Claims (7)
- Geni za biosintezo spojin tetraciklina in njihove uporabe Patentni zahtevki1. Gensko spremenjena celica, kjer je omenjena celica vrste Aktinomiceta, kjer je omenjena celica zmožna proizvajati spojino tetraciklina po modificirani poti biosinteze tetraciklina, kjer omenjena modificirana biosintetična pot tetraciklina vključuje polipeptide iz SEQ ID No: od 1 do 8, kjer je vsak izmed omenjenih polipeptidov iz SEQ ID No: od 1 do 8, lahko substituiran s polipeptidom, ki je vsak 80% enak in ima enako katalitsko funkcijo kot omenjeni substituirani polipeptid; in, kjer omenjena calica ni Amycolatopsis sulphurea sev NRRL2822.
- 2. Celica po zahtevku 1, kjer omenjena modificirana biosintetična pot tetraciklina vsebuje reakcijo katalizirano dodatnim polipeptidom izbranim iz skupine, ki vsebuje SEQ ID No: od 9 do 18, kjer je omenjeni dodatni polipeptid lahko substituiran s polipeptidom, kije vsaj 80% enak in ima enako katalitsko funkcijo kot omenjeni substituirani dodatni polipeptid.
- 3. Celica po zahtevku 2, kjer omenjena modificirana biosintetična pot tetraciklina vsebuje nadalje vsebuje vsaj eno reakcijo, katalizirano s polipeptidom izbranim iz skupine, ki vsebuje katerokoli izmed SEQ ID No: od 19 do 26 in polipeptid, ki je vsaj 80% enak in ima enako katalitsko funkcijo kot omenjeni polipeptid kateregakoli izmed SEQ ID No: od 19 do 26.
- 4. Genski grozd, ki kodira biosintetična pot tetraciklina, kjer omenjeni genski grozd vsebuje nukleinsko kislino, ki kodira vse polipeptide iz SEQ ID NO: od 1 do 8, kjer je vsak omenjeni polipeptid iz SEQ ID NO: od 1 do 8 lahko substituiran s polipeptidom, kije vsaj 80% enak in ima enako katalitsko funkcijo kot omenjeni substituirani polipeptid.
- 5. Genski grozd po zahtevku 4, kjer omenjeni genski grozd vsebuje nukleinsko kislino, ki kodira nadaljnji polipeptid izbran iz skupine, ki sestoji iz SEQ ID NO: od 8 do 18, kjer je omenjeni nadaljnji polipeptid lahko substituiran s polipeptidom, ki je vsaj 80% enak in ima enako katalitsko funkcijo kot omenjeni nadaljnji polipeptid.
- 6. Postopek biosintetične proizvodnje spojine tetraciklina, kjer omenjeni postopek obsega naslednje korake: zagotavljanje celice po kateremkoli zahtevku od 1 do 3; zagotavljanje substrata; inkubiranje omenjenega substrata z omenjeno celico pri dovoljenih pogojih, in pridobivanje omenjene spojine tetraciklina.
