Способ заключается в том, что раствор, содержащий инсулин или его биотехнологический предшественник, хроматографируют на катионообменнике на основе сополимера метакрилатного мономера с кросс-агентом при рН 4,5 - 5,8, элюируют белки, связавшиеся с катионообменником, в градиенте соли, фракции, содержащие целевой продукт, обессоливают и хроматографируют на анионообменнике на основе сополимера метакрилатного мономера с кросс-агентом, имеющем четвертичные аминогруппы, при рН 9,0 - 10,5. Предпочтительными вариантами осуществления способа являются использование в качестве катионообменника сорбента на основе сополимера метакрилатного мономера с кросс-агентом, модифицированного фосфоргруппами, и осуществление обессоливания с помощью ультрафильтрации. Обессоливание элюата можно осуществлять и с помощью ступенчатой гидрофобной хроматографии, используя сорбент на основе сополимера метакрилатного мономера с кросс-агентом, модифицированный алкильными группами. Preferred embodiments of the process are the use of a sorbent based on a copolymer of a methacrylate monomer with a cross-agent modified with phosphorus groups as a cation-exchanger and the implementation of desalting using ultrafiltration. Desalting of the eluate can also be carried out using stepwise hydrophobic chromatography using a sorbent based on a copolymer of a methacrylate monomer with a cross-agent modified with alkyl groups. The method allows to obtain insulin or its biotechnological precursors of high purity - more than 97.5%.
