Abstract
Yaprak sapi dokusunun Agrobacterium vasitali dönüsümü yoluyla transgenik pamuk bitkisi üretmek için bir yöntem ifsa edilmistir. Yöntem asagidaki adimlari ihtiva eder: (a) pamuk yaprak sapi eksplantlari elde etmek: (b) yaprak sapi eksplantlarini bir dissal gen ve seçilebilir bir isaret ihtiva eden bir vektör bulunduran Agrobacterium tumefaciens kültürüne tabi tutmak; Agrobacterium dissal genin ve ayiklama ajani direnç geninin yaprak sapi eksplanti hücrelerinin genomuna karali transferini gerçeklestirebilir; (c) yaprak sapi eksplantlarini kalus olusumu dogurtmak üzere yetistirmek; (d) dissal geni sentezleyen kalusu ayiklamak; (e) embriyoit olusumu dogurtmak üzere kalusu süspansiyon kültürü içinde yetistirmek; (f) embriyoitleri tam transgenik pamuk bitkileri olarak islah etmek.
