Abstract
В результате выход биомассы дрожжей с высоким содержанием белка увеличиваетс на 20-50% при сокращении времени культивировани .The invention relates to the microbiological industry and is a method for producing biomass of fodder yeast. The purpose of the invention is to increase the biomass yield and accelerate the process. Yeast is grown on a nutrient medium, the basis of which is brown juice of green plants, while cultivation is carried out in the presence of 0.3-0.5% (w / v) of the solid residue obtained from green plants after the separation of brown juice. As a result, the biomass yield of high protein yeast is increased by 20-50% while reducing the cultivation time.
Description
The invention relates to the microbiological industry, namely to a method for producing biomass of fodder yeast using brown juice of green plants.
The purpose of the invention is to increase the biomass yield and accelerate the process.
The method is carried out as follows.
Cultivation of fodder yeast is carried out on a nutrient medium, the basis of which is brown juice of green plants. Brown juice of green plants is obtained by grinding, pressing, pressed product is moistened with water, re-pressed, added antioxidant
к жидкости, коагулируют подлинно-протеиновую фракцию, отдел ют осадок белка, промывают водой и разбавленной кислотой и объедин ют жидкую фазу с промывной жидкостью.to a liquid, a genuine protein fraction is coagulated, the protein precipitate is separated, washed with water and dilute acid, and the liquid phase is combined with the washing liquid.
A source of nitrogen and 0.3-0.5% (w / v) of the solid residue obtained after separation of the brown juice is added to the resulting brown juice and inoculated with microorganisms. As a result of the cultivation in a shorter time, the biomass yield is 1.5 times the biomass yield without the addition of a solid residue. When the concentration of the solid residue is less than 0.3 and more than 0.5%, the biomass yield decreases.

4,5.Example 1. Fodder yeast Trichosporon cutaneum Nem-2 is grown on a nutrient medium based on brown juice of green plants with a Sugar content of 10 g / l. The amount of phosphorus source introduced was 425 mg / l, nitrogen source 800 mg / l, pH 4.5.
1 Nutrient medium 100 ml each was added to the flasks, sterilized for 15 min at 0.5 atm. The solid residue, pulp, obtained by treating the green mass of plants in the line for the production of protein-green concentrates was sterilized separately for 2 hours at 1.5 atm in flasks in an amount of 0.3% (weight / volume). Then the culture medium was sterilely introduced into the flasks with the pulp and seeded with the Ne-2 Trichosporon cutaneum vegetative culture. The cultivation was carried out in flasks on a shaker at 180 rpm and a temperature of 36 C. At the same time, cultivation was carried out on the medium without the pulp in similar samples. The maximum accumulation of biomass during heterophase cultivation (in the presence of pulp) is reached 12 hours earlier than in the control experiment. The biomass yield per 24 h of cultivation is 14.4 g / l, in the control 9 g / l.
The protein content in biomass is 0.654 g / l, in the control 0.436 g / l.
0,5%.Example 2. The method is carried out according to Example 1, but the Candida scottii Tul-1 yeast is used as a biomass producer. The amount of beet pulp is 0.5%.
As a result, the biomass yield is 12.1 g / l, in the control 10.7 g / l. The duration of cultivation in the control experiment is 9 hours longer.
