應用。
肝細胞癌(Hepatocellular Carcinoma,HCC)簡稱肝癌,其臨床表現隱匿,早期缺乏典型癥狀,儘管近年來肝癌的診斷和治療有了一定的進展,其預後仍然不理想,五年生存率極低。究其原因,一方面是肝癌對目前化療藥物不敏感,大多數肝癌患者缺乏有效的治療方法;另一方面,肝癌往往在疾病的晚期才被診斷,這就排除了局部消融這些原本可以改善病人病情的方法。目前,手術切除與肝移植仍然是治療肝癌最有效的方法。但是肝癌發展到一定階段,腫瘤細胞可能轉移到其它器官(如肺部、骨骼、腦等),行肝移植前,現階段的檢查檢測不到,術後因免疫抑制狀態,潛伏在其它器官的微病灶可能導致肝癌復發,患者術後存留期不理想。因此,極需找尋新的有效輔助治療方案。
免疫細胞治療技術是生物醫藥領域中發展最為迅速的板塊。2013年,科學轉化醫學(Science-Translational Medicine)雜誌預測,細胞治療將成為"未來醫學第三大支柱"。2017年10月以來,美國FDA相繼批准諾華與KITE公司的CAR-T藥物上市,開創免疫細胞治療藥物的新時代,在臨床治療中展現出廣闊的應用前景。在實體腫瘤免疫細胞治療領域,理想的目標抗原是僅在腫瘤細胞表面表達的腫瘤特異性抗原。不幸的是腫瘤表達的大多數抗原不具備腫瘤特異,因此大多數的CAR-T、TCR-T都以腫瘤相關性抗原作為靶點,但這往往會導致"脫靶"的可能性。目前使用較多的一些腫瘤相關抗原,甲胎蛋白、NY-ESO、MAGE等蛋白都屬於這一狀態。
在諸多肝癌相關環境危險因素中,乙型肝炎病毒(Hepatitis B Virus,HBV)或丙型肝炎病毒(Hepatitis C Virus,HCV)感染與肝癌的發生直接相關。高達80%的肝癌歸因於HBV或HCV感染,在中國主要是HBV感染為主,90%肝癌患者HBsAg呈陽性。HBV感染人體後可刺激機體產生一系列體液免疫和細胞免疫反應,通常可以清除感染的病毒而痊癒;但如果機體免疫反應低下,不足以清除病毒,則病毒可持續存在,可發展成為慢性乙肝。慢性乙肝是肝硬化和肝癌發生的重要危險因素。慢性乙肝主要涉及免疫反應各個階段的抗病毒免疫反應缺陷,尤其是HBV特異性CD8+
T細胞的數量下降、功能降低,引起免疫耐受;同時伴隨免疫介導的炎症性肝損傷。
目前,尚未發現能夠治癒乙肝的藥物。臨床上針對慢性乙肝的治療手段主要為抗病毒相關藥物的治療,包括干擾素類(IFNs)及核苷和核苷酸類藥物等,通過免疫調節或干擾HBV複製發揮抗病毒作用。然而,這些治療手段不能徹底清除病毒,使得患者容易出現耐藥、病毒變異以及病情反覆等情況。
HBV特異性CD8+
T細胞在控制病毒的複製、清除病毒和HBV感染的臨床恢復上起著決定性作用。HBV特異性TCR基因修飾的T細胞(TCR-T)過繼轉移,已初步證明有非常好的抗病毒活性。在肝癌領域,HBV也可作為一種獨特的腫瘤相關性抗原。雖然已知的HBV-HCC肝癌細胞不表達完整的HBV抗原,但是在慢性乙型肝炎的自然史感染中,病毒往往將自身整合到人類基因組中,最終形成HCC細胞並攜帶這些HBV基因組。新加坡學者研究發現,儘管不表達完整HBV抗原,HBV-HCC肝癌細胞卻帶有短片段的HBV mRNAs,這些mRNAs可以編碼能夠被HBV特異性T細胞識別並活化的表位多肽,他們入組了2例肝移植患者肝癌復發和肺部轉移瘤,根據腫瘤中HBV mRNA表達選擇HBV特異性T細胞受體(TCRs),利用基因工程手段讓自體T細胞表達這些TCRs,並過繼轉移到病人體內。這些TCR-T細胞不影響肝功能,在1例患者中,5/6肺轉移瘤在1年內體積減小。該結果提示在肝癌病人,特別是肝移植患者治療中,將肝癌細胞上HBV抗原表位當作腫瘤相關抗原進行T細胞過繼免疫治療有可能將HBV-HCC肝癌細胞清除掉。這種肝特異性標記意味著腫瘤外的不良反應在很大程度上是可以預測的,很少或不涉及其它器官。此外,也有其它團隊臨床研究通過體外誘導TCR重定向的T細胞的方法,將特異識別乙肝病毒HLA-A2限制性表位HBs183-191
的TCR-T細胞用於治療肝移植患者中的肝癌轉移,證明了這種重定向TCR-T細胞具有靶向特定肝癌細胞的活性,展現出巨大的應用潛力和價值。
現有報導的治療性T細胞的研究主要局限在HLA-A2人群的研究,而針對其它HLA人群的關注卻明顯不足。HLA-A3超家族(包含HLA-A11、HLA-A33、HLA-A68、HLA-A31等)在中國人群HLA分型中佔著最大比例(約52.7%)。HLA-A3超家族成員的顯著特徵之一是不同的成員具有共同的多肽結合特性,即偏好結合C末端帶鹼性氨基酸的多肽表位。因此,針對HLA-A3超家族限制性的人群進行肝癌的治療研究將具有很好的大眾性和通用性。其中,HLA-A11是A3超家族中分佈最廣的,統計分析也表明中國乙肝患者中HLA-A11基因頻率最高。
HLA轉基因動物模型在臨床前試驗和基礎實驗研究中發揮了重要作用。HLA-A11轉基因小鼠之前已有報導,然而多方面的證據表明,HLA-A11分子偏好結合C末端鹼性的表位多肽,與小鼠自身抗原提呈系統中抗原相關轉運蛋白(TAP)的多肽結合偏好性不符,因而HLA-A11轉基因小鼠在遞呈HLA-A11限制性表位上存在明顯缺陷。
將含人源TAP-LMP基因簇的BAC轉基因小鼠(hTAP-LMP轉基因小鼠)與HLA-A11轉基因小鼠雜交,得到優化型的HLA-A11/hTAP-LMP轉基因小鼠。與HLA-A11轉基因小鼠相比,HLA-A11/hTAP-LMP轉基因小鼠具有更強的提呈HLA-A11限制性CTL表位的能力。
綜上所述,免疫細胞技術治療乙肝和肝癌進入到臨床試驗階段並取得了進展,顯示出廣闊的應用前景。但是在中國人群為主的HLA-A11限制性肝癌治療極需進行深入的研究。
本發明的目的是製備用於預防和/或治療由HBV感染所致的疾病的藥物。
本發明首先保護一種識別HLA-A11限制性乙型肝炎病毒HBc141-151
表位肽的T細胞受體,可包含α鏈和β鏈。所述α鏈可包含三個互補決定區,氨基酸序列分別如SEQ ID No. 2的第48-53位、第71-77位和第112-121位所示;或這些序列的具有至多3個、2個或1個氨基酸改變的變體。所述β鏈可包含三個互補決定區,氨基酸序列分別如SEQ ID No. 4的第46-50位、第68-73位和第111-122位所示;或這些序列的具有至多3個、2個或1個氨基酸改變的變體。
所述T細胞受體中,所述α鏈的可變區的氨基酸序列可如SEQ ID No. 2的第22-112位所示;或這些序列的具有至多3個、2個或1個氨基酸改變的變體。所述β鏈的可變區的氨基酸序列可如SEQ ID No. 4的第20-113位所示;或這些序列的具有至多3個、2個或1個氨基酸改變的變體。

具有上述任一所述T細胞受體的T 細胞也屬於本發明的保護範圍。
含有上述任一所述T細胞受體的T細胞或"上述任一所述表達盒、載體或細胞"的藥物組合物也屬於本發明的保護範圍。所述藥物組合物可用於預防和/或治療由HBV感染所致的疾病。
本發明還保護上述任一所述T細胞受體、或上述任一所述核酸分子、或上述任一所述載體或細胞、或上述任一所述T細胞受體的T細胞的應用,其應用可為A1)~A4)中的至少一種:
A1)製備預防和/或治療由HBV感染所致的疾病的藥物;
A2)預防和/或治療由HBV感染所致的疾病;
A3)體內或體外殺傷靶細胞;
A4)清除HBV的慢性感染。

上述任一所述由HBV感染所致的疾病可為慢性乙型肝炎或肝細胞癌。
本發明的發明人經過大量實驗,分離並鑑定了一對HBV特異性的TCR序列,成功構建了這對TCR的轉基因小鼠,並且在體外驗證了TCR轉基因陽性的CD8細胞(即TCR-T細胞)具有HBV多肽表位依賴性的活化和增殖能力;同時利用動物體內和體外殺傷靶細胞實驗,證明了這對TCR具有很好的殺傷靶細胞(HLA-A11轉基因小鼠負載多肽HBc141-151
的脾細胞或PBMC細胞)的活性;體外驗證Human TCR-T也具有特異性的殺傷靶細胞(負載多肽HBc141-151
的HLA-A11限制性人的PBMC細胞)的能力;此外,動物實驗表明這對TCR序列是清除HBV感染細胞的有效方法之一。本發明提供的HLA-A11 限制性HBc141-151
表位肽的T細胞受體具有重要的應用價值。
以下的實施例便於更好地理解本發明,但並不限定本發明。下述實施例中的實驗方法,如無特殊說明,均為常規方法。下述實施例中所用的試驗材料,如無特殊說明,均為自常規生化試劑商店購買得到的。以下實施例中的定量試驗,均設置三次重複實驗,結果取平均值。

[實施例1、TCR-T細胞的獲得及其應用]
一、應用HLA-A11/hTAP-LMP轉基因小鼠篩選獲得A11限制性HBV特異性TCR序列
1、將100μL含100μg多肽HBc123-157
和100μg輔助多肽HBc128-140
的PBS緩衝液和100μL IFA混合,乳化;然後皮下多點注射至HLA-A11/hTAP-LMP轉基因小鼠。HLA-A11/hTAP-LMP轉基因小鼠體內將誘導出針對HBc141-151
表位的初次CTL免疫應答。
2、完成步驟1後第14天,採用尾靜脈高壓注射法向HLA-A11/hTAP-LMP轉基因小鼠的尾靜脈注射pAAV/HBV1.2質體,每隻小鼠注射10μg pAAV/HBV1.2質體。
pAAV/HBV1.2質體包含HBV病毒1.2個拷貝的基因組,採用尾靜脈高壓注射法可瞬時轉染小鼠肝細胞並表達乙肝病毒抗原以及產生病毒顆粒,因而可用於在小鼠體內類比HBV的早期感染。進行該步驟的目的是HBV的早期感染後,誘導針對HBc141-151
的二次CTL免疫應答,產生大量HBc141-151
抗原特異性的CD8+
T細胞。
3、完成步驟2後第8天,處死HLA-A11/hTAP-LMP轉基因小鼠並收集外周血單核細胞,進行Tetramer染色,之後流式分選出HLA-A11/HBc141-151
Tetramer(NIH Tetramer Facility)和CD8雙陽性的T細胞,使用口吸式顯微注射針將抗原特異性CTL在顯微鏡下手動吸取單個細胞,即獲得單個抗原特異性CTL細胞。
4、完成步驟3後,首先配製反轉錄體系Mix1(由0.5μL Random引物和4.5μL DEPC處理的H2
O組成)和反轉錄體系Mix2(由5μL 2×buffer和0.5μL RT酶組成);然後在顯微鏡下使用顯微注射針將單個抗原特異性CTL細胞吹入反轉錄體系Mix1中,70°C 5min,冰浴2min;之後加入反轉錄體系Mix2,25°C 5min,42°C 30min,85°C 5min,合成單細胞cDNA。
前述Random引物為生工生物工程(上海)股份有限公司的產品。以及前述2×buffer和RT酶皆為北京全式金生物技術有限公司的產品。
5、完成步驟4後,進行兩輪TCR特異性兼併引物PCR。第一輪PCR:TCRα-mix,V區23條,C區1條;TCRβ-mix,V區19條,C區1條。第二輪PCR:TCRα-mix(in),V區23條,C區1條;TCRβ-mix(in);V區19條,C區1條。引物的核苷酸序列具體見表1。

6、完成步驟5後,兩輪PCR結束後得到配對的TCR-α和TCR-β可變區序列。切膠回收PCR擴增產物,測序。將測序結果在IMGT網站(網址為:http://www.imgt.org/IMGT_vquest/vquest?livret=0&Option=mouseTcR)上進行分析,得到單個細胞的α鏈序列和β鏈序列。
共檢測40個抗原特異性CTL。其中5個抗原特異性CTL可以獲得α鏈序列和β鏈序列,即得到5對TCR受體序列。經過分析,這5個抗原特異性CTL的α鏈和β鏈完全相同。
α鏈可變區的編碼基因如SEQ ID No.1的第64-336位所示;其中,SEQ ID No.1中,第142-159位、第211-231位和第334-363位分別為三個CDR的編碼基因。
α鏈可變區的氨基酸序列如SEQ ID No.2的第22-112位所示;其中,SEQ ID No.2中,第48-53位、第71-77位和第112-121位分别為三個互補決定區。
β鏈可變區的編碼基因如SEQ ID No.3的第58-339位所示;其中,SEQ ID No.3中,第136-150位、第202-219位和第331-366位分別為三個CDR的編碼基因。
β鏈可變區的氨基酸序列如SEQ ID No.4的第20-113位所示;其中,SEQ ID No.4中,第46-50位、第68-73位和第111-122位分别為三個互補決定區。
這對α鏈序列和β鏈序列組成的TCR受體可能對HLA-A11限制性CTL表位HBc141-151
具有高親和力,並來自同一個T細胞選殖(Clone)。
前述步驟3~6的實驗流程圖請參見圖1。
二、表達TCR的Jurkat細胞的獲得及鑑定
1、根據步驟一得到的α鏈可變區和β鏈可變區的序列,參考NCBI上小鼠基因組α鏈和α鏈的恆定區序列,獲得並人工合成HLA-A11限制性乙型肝炎病毒HBc141-151
特異性TCR受體α鏈和β鏈的完整編碼基因。
α鏈的完整編碼基因如SEQ ID No.1所示,編碼SEQ ID No.2所示的α鏈。
β鏈的完整編碼基因如SEQ ID No.3所示,編碼SEQ ID No.4所示的β鏈。
2、將逆轉錄病毒載體MSCV-IRES-GFP的限制性內切酶XhoI和EcoRI之間的DNA小片段替換為SEQ ID No.5所示的DNA分子,其它序列均不變,得到重組質體MSCV-TCR-IRES-GFP(即重組質體MSCV-TCR-GFP)。SEQ ID No.5中,第1至804位為α鏈的完整編碼基因,第805至867位為T2A自剪切多肽的編碼基因,第868至1785位為β鏈的完整編碼基因。
逆轉錄病毒載體MSCV-IRES-GFP是將MO載體的限制性內切酶XhoI和ClaI之間的DNA小片段替換為IRES核苷酸序列(Genbank:MG550106.1)和螢光標記蛋白GFP核苷酸序列(Genbank:MH777595.1),得到的重組質體。
MO載體記載於如下文獻:Tanyu Hu, Krung Phiwpan, Jitao Guo, et al. MicroRNA-142-3p Negatively Regulates Canonical Wnt Signaling Pathway. PLOS ONE. 2016, DOI: 10.1371/journal.pone.0158432。
3、培養Jurkat細胞至數量2×107
以上,收集細胞,用不含抗生素的1640培養基清洗2遍,最後一遍洗後用不含抗生素1640培養基重懸至5×107
/mL,將上述細胞分為400μL/份加至電轉杯(BIO-RAD,貨號:165-2088),同時加入40μg重組質體MSCV-TCR-GFP,混勻,將電轉杯放入電轉儀(BIO-RAD,Gene Pulser Xcell™)中,電壓250V,電容950μF電轉,將重組質體MSCV-TCR-GFP導入Jurkat細胞,得到表達TCR的Jurkat細胞。
按照上述方法,將重組質體MSCV-TCR-GFP替換為逆轉錄病毒載體MSCV-NGFR-GFP,其它步驟均不變,得到表達NGFR的Jurkat細胞。
逆轉錄病毒載體MSCV-NGFR-GFP是將MO載體的限制性內切酶XhoI和EcoRI之間的DNA小片段替換為NGFR核苷酸序列(序列來源於addgene NGFR質體,Plasmid #27489),再將限制性內切酶EcoRI和ClaI之間的DNA小片段替換為IRES核苷酸序列(Genbank:MG550106.1)和螢光標記蛋白GFP核苷酸序列(Genbank:MH777595.1),得到的重組質體。
4、利用Tetramer(HLA-A11/HBc141-151
)對步驟3得到的表達TCR的Jurkat細胞或表達NGFR的Jurkat細胞進行染色。
染色結果請參見圖2。結果表明,TCR對HLA-A11限制性表位具有較強的親和力。
三、HBc141-151
特異性TCR轉基因小鼠(以下簡稱TCR轉基因小鼠)的構建和鑑定
1、重組質體phCD2-TCR-α和重組質體p428-TCR-β的獲得:
(1)向phCD2質體的限制性內切酶EcoRI的識別位點處插入SEQ ID No.1所示的TCR-α基因序列,其它序列均不變,得到重組質體phCD2-TCR-α。
phCD2質體記載於如下文獻:Zhumabekov T, Corbella P, Tolaini M. Kioussis D. Improved version of a human CD2 minigene based vector for T cell-specific expression in transgenic mice. Journal of Immunological Methods. 1995 Sep 11;185(1):133-40。
(2)向p428質體的限制性內切酶SalI的識別位點處插入SEQ ID No.3所示的TCR-β基因序列,其它序列均不變,得到重組質體p428-TCR-β。
p428質體記載於如下文獻:Sawada S, Scarborough JD, Killeen N, Littman DR. A lineage-specific transcriptional silencer regulates CD4 gene expression during T lymphocyte development. Cell. 1994 Jun 17;77(6):917-29。
2、超級排卵(Superovulation)以及假孕母鼠的準備:
(1)超級排卵:向3週齡的C57BL/6雌性小鼠腹腔注射PMSG(5U/只);48h以後,腹腔注射HCG(5U/只);注射HCG後立即與B6D2F1雄鼠1:1合籠,合籠18~24h後檢查陰道栓,有陰道栓的小鼠提出待用。
(2)假孕母鼠的準備:在步驟(1)中C57BL/6雌性小鼠與B6D2F1雄鼠合籠的同時,將ICR雌鼠與結紮的ICR雄鼠以2:1的比例合籠,合籠18~24h後檢查,有陰道栓的小鼠(即假孕母鼠)提出待用。
3、取卵:
將步驟2中(1)有陰道栓的C57BL/6雌性小鼠採頸椎脫位處死,並解剖小鼠,用鑷子夾住子宮上部,將輸卵管分離至M2培養基中,在顯微鏡下劃開輸卵管的壺腹部,使卵流到培養液中。向培養液裡加入1mg/mL的透明質酸酶,以去除受精卵周圍的顆粒細胞。選擇狀態較好的受精卵,並轉移至塑膠皿(直徑35mm)裡由礦物油覆蓋著的M2培養基液滴之中,在二氧化碳培養箱(37°C,5%二氧化碳,95%空氣)中培養直到受精卵適合注射為止。
4、向受精卵原核注入DNA溶液:
首先,將1mL注射器的活塞切兩段長度為0.5cm的小柱,用砂輪切割一片寬度為0.5cm的蓋玻片,75%(v/v)乙醇水溶液消毒。用凡士林把切下來的兩段小柱按蓋玻片的長度黏在載玻片上,利用1mL的注射器在兩小柱中央滴兩小滴M2溶液,同時在蓋玻片上滴一滴,然後將蓋玻片反轉蓋在兩小柱上壓緊,將注射器針頭插入蓋玻片的M2液滴裡緩慢推入M2溶液,使圍成的小室充滿M2溶液。隨後將完成的注射室放到顯微操作儀的載物台上,用胚胎移植管將一組受精卵(約100枚)移到注射室中。將固定針和注射針裝上並調整到視野中央,調整各自的X、Y和Z軸,使固定針、注射針和受精卵處於同一水平面上。推動注射針透過透明質帶,進入原核,利用壓力泵的持續壓力(約150hPa)向原核注入DNA(濃度為3-5ng/μL的重組質體phCD2-TCR-α和濃度為3-5ng/μL的重組質體p428-TCR-β按體積比1:1混合而成)至原核稍微膨大,然後將注射針迅速撤出。調節固定針為正壓,使注射過的受精卵脫落,再調節為負壓吸附另一個受精卵進行注射。受精卵注射完畢後,立即移回M16培養基中,至於37°C的培養箱培養(約12h)。
5、受精卵移植:
將假孕母鼠麻醉,手術取出卵巢連接輸卵管並用脂肪鑷固定,在顯微鏡下找到輸卵管開口。移植管吸取經顯微注射DNA後培養成活的受精卵,將移植管口插入輸卵管口後輕輕將移植管內的液體吹入,將卵巢連同輸卵管放回腹腔,縫合肌肉和皮膚。
6、TCR轉基因小鼠的鑑定:
取步驟5處理後假孕母鼠分娩的首代小鼠進行基因型鑑定,具體步驟如下:取首代小鼠的尾靜脈血,使用ACK紅血球裂解液裂解紅血球,離心,收集白血球。使用anti-mouse CD8a(Biolegend,克隆號:53-6.7)以及anti-mouse TCRVβ10(BDBioscience,克隆號:B21.5)的抗體對獲得的白細胞進行染色。
按照上述方法,將首代小鼠替換為野生型小鼠,其它步驟均不變,作為對照。
染色結果請參閱圖3。結果表明,TCR轉基因小鼠的體內可以檢測到CD8細胞高表達特異性TCRVβ。
四、TCR轉基因小鼠的CD8+
T細胞(即TCR-T細胞)具有HBV多肽依賴的活化增殖能力
TCR轉基因小鼠中特異性TCR轉基因陽性T細胞即為TCR-T細胞。
1、取TCR轉基因小鼠淋巴結以及脾臟細胞,計數並將細胞濃度稀釋至3×107
/mL。每3×107
個細胞加入1μg anti-CD4抗體(BioXcell,克隆號:GK1.5),4°C旋轉孵育30min。
2、完成步驟1後,加入2%FBS DMEM培養基使體積達到15mL,4°C、2000rpm離心2min,去除上清,以洗去未結合的抗體。然後每3×107
個細胞加入1mLGoatAnti-Mouse IgG(QIAGEN,貨號:310007)、1mL GoatAnti-Rat IgG(QIAGEN,貨號:310107)和磁珠重懸液(取磁珠,用含0.5%(v/v)BSA和2mM EDTA的1×PBS緩衝液洗滌2次,然後用含0.5%(v/v)BSA和2mM EDTA的1×PBS緩衝液重懸獲得),4°C旋轉孵育30min。
3、完成步驟2後,用磁鐵吸附磁珠,除去CD4+
T細胞和B細胞,上清液中剩餘細胞大約有90%為CD8+
T細胞,隨後對得到的CD8+
細胞進行CFSE標記,CFSE標記的基本過程為:取準備標記的細胞,根據需要加入CFSE稀釋液或CFSE原液,充分混勻,37°C、5% CO2
培養10min;之後加入10倍體積預熱的1640完全培養基以停止標記,並用PBS重懸,得到CFSE標記的TCR-T細胞。
4、取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數,加入新鮮1640培養基將細胞稀釋至1×107
個/mL,然後向HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞中加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37°C 、5% CO2
培養1h。
5、完成步驟4後,用1640培養基洗滌兩次,細胞計數。
6、將步驟5得到的細胞和CFSE標記的TCR-T細胞1:1混合,37°C、5% CO2
培養1d、2d或3d。
7、完成步驟6後,進行流式檢測,分析TCR-T細胞的增殖和活化情況。
將上述方法中的多肽HBc141-151
替換為對照多肽,其它步驟均不變,作為對照。對照多肽具體為NP91(NP91為PR8流感病毒NP蛋白91-99肽段,由北京旭和源生物科技有限公司合成)。
將上述方法中的多肽HBc141-151
替換為Anti-CD3抗體(Bioxcell,克隆號:145-2C11),其它步驟均不變,作為陽性對照。
檢測結果見圖4(特異性多肽為多肽HBc141-151
,Anti-CD3為Anti-CD3抗體)。結果表明,HBc141-151
可以有效的活化TCR-T細胞,並且可以有效的刺激TCR-T細胞的增殖。由此可見,TCR-T細胞具有HBV多肽依賴的體外活化增殖能力,當把TCR-T細胞回輸給HBV陽性的動物或者患者時,可以有效的體內激活和擴增回輸的TCR-T細胞。
五、TCR-T細胞具有體外殺傷靶細胞的功能
為了體外驗證TCR具有特異性的殺傷靶細胞的能力,進行體外殺傷實驗。具體步驟如下:
1、TCR-T細胞的活化
(1)取TCR轉基因小鼠淋巴結以及脾臟細胞,用結合CD4+
T細胞以及B細胞的磁珠處理樣品,去除CD4+
T細胞以及B細胞,從而富集CD8+
T細胞,細胞計數。
(2)取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數,加入新鮮1640培養基將細胞稀釋至1×107
個/mL,然後向HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞中加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37°C、5% CO2
培養1h。
(3)完成步驟(2)後,用1640培養基洗滌兩次,細胞計數。
(4)將步驟(3)得到的細胞和TCR轉基因小鼠的CD8+
T細胞等數量混合,加入400U/mL IL-2,37°C、5% CO2
培養5d,得到體外活化的TCR-T細胞。
2、取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數,加入新鮮1640培養基將細胞稀釋至1×107
個/mL,然後向HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞中加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37°C、5% CO2
培養1h,之後1640培養基洗滌兩次,細胞計數,得到負載多肽HBc141-151
的靶細胞。
3、將步驟2得到的負載多肽HBc141-151
的靶細胞加入96孔板中,1×104
個/孔;隨後按10:1、5:1或1:1的比例加入體外活化的TCR-T細胞,37°C、 5% CO2
培養5h。
4、取96孔板,250g離心4min;然後將50μL上清轉移至新的96孔板,每孔加入底物50μL,室溫、避光孵育30min;之後每孔加入50μL終止液,使用酶標儀(波長490nm)讀取數值,即實驗組數值。
底物、終止液和細胞裂解液均來自CytoTox 96® Non-Radioactive Cytotoxicity Assay試劑盒(Promega公司,貨號G1780)。
5、殺傷實驗本底數值檢測
(1)按照上述步驟1~4,將步驟3替換為步驟3A,其它步驟均不變,得到T細胞自釋放值。步驟3A為:取96孔板,加入與實驗組同等數量的體外活化的TCR-T細胞,37°C、5% CO2
培養5h。
(2)按照上述步驟1~4,將步驟3替換為步驟3B,其它步驟均不變,得到靶細胞自釋放值。步驟3B為:取96孔板,加入與實驗組同等數量的負載多肽HBc141-151
的靶細胞,37°C、5% CO2
培養5h。
(3)按照上述步驟1~4,將步驟3替換為步驟3C,其它步驟均不變,得到靶細胞最大釋放值。步驟3C為:取96孔板,加入與實驗組同等數量的負載多肽HBc141-151
的靶細胞和10μL細胞裂解液,37°C、5% CO2
培養5h。
6、計算殺傷活性
將上述方法中的多肽HBc141-151
替換為NP91,其它步驟均不變,作為對照。
檢測結果見圖5(E:T表示TCR-T細胞和靶細胞的比例)。結果顯示,TCR-T細胞在體外可以有效的殺傷靶細胞。
六、TCR-T細胞具有體內殺傷靶細胞的功能
為了體內驗證TCR具有特異性的殺傷靶細胞的能力,進行體內殺傷實驗。將負載多肽HBc141-151
的HLA-A11/hTAP-LMP細胞用較低濃度CFSE(CFSElow
)標記,未負載多肽的HLA-A11/hTAP-LMP細胞作為對照用較高濃度CFSE(CFSEhigh
)標記,將兩種細胞等量混合後回輸至C57/B6J小鼠體內,隨後回輸預先活化的TCR-T細胞,使用流式細胞儀檢測C57/B6J小鼠體內CFSElow
標記的細胞群是否會被TCR-T細胞殺傷。具體步驟如下:
1、TCR-T細胞的活化
(1)取TCR轉基因小鼠淋巴結以及脾臟細胞,用結合CD4+
T細胞以及B細胞的磁珠處理樣品,去除CD4+
T細胞以及B細胞,從而富集CD8+
T細胞,細胞計數。
(2)取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數,加入新鮮1640培養基將細胞稀釋至1×107
個/mL,然後向HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞中加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37°C、5% CO2
培養1h。
(3)完成步驟(2)後,用1640培養基洗滌兩次,細胞計數。
(4)將步驟(3)得到的細胞和TCR轉基因小鼠的CD8+
T細胞等數量混合,加入400U/mL IL-2,37°C、5% CO2
培養5d,得到體外活化的TCR-T細胞。
2、靶細胞的製備
(1)取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數,加入新鮮1640培養基將細胞稀釋至1×107
個/mL,然後向HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞中加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37°C、5% CO2
培養1h,之後1640培養基洗滌兩次,細胞計數。
(2)完成步驟(1)後,用PBS重懸細胞,得到濃度為5×107
/mL的負載多肽HBc141-151
的脾細胞重懸液。
(3)完成步驟(2)後,用較低濃度的CFSE(CFSElow
)標記負載多肽HBc141-151
的脾細胞(每mL負載多肽HBc141-151
的脾細胞需加入1μL濃度為0.5mM的CFSE稀釋液;進行CFSE標記時,CFSE的濃度為0.5μM),得到負載多肽HBc141-151
且CFSElow
標記的脾細胞。
(4)取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數;然後37°C、5% CO2
培養1h(培養基為1640培養基)。
(5)完成步驟(4)後,用1640培養基洗滌兩次,細胞計數。
(6)完成步驟(5)後,用PBS緩衝液重懸,得到濃度為5×107
/mL的細胞重懸液。
(7)完成步驟(6)後,用較高濃度的CFSE(CFSEhigh
)標記未負載多肽的脾細胞(每mL未負載多肽的脾細胞需加入1μL濃度為5mM的CFSE原液;進行CFSE標記時,CFSE的濃度為5μM),得到CFSEhigh
標記的脾細胞。
3、TCR-T細胞具有體內殺傷靶細胞的功能
(1)將負載多肽HBc141-151
且CFSElow
標記的脾細胞和CFSEhigh
標記的脾細胞等數量混合,得到混合細胞。
(2)取C57/B6J小鼠,每隻C57/B6J小鼠回輸(尾靜脈輸入)混合細胞2×107
個。
(3)完成步驟(2)的2h後,每隻C57/B6J小鼠回輸(尾靜脈輸入)步驟1獲得的體外活化的TCR-T細胞1×107
個。
(4)完成步驟(3)的24h後,處死小鼠,收集外周血和脾細胞,裂解紅血球,離心,獲得白血球,製成單細胞懸液。流式細胞儀檢測外周血以及脾臟中CFSElow
與CFSEhigh
的比例。通過CFSElow
與CFSEhigh
的比例定量分析TCR-T細胞的殺傷功能。
按照上述步驟(1)~(4)將步驟(3)替換為步驟K,其它步驟均不變,作為回輸PBS對照。步驟K為:完成步驟(2)的2h後,每隻C57/B6J小鼠回輸與步驟(3)中體外活化的TCR-T細胞等體積的PBS緩冲液。
檢測結果見圖6。與回輸PBS對照組相比,不論在外周血或是脾臟中,CFSElow
標記的細胞群相較CFSEhigh
的細胞群均有明顯的下降。結果表明,TCR-T細胞在體內可以有效的殺傷靶細胞。
七、TCR-T細胞可以有效的清除體内HBV的慢性感染
1、小鼠慢性HBV感染模型的建立
小鼠慢性HBV感染模型的建立的方法記載於如下文獻中:董小岩,尉遲捷,王剛,等。高嗜肝性8型重組腺相關病毒體內轉導法製備乙型肝炎病毒持續感染小鼠模型。病毒學報;2010, 26 (6): 425-431。具體步驟如下:
(1)向HLA-A11/hTAP-LMP轉基因小鼠尾靜脈注射rAAV/HBV1.3病毒(北京五加和分子醫學研究所有限公司的產品),注射劑量為5×109
vg/隻。
完成步驟(1)後,觀察自然情況下病毒梯度(乙肝病毒s抗原、乙肝病毒e抗原或病毒DNA)變化曲線,並根據變化曲線預測TCR-T細胞的給藥時間。
(2)完成步驟(1)後1個月,得到小鼠慢性HBV感染模型。小鼠慢性HBV感染模型中HBsAg和HBeAg的表達受HBV病毒DNA自身的啟動子等元件調控,在肝臟和外周血中可以連續10周觀察到持續表達的HBsAg和HBeAg。
將上述方法中的HLA-A11/hTAP-LMP轉基因小鼠替換為C57BL/6小鼠,其它步驟均不變,獲得慢性HBV感染的C57BL/6小鼠,作為陰性對照。
2、TCR-T細胞治療HLA-A11/hTAP-LMP小鼠的慢性HBV感染
(1)取TCR轉基因小鼠淋巴結以及脾臟細胞,用結合CD4+
T細胞以及B細胞的磁珠處理樣品,去除CD4+
T細胞以及B細胞,從而富集CD8+
T細胞,細胞計數。
(2)取HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞,裂解紅血球,細胞計數,加入新鲜1640培養基將細胞稀釋至1×107
個/mL,然後向HLA-A11/hTAP-LMP轉基因小鼠的脾臟細胞中加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37℃、5% CO2
培養1h。
(3)完成步驟(2)後,用1640培養基洗滌兩次,細胞計數,然後將細胞和TCR轉基因小鼠的CD8+
T細胞等數量混合,加入400U/mL IL-2,37℃、5% CO2
培養5d,得到體外活化的TCR-T細胞。
(4)取HLA-A11/hTAP-LMP轉基因小鼠慢性HBV感染模型,每隻小鼠回輸(尾靜脈輸入)體外活化的TCR-T細胞1×107
個。
(5)完成步驟(4)後記為第0天,每隔兩天檢測血清中HBsAg的OD450nm
值(HBsAg檢測試劑盒,上海科華)和ALT濃度(ALT/GPT kit,全自動生化分析儀MEDSOUL AMS-18),從而判斷體內HBV病毒的感染情況。共檢測8天。
(6)完成步驟(4)後第7天,取HLA-A11/hTAP-LMP轉基因小鼠慢性HBV感染模型,二次回輸體外活化的TCR-T細胞1×107
個。
(7)完成步驟(6)後2天,檢測血清中HBsAg的OD450nm值和ALT濃度。
(8)將步驟(4)~(7)中的HLA-A11/hTAP-LMP轉基因小鼠慢性HBV感染模型替換為C57BL/6小鼠慢性HBV感染模型,其它步驟均不變,作為對照。
血清中HBsAg的OD450nm
值的檢測結果見圖7中A(WT 1#,2#,3#,4#均為C57BL/6小鼠慢性HBV感染模型,A11. hTAP 36#、A11. hTAP 37#、A11. hTAP 39#、A11. hTAP 40#和A11. hTAP 43#均為HLA-A11/hTAP-LMP轉基因小鼠慢性HBV感染模型)。血清中ALT濃度檢測結果見圖7中B(WT 1#,2#,3#,4#均為C57BL/6小鼠慢性HBV感染模型,A11. hTAP 36#、A11. hTAP 37#、A11. hTAP 39#、A11. hTAP 40#和A11. hTAP 43#均為HLA-A11/hTAP-LMP轉基因小鼠慢性HBV感染模型)。結果表明,TCR-T細胞可以有效的清除HLA-A11/hTAP-LMP轉基因小鼠慢性HBV感染模型體內的HBV病毒感染。
八、Human TCR-T細胞具有體外殺傷靶細胞的功能
為了體外驗證Human TCR-T具有特異性的殺傷靶細胞的能力,進行體外殺傷實驗。具體步驟如下:
1、慢病毒的包裝與濃縮
(1)將慢病毒包裝載體pCDH-MSCV-MCS-IRES-GFP(SystemBiosciences,編號:CD731B-1)的限制性內切酶EcoRI與BamHI之間的DNA小片段替換為TCR DNA片段(SEQ ID No.5所示),得到pCDH-MSCV-TCR-GFP質體。
(2)將293T細胞吹打重懸成單細胞,計數,然後用含10%(v/v)FBS的DMEM培養基調節,得到濃度為5×105
個/mL的細胞懸浮液。取培養皿(規格為10cm),鋪10mL細胞懸浮液,培養過夜。
(3)完成步驟(2)後,待293T細胞匯合度為75%時進行轉染,轉染前30min更換培養基為DMEM培養基。
(4)轉染預混物準備
向500μL DMEM培養基中加入12μg pCDH-MSCV-TCR-GFP質體、9μg psPAX2和6μg pMG2.D,渦旋充分混勻,得到質體混合物(psPAX2和pMG2.D均為北京天恩澤基因科技有限公司的產品)。
向500μL DMEM培養基中加入27μg PEI,渦旋混勻,靜置5min,得到PEI混合物。
(5)完成步驟(4)後,向500μL質體混合物中加入500μL PEI混合物,渦旋充分混勻,室溫孵育20min;然後將孵育完成的混合物輕柔沿培養皿側壁加入293T細胞中,輕晃培養皿混勻,37°C培養箱培養。6-8h後將培養基更換為10mL含10%(v/v)FBS的DMEM培養基。
(6)完成步驟(5)後第48h,收集第一次病毒上清,補充新鮮DMEM培養基,病毒上清4°C保存。
(7)完成步驟(5)後第72h,收集第二次病毒上清。將第一次病毒上清和第二次病毒上清合併,800g、室溫離心5min,收集上清。
(8)完成步驟(7)後,將所述上清使用0.45μm PES濾膜過濾除去細胞碎片,收集病毒上清。
(9)完成步驟(8)後,將所述病毒上清轉移至超速離心管,吸取2mL 20%無菌蔗糖加入到管底。4°C、70000g離心120min,小心棄上清,收集沉澱(白色病毒顆粒)。
(10)完成步驟(9)後,取所述沉澱,加入100倍濃縮體積的1640培養基重懸,4°C溶解過夜,得到濃縮慢病毒。將濃縮慢病毒分裝儲存於-80°C超低溫冰箱,備用。
2、人外周淋巴細胞的分離
(1)抽取人外周靜脈血5mL。
(2)完成步驟(1)後,取離心管(規格為50mL),加入5mL外周血和5mL PBS緩衝液,充分混匀。
(3)完成步驟(2)後,向離心管中加入5mL人外周血淋巴細胞分離液(天津市灝洋生物製品科技有限公司,貨號:LTS1077),使用一次性無菌滴管取稀釋後外周血沿管壁小心疊加於分離液液面之上,注意保持清楚界面。
(4)完成步驟(3)後,將離心管置於離心機中,調節升速和降速為最低,800g離心20min。
(5)完成步驟(4)後,離心結束後管內分為四層,第一層為血漿和PBS,第二層為環狀乳白色淋巴細胞層,第三層為透明分離液層,第四層為紅血球和粒細胞層。用移液器小心吸取第二層環狀乳白色淋巴細胞層至無菌離心管(規格為50mL),向離心管中加入40mL PBS緩衝液,混勻細胞,800g離心5min,棄上清,之後使用1640培養基重懸細胞,計數備用。
3、人外周T淋巴細胞的活化
(1)取24孔板,每孔加入500μL anti-CD3抗體稀釋液和500μL anti-CD28抗體稀釋液,4°C包被(Coating)過夜。
anti-CD3抗體稀釋液:用PBS緩衝液稀釋anti-CD3抗體(BioXcell,克隆號:OKT3)至濃度為3μg/mL獲得。
anti-CD28抗體稀釋液:用PBS緩衝液稀釋anti-CD28抗體(BioXcell,克隆號:CD28.2)至濃度為1μg/mL獲得。
(2)完成步驟(1)後,取所述24孔板,移除液體,並用PBS緩衝液清洗一次。
(3)完成步驟(2)後,取所述24孔板,加入500μL人外周T淋巴細胞稀釋液,37°C培養箱培養48h;之後400g離心5min,收集沉澱並使用1640培養基重懸,得到活化的人外周T淋巴細胞。活化的人外周T淋巴細胞用於感染慢病毒。
人外周T淋巴細胞稀釋液:用1640培養基將步驟2獲得的人外周T淋巴細胞稀釋至4×106
個/mL獲得。
4、慢病毒感染人外周T淋巴細胞
(1)取24孔板,每孔加入5×105
個活化的人外周T淋巴細胞和200μL濃縮慢病毒,然後用1640培養基補體積至500μL;最後加入polybrene和IL-2,並使polybrene和IL-2在體系中的濃度分別為8μg/mL和40U/mL。
(2)完成步驟(1)後,取24孔板,600g、32℃離心90min;然後將24孔板放入37℃培養箱感染24h。
(3)完成步驟(2)後,取所述24孔板,小心吸掉感染孔中350μL培養基,然後加入1640培養基補足至體積2mL並吹打混勻,37℃培養箱繼續培養48h。
(4)完成步驟(3)後,先取適量感染後T細胞進行流式檢測感染效率;然後使用anti-mouse CD8a抗體(Biolegend,克隆號:53-6.7)和anti-mouse TCRVβ10抗體(BD Bioscience,克隆號:B21.5)對感染後T細胞進行染色,4°C染色30min後進行上機檢測,感染效率(TCRVβ10陽性率)大於15%,陽性細胞即為成功轉入TCR的Human TCR-T細胞,用於後續殺傷實驗。
5、Human TCR-T體外具有殺傷靶細胞的功能
底物、終止液和細胞裂解液均為CytoTox 96® Non-Radioactive Cytotoxicity Assay試劑盒(Promega公司,貨號:G1780)中的組件。
(1)抽取HLA-A11陽性健康人外周靜脈血5mL,然後按照步驟2的方法分離獲得淋巴細胞,即HLA-A11+
PBMC細胞。
(2)完成步驟(1)後,用1640培養基將HLA-A11+
PBMC細胞稀釋至濃度為1×107
個/mL,然後加入多肽HBc141-151
並使其在體系中的濃度為10μg/mL,37°C、5% CO2
培養1h,之後1640培養基洗滌兩次,細胞計數,得到負載多肽HBc141-151
的靶細胞。
(3)將步驟(2)得到的負載多肽HBc141-151
的靶細胞加入96孔板中,2×104

(5)殺傷實驗本底數值檢測
①按照上述步驟(1)~(4),將步驟(3)替換為步驟3A,其它步驟均不變,得到T細胞自釋放值。步驟3A為:取96孔板,加入與實驗組同等數量的Human TCR-T細胞,37°C、5% CO2
培養5h。
②按照上述步驟(1)~(4),將步驟(3)替換為步驟3B,其它步驟均不變,得到靶細胞自釋放值。步驟3B為:取96孔板,加入與實驗組同等數量的負載多肽HBc141-151
的靶細胞,37°C、5% CO2
培養5h。
③按照上述步驟(1)~(4),將步驟(3)替換為步驟3C,其它步驟均不變,得到靶細胞最大釋放值。步驟3C為:取96孔板,加入與實驗組同等數量的負載多肽HBc141-151
的靶細胞和10μL細胞裂解液,37°C、5% CO2
培養5h。
6、計算殺傷活性
將上述方法中的多肽HBc141-151
替換為NP91,其它步驟均不變,作為對照。
將上述方法中的Human TCR-T替換為未轉染TCR的Human T細胞,其它步驟均不變,作為對照。
檢測結果見圖8(A為Human TCR-T細胞的獲得;B為Human TCR-T細胞體外殺傷靶細胞實驗,其中E:T表示Human TCR-T細胞和靶細胞的比例,T+HBc141-151

多肽的靶細胞)。結果表明,Human TCR-T細胞在體外可以有效的殺傷靶細胞。
綜上所述,本發明的發明人分離並鑑定了一對HBV特異性的TCR序列,成功構建了這對TCR的轉基因小鼠,並且在體外驗證了TCR轉基因陽性的CD8細胞(即TCR-T細胞)具有HBV多肽表位依賴性的活化和增殖能力;利用動物體內和體外殺傷靶細胞實驗,證明了這對TCR具有很好的殺傷靶細胞的活性;體外驗證Human TCR-T也具有特異性的殺傷HLA-A11+
靶細胞的能力;此外,動物實驗提示這對TCR序列可能是清除HBV感染細胞的有效方法之一。

Claims (13)
- 一種識別HLA-A11限制性HBc141-151 表位肽的T細胞受體,包含α鏈和β鏈; 該α鏈包含三個互補決定區,氨基酸序列分別如SEQ ID No. 2的第48-53位、第71-77位和第112-121位所示; 該β鏈包含三個互補決定區,氨基酸序列分別如SEQ ID No. 4的第46-50位、第68-73位和第111-122位所示。
- 如請求項1所述的T細胞受體,其特徵在於: 該α鏈的可變區的氨基酸序列如SEQ ID No. 2的第22-112位所示; 該β鏈的可變區的氨基酸序列如SEQ ID No. 4的第20-113位所示。
- 如請求項1或2任一所述的T細胞受體,其特徵在於: 該α鏈的氨基酸序列如SEQ ID No. 2所示; 該β鏈的氨基酸序列如SEQ ID No. 4所示。
- 編碼請求項1至3任一所述T細胞受體的核酸分子。
- 如請求項4所述的核酸分子,其特徵在於:編碼所述T細胞受體的核酸分子包含編碼所述T細胞受體的α鏈的核酸分子和編碼所述T細胞受體的β鏈的核酸分子; 編碼所述T細胞受體的α鏈中三個互補決定區的核苷酸序列分別如SEQ ID No. 1的第142-159位、第211-231位和第334-363位所示; 編碼所述T細胞受體的β鏈中三個互補決定區的核苷酸序列分別如SEQ ID No. 3的第136-150位、第202-219位和第331-366位所示。
- 如請求項4或5任一所述的核酸分子,其特徵在於: 編碼所述α鏈的可變區的核苷酸序列如SEQ ID No. 1的第64-336位所示; 編碼所述β鏈的可變區的核苷酸序列如SEQ ID No. 3的第58-339位所示。
- 如請求項4至5任一所述的核酸分子,其特徵在於: 編碼所述α鏈的核酸分子的核苷酸序列如SEQ ID No. 1所示; 編碼所述β鏈的核酸分子的核苷酸序列如SEQ ID No. 3所示。
- 含有如請求項4至7任一所述核酸分子的表達盒、載體或細胞。
- 如請求項8所述的載體,其特徵在於:該載體為逆轉錄病毒載體或慢病毒載體; 該逆轉錄病毒載體為向逆轉錄病毒載體MSCV-IRES-GFP的多重選殖位點(Mutiple cloning site;MCS)之間插入編碼所述T細胞受體的α鏈的核酸分子和編碼所述T細胞受體的β鏈的核酸分子,得到的重組質體; 該慢病毒載體為向慢病毒包裝載體pCDH-MSCV-MCS-IRES-GFP的多重選殖位點(Mutiple cloning site;MCS)之間插入編碼所述T細胞受體的α鏈的核酸分子和編碼所述T細胞受體的β鏈的核酸分子,得到的重組質體。
- 如請求項8所述的細胞,其特徵在於:該細胞為T細胞或Jurkat細胞。
- 具有如請求項1至3任一所述T細胞受體的T細胞。
- 含有如請求項8至10任一所述的載體或細胞、或含有如請求項11所述的T細胞的藥物組合物。
- 如請求項1至3任一所述的T細胞受體、或如請求項4至7任一所述的核酸分子、或如請求項8至10任一所述的載體或細胞、或如請求項11所述T細胞受體的T細胞的應用,其為A1)~A4)中的至少一種: A1)製備預防和/或治療由HBV感染所致的疾病的藥物; A2)預防和/或治療由HBV感染所致的疾病; A3)體內或體外殺傷靶細胞; A4)清除HBV的慢性感染。
