Далее была проведена кодон - оптимизация под клеточную линию яичников китайского хомячка СНО-K1 в онлайн сервисе https://www.thermofisher.com для увеличения выхода целевого белка. Синтез праймеров (табл. 1) и нуклеотидной последовательности, кодирующей тример GPC LASV, был осуществлен в коммерческой научно-производственной фирме OOO «ДНК-Синтез» (г. Штамм клеток яичника китайского хомячка CHO-K1-GPC-LASV, получен на основе клеточной линии яичников китайского хомячка СНО-K1 с использованием разработанной конструкции PVEAL3-Lassa-Trim. Суспензию клеток CHO-K1, растили в инкубаторе при 5% содержания CO2, 80%-ной влажности температуре (37±1)°С во влажной атмосфере 5%СО2 и скорости вращения платформы 185 об/мин. При достижении плотности суспензии 5х106 клеток/мл проводили трансфекцию клеток с помощью полиэтиленамина (PEI 25K™) в соответствии с инструкцией производителя. Трансфецировали клетки смесью плазмид PVEAL3-Lassa-Trim и pCMV(CAT)T7-SB100, взятых в соотношении 1:10. Через 48 часов после трансфекции добавляли селективный антибиотик пуромицин в концентрации 2 мкг/мл. Селекцию проводили в течение 12 суток с постепенным увеличением концентрации антибиотика с 2 до 10 мкг/мл. Полученный пул клеток был криоконсервирован, после чего было проведено клонирование с целью отбора индивидуальных клонов с высокой продукцией. Клонирование выполнялось методом лимитирующих разведений в 96-луночных планшетах. Для идентификации тримеризованного белка поверхностного гликопротеина GPC LASV использовали иммуноферментный анализ. В качестве антитела брали нейтрализующее моноклональное антитело 37.7H против LASV. Культуральную жидкость, содержащую тримеризованный GPC LASV разводили в соотношение 1:100 в растворе натрия углекислого кислого (pH 8,0) и вносили в лунки планшета для ИФА по 150 мкл. Антиген инкубировали при 4°С в течение 16 часов, а затем удалили из лунок путем встряхивания. Блокировка проходила при добавлении в лунки к сорбированным антигенам по 200 мкл блокирующего буфера (фосфатно-солевой буфер (ФСБ) + 1% BSA) и инкубации при 37°С в течении 2 часов. После удаления растворов четырехкратно промыли лунки промывочным буфером ФСБ-0,5% Tween20 (ФСБ-Т). Для проверки специфичности тримера использовали рекомбинантные человеческие моноклональное антитела 37.7H, iB20 (отрицательный контроль) и MR191 (отрицательный контроль). Описание антител приведено выше. Антитела разводили в блокирующем буфере до концентрации 4 мкг/мл и вносили в лунки по 150 мкл. Планшеты с растворами инкубировали 2 часа при 37°С. Затем отмывали лунки четырехкратно промывочным буфером и в лунки вносили по 150 конъюгата Goat anti-human IgG-HRP (Sigma-Aldrich, США) в разведении 1:30.000. Инкубировали 2 часа при 37°С и промыли лунки четырехкратно с ФСБ-Т. Для визуализации результатов в лунки вносили по 100 мкл 0,02% раствора хромогена ТМБ в цитрат-фотфатном буфере с перекисью водорода (БРС). После 15 мин инкубации при 37°С в лунки нанесли по 50 мкл 0,9М раствора серной кислоты в качестве стоп-реагента. Результаты регистрировали в планшетном ридере при длине волны 450 нм.
