Abstract
Description
The present invention relates to a detergent and disinfectant for floors and walls, in particular in hospitals or in the antiseptic treatment of healthy skin.
Doterajší stav technikyBACKGROUND OF THE INVENTION
Nosocomial infections are a major problem for public health in terms of their prevalence, potentially severe degree and their economic and social consequences. They may be alleviated by the use of effective measures, for example by adhering to strict hygiene and disinfection regulations in various areas, such as floors, walls and surfaces of any kind, as well as medical - surgical instruments and apparatus.
The French standard AFNOR defines disinfection as a short-term outcome procedure that allows the removal or killing of micro-organisms and / or the inactivation of unwanted viruses found in contaminated, neutral environments.
So far, the most commonly used active substances for disinfection in hospitals are aldehydes, quaternary ammonium compounds and phenols. Of the aldehydes, formaldehyde and glutaraldehyde are usually used for disinfection. Although they have a wide spectrum of efficacy, they are characterized by considerable toxicity to both users and patients.
Vdychovanie formaldehydu od 0,1 ppm spôsobuje u ľudí slzenie, od 1 do 5 ppm dráždenie horných dýchacích ciest so zápalmi sliznice nosa, hltanu a hrtanu a pri 50 ppm edém pľúc. V nemocniciach spôsobilo používanie väčších množstiev formaldehydu v anatomicko-patologických laboratóriách a hemodialyzačných jednotkách akútne otravy. Inhaling formaldehyde from 0.1 ppm causes tearing in humans, from 1 to 5 ppm irritates the upper respiratory tract with inflammations of the mucous membranes of the nose, pharynx and larynx and at 50 ppm lung edema. In hospitals, the use of larger amounts of formaldehyde in anatomical-pathological laboratories and hemodialysis units has caused acute poisoning. In addition, ocular irritation may occur in certain persons exposed to formaldehyde in concentrations of 0.4 ppm or more. By local action on the mucosa, formaldehyde may result in irritation, changes in the corneal layer of the skin or exematous lesions. Glutaraldehyde may cause respiratory irritation associated with coughing attacks in humans if prolonged inhalation. When applied locally to the mucous membranes, glutaraldehyde causes irritation, erythema or changes in the corneal layer of the skin.
Quaternary ammonium compounds are often used as disinfectants in hospitals because of their bactericidal and cleaning properties. However, there have been numerous cases of skin diseases with contact allergies in humans. Hypersensitivity reactions have been reported in users who have repeatedly or for a long time used quaternary ammonium products. Inhalation of vapors or aerosols containing quaternary ammonium salts can result in pulmonary edema. Accordingly, there is a need for a non-toxic detergent and disinfectant which can be used in hospitals instead of aldehydes and quaternary ammonium compounds.
Podstata vynálezuSUMMARY OF THE INVENTION
Podstata vynálezu spočíva v príprave nového čistiaceho a dezinfekčného prostriedku, ktorý nemá žiadne toxické účinky na používateľa a ktorý možno jednoducho použiť na dezinfekciu povrchov.SUMMARY OF THE INVENTION The present invention is based on the preparation of a novel detergent and disinfectant which has no toxic effects on the user and which can be easily used for surface disinfection.
Vynález sa vzťahuje na čistiaci a dezinfekčný prostriedok, ktorý obsahuje terciárny alkylamín a aminoxid v hmotnostnom pomere medzi 0,2 a 2.The invention relates to a cleaning and disinfecting composition comprising a tertiary alkylamine and an amine oxide in a weight ratio between 0.2 and 2.
U terciárneho alkylamínu ide výhodne o zlúčeninu všeobecného vzorca:According to the invention, according to the invention, it is an alkylamine which has bactericidal, fungicidal and virucidal properties. This substance is rarely inhibited by the predominance of interfering substances. It is also completely toxicologically and ecologically safe. It has been found to be advantageous to combine a synergistically effective amount of amine oxide with the aforementioned alkylamine in order to achieve a marked increase in the antimicrobial effect in conjunction with the simultaneous cleaning effect. In a preferred embodiment of the invention and with a view to maximum efficiency, the tertiary alkylamine comprises from 2 to 15% by weight, preferably from 2 to 9% by weight, based on the total composition. The tertiary alkylamine is preferably a compound of the formula:
(ch2)3 - nh2 (ch2)3 - nh2, kde r! je alkylový radikál s 8 až 16, výhodne 10 až 14 atómami uhlíka. U aminoxidu ide výhodne o zlúčeninu všeobecného vzorca:(ch 2 ) 3 - nh 2 (ch 2 ) 3 - nh 2 where r! is an alkyl radical of 8 to 16, preferably 10 to 14 carbon atoms. The amine oxide is preferably a compound of the formula:
CH3 CH 3
R2 - N -> 0, R2 - C -> 0,
CH3 CH 3
O kde R je alkylový radikál s 8 až 16, výhodne 10 až 14 atómami uhlíka.Where R is an alkyl radical of 8 to 16, preferably 10 to 14 carbon atoms.
It has been found that the best results are obtained when R and R are identical, especially when R x or R is a lauryl radical. In this case, the alkylamine is diaminopropyllaurylamine and the amine oxide is dimethyl laurylamine oxide. The composition according to the invention may furthermore comprise a nonionic surfactant having a low foaming capacity with a proportion of 2 to 20% by weight, preferably 2 to 13% by weight, based on the total composition, and a fragrance with a proportion by weight of 0.005 to 1%, preferably 0.05 up to 0.6% based on the total composition. The nonionic surfactant may be an aliphatic polyalkoxy derivative, for example, a fatty alcohol ethylene oxide dipropylene oxide. It is preferably used in a weight ratio of from 0.2 to 4 to the alkylamine or to the amine oxide. A preferred composition according to the invention in which efficacy has been determined is as follows:
diaminopropyllaurylamín 5,4% dimetyllaurylaminoxid 6 % polyalkoxylderiváx 8 % vonná látka 0,1% pomocné látky v roztoku QSP 100diaminopropyllaurylamine 5.4% dimethyllauryl amine oxide 6% polyalkoxylderivax 8% fragrance 0.1% excipients in QSP 100 solution
The aforementioned composition is produced in a mixer, in which, respectively, purified water (aqua purificata), dimethyl lauryl oxide, diaminopropyl laurylamine and a polyalkoxy derivative are added under stirring. For end use, dilution with water in a ratio of 20 ml of solution and 8 L of water corresponds to a concentration of 0.25%. The product of the invention is a pure, amber-colored solution having a pH of 11.2 ± 0.5. Filling can be in 20 ml bags or in 5 L cans. Due to its antiseptic effect and non-toxic action according to AFNOR standards, which will be discussed in more detail below, it is possible to use the composition according to the invention for the antiseptic treatment of healthy skin.
Pri týchto testoch ide o nasledovné stanovenia:The antimicrobial action of the preferred composition of the invention (above example) has been demonstrated according to AFNOR standards. The results presented in the Annex relate to the various tests carried out after membrane filtration. These tests are as follows:
- stanovenie baktericídneho účinku podlá normy AFNORM NF T 72-151 (príloha 1)- determination of bactericidal activity according to AFNORM NF T 72-151 (Annex 1)
- stanovenie baktericídneho účinku v prítomnosti interferenčnej látky (tvrdá voda) podľa normy NF T 72-171 (príloha 2)- determination of bactericidal action in the presence of an interfering substance (hard water) according to NF T 72-171 (Annex 2)
- Stanovenie baktericídneho účinku v prítomnosti interferenčnej látky (albumín) podľa normy NF T 72-171 (príloha 3),- Determination of bactericidal activity in the presence of an interfering substance (albumin) according to NF T 72-171 (Annex 3),
- stanovenie fungicídneho účinku podľa normy NF T 72-201 (príloha 4).- determination of fungicidal activity according to NF T 72-201 (Annex 4).
Stanovenie baktericídneho účinku proti Mycobacterium tuberculosis podľa Bactec TB-metódy bolo tak isto prevedené.The determination of the bactericidal activity against Mycobacterium tuberculosis according to the Bactec TB method was also carried out.
U Bactec-TB-média (12B) ide o kultivačnú pôdu na báze obohateného Middelbrook 7H9. Mykobaktérie využívajú mastnú kyselinu značenú C14 a uvoľňujú do vzduchu C02.Bactec-TB medium (12B) is a culture medium based on enriched Middelbrook 7H9. Mycobacteria using labeled fatty acid C14 released into the air and C0 second
When mycobacterial growth takes place in the 128B-bottles, Bactec 460 metered radioactivity conditional release 14C0 2 and expressed numerically on a scale of 0 to 999. Tj.eto values are designated as growth index (GI). The increase in GI is proportional to the number of bacteria. Treatment of the Mycobacterium tuberculosis suspension with the disinfectant is followed by a calculation of the number of remaining mycobacteria by injecting each dilution of the microorganism suspension into 2,128B multi-dilution bottles. The GI increase corresponds to the incidence of mycobacteria.
Nakoniec sa látka naplní do 2 12B-fliaš, ktoré obsahujú 4 ml živného roztoku:The method consists in using a McFarland 3-suspension in sterile distilled water, which is prepared from a pure culture of Mycobacterium tuberculosis, which is taken from Löwenstein's culture broth. According to this mother liquor, further dilutions are made by a factor of 10 to 10-9 . 100 ml of each dilution is inoculated into 12B medium containing 3 ml of broth (1/40 dilution in each 12B bottle). Finally, 200 ml of pure or diluted disinfectant is mixed with 200 ml of Mycobacterium tuberculosis with McFarland 3 for a period of time while shaking in a 35 ml centrifuge tube. After the contact time has elapsed, the tube is filled with sterile distilled water and finally centrifuged for 30 min. at 3000 rpm. The excess is collected and the residue is diluted in 2 ml of distilled water (1/10 to 10 µl dilution). Finally, the substance is filled into 2 12B-bottles containing 4 ml of nutrient solution:

Odčítanie vypočítanej hodnoty a pokusu nasleduje na čítači Bactec-TB. Výsledky udávané ako GI možno získať z dodatku 5 z tabuľky pre jednotlivé kontaktné časy, pričom od6 čítanie nasleduje po dvojitom naočkovaní živnej pôdy k časovému bodu TI, T2, T3, T6.T7, T8, T10, T13, T16, T17, T20.Subtracting the calculated value and the experiment follows on a Bactec-TB counter.
