Abstract

1526237 Glucoamylase synthesis CPC INTERNATIONAL Inc 5 Dec 1975 [5 Dec 1974] 50095/75 Heading C3H A process for the production of glucoamylase is characterized by fermenting a microorganism capable of producing glucoamylase in a nutrient medium containing a carbon source, said carbon source comprising liquefied ground corn enzymatically liquefied with a bacterial alphaamylase enzyme preparation derived from Bacillus licheniformis and having a solids content of from 16% to 25% dry substance basis, preferably from 18% to 22%, dry substance basis. The nutrient medium may contain a nitrogen source primarily derived from corn steep liquor solids. Preferred enzyme preparations are selected from Bacillus licheniformis NCIB 8061, NICB 8059, ATCC 6598, ATCC 6634, ATCC 8480, ATCC 9945a and ATCC 11945. The process may be effected by treating an aqueous slurry containing the ground corn at a pH in the range of from 4À5 to about 7À5 and at a temperature in the range of from about 55‹ C. to about 80‹ C. to substantially liquefy said ground corn, then in a second step, increasing the temperature of the fermentation nutrient medium to a temperature of at least about 110‹ C. to sterilize the nutrient medium. Glucoamylase may be obtained by cooling the slurry after the sterilization, inoculating the cooled slurry with the microorganism capable of producing glucoamylase and fermenting the inoculated slurry.
