Description
The invention relates to the field of medicine, namely to morphology, and can be used, in particular, for the preparation of preparations for light and electron microscopy from biological objects.
The purpose of the invention is to detect water-soluble calcium compounds by fixing a solution of tetroxide osmium in a mixture of organic solvents and dehydration in a mixture of these solvents with a varying ratio of ingredients.
The method is carried out as follows.
The solution is fixed in a 1-3% solution of osmium tetroxide in a mixture of absolute ethyl and acetone ether in a volume ratio of 2: G for 30-60 minutes at 3-5 ° C and dehydration is carried out in an anhydrous mixture of ether and acetone at successive ratios 2: 1, 1: 1, 1: 2 respectively. Then, in three shifts, for 20-40 minutes each, immerse the samples in the casting medium. Four stages of dehydration are carried out at
..
Example 1. Samples of embryonic calcific human cartilage with dimensions 1x1x1 mm are processed. The samples were fixed in a 2% solution of osmium tetroxide in a mixture of ethyl ether and acetone absolute in a volume ratio of 2: 1 at 4 ° C for 60 minutes
Then the samples are successively transferred to a series of dehydrating liquids having a temperature of 4 ° C. The first of them is a mixture of ethyl ether and acetone in an absolute volume ratio of 2: 1. Samples are contained in it for 1 hour. Then they are transferred to a mixture of ethyl ether and acetone absolute in a 1: 1 volume ratio also for 1 hour. A third liquid is a mixture of ethyl ether and acetone absolute in a volume ratio of 1: 2. There are 1 hour samples in it.
The next stage of dehydration is the threefold immersion of samples each time with a portion of acetone absolute for 30 minutes each.

Next, the samples are poured into epoxy resins using absolute acetone according to known methods.
Температура фиксатора .As a result of this treatment, serial ultrathin histological sections are obtained. The structure of the studied tissue is clearly preserved and clearly revealed both at the level of light and electron microscopy. 0 Example 2. Samples of embryonic calcific human cartilage sized 1x1x x1 mm are processed. Fixation was carried out in a 1% solution of osmium tetroxide in a mixture of ethyl ether and acetone absolute at a volume ratio of 2: 1 for 30 minutes. Latching temperature
1:2.Next, dehydration is carried out in three mixtures of ethyl ether and absolute acetone with various volume ratios at 3 ° C for 30 minutes each. 1 mixture - ethyl ether and acetone absolute in the ratio 2: 1, 2 mixture ethyl ether and acetone absolute in the ratio 1: 1, 3 mixture ethyl ether and acetone absolute in the ratio 1: 2.
Continue dehydration in three shifts of absolute acetone at 3 ° C for 20 minutes each.
