Abstract
An in vitro method for assaying tPA-activity, optionally in combination with assaying PAI-1-activity. The method is characterized in that it includes the steps (i) of incubating a sample containing tPA with a solid phase that specifically binds tPA under pH conditions which inhibit the formation of a tPA-PAI-1 complex, whereafter (ii) the tPA-activity of the solid phase is determined. The pH conditions mean that PAI-1 will not be denatured irreversibly. If PAI-1 is to be determined functionally, the sample is conditioned with regard to pH prior to step (i). A first portion and a second portion of the sample are then incubated in accordance with step (i) with the exception of the pH conditions for the second portion, which are selected so that a PAI-1-tPA complex can form. The activity of the tPA bound to the solid phase is determined separately for both portions as a final stage, and the PAI-1-activity is calculated from the ratio between the measured tPA-activity of the different sample portions.
