Abstract

Prodn. of embryogenic conifer callus comprises culturing somatic tissue, capable of growth, from a conifer t form non-embryogenic callus (NEC). This is then cultured in presence of a nurse culture to form embryogenic callus (EC). Pref. somatic tissue is an explant of needle, bud, shoot or root tissue, partic. taken from a mature tree. The nurse culture comprises EC of juvenile origin, derived from the same species as the NEC being cultured with it. Pref. the EC and NEC are placed close together on opposite sides of a microporous membrane (to allow chemical but not physical contact.
