Abstract
Description
The invention relates to the veterinary field and concerns the use of L-ornithine-L-aspartate as a hepatoprotector in domestic animals.
- 2012. - №6).In modern animal husbandry, the problem of fatty hepatosis in domestic animals is the most economically significant. Fatty degeneration of the liver is detected in 80% of culled cows (Mishchenko V.A. et al. Analysis of metabolic disorders in high-yielding cows // Veterinary Kuban. - 2012. - No. 6).
Fatty hepatosis is a pathological syndrome in which an excessive amount of lipids (fats) accumulate in the liver and dystrophic changes in its functional cells (hepatocytes) occur. As a result of degeneration, hepatocytes are replaced by fibrous tissue.
Hepatoprotectors are a broad class of drugs for the treatment of fatty liver and other liver diseases. The funds included in this group can be used at different stages of organ damage, but mainly before the development of cirrhosis. Hepatoprotectors act in a complex way:
- стимулируют регенерацию клеток печени;- stimulate the regeneration of liver cells;
- восстанавливают гомеостаз;- restore homeostasis;
- нормализуют функциональную активность печени;- normalize the functional activity of the liver;
- повышают устойчивость гепатоцитов к неблагоприятным факторам.- increase the resistance of hepatocytes to adverse factors.
№4 - 2016. - С. 42-45). Until recently, the arsenal of means for the prevention and treatment of fatty liver hepatosis was extremely small (Semenenko M.P. et al. Etiopathogenesis and features of hepatotropic therapy of cows with hepatoses. Veterinary Medicine. No. 4 - 2016. - P. 42-45).
A known method for the treatment and prevention of hepatosis in animals (RF Patent No. 2385728, 10.04.2010), including injections of hydrolyzed liver tissues and mineral salts to sick cows and calves. Intramuscular doses for cows are 20-40 ml, for calves 10-12 ml every 7-10 days, 5-6 injections. Clinical efficacy is quite high. However, tissue preparations from animal organs pose a potential hazard against prion infections such as spongiform encelophalopathy. The injection method of using the drug cannot be considered effective enough for the most severe metabolic pathologies, such as ketosis, hepatosis, mycotoxicosis.
From the patent of the Russian Federation RU 2500402 C2, a method is known to increase the ability to learn in an animal, which involves the introduction of an edible composition to the animal, including one or more unsaturated fatty acids (FFA) in an amount of from 0.1% to 50%, one or more compounds releasing nitric oxide (NORC) - L-ornithine L-aspartate, according to one of the variants of the invention, in an amount of 0.1% to 20%, as well as a composition contains 0.1-40 times more B vitamins than the recommended daily intake of B vitamins, and from 0.0001% to 25% antioxidants, while the composition is formulated as a food composition for animals or a dietary supplement, and the food composition additionally contains from 15% to 50% protein, from 5% to 40% fat, from 5% to 10% minerals and has a moisture content from 5% to 20%. The disadvantage of this technical solution is that the composition is not intended for the treatment of hepatosis.
From RF patent RU 2734417 C2 a group of inventions related to the pharmaceutical industry is known, in particular to a method for manufacturing an effervescent composition based on ornithine aspartate, according to which a mixture containing ornithine aspartate and a gas-forming component, which is sodium bicarbonate, is granulated to obtain granules G; then the granules G are mixed with a sweetener and with at least one component, which is a pharmaceutically acceptable polybasic organic acid or its partial salt with alkali or alkaline earth metals, to obtain the specified effervescent composition. The disadvantage of this technical solution is that it is not intended for the treatment of pets.
As the closest analogue, a composition is known for stimulating energy-metabolic processes and a method for preventing birth pathologies and postpartum diseases of cows, including the use of succinic acid and beet molasses as an energy stimulant before and after calving (RF Patent No. 2553360, 05/18/2015). This composition is technologically simple, environmentally friendly and efficient. However, in the course of clinical trials, it turned out that this composition is not optimal for the prevention of fatty hepatosis.
The objective of the invention is to develop an effective, technologically simple, environmentally safe method for the prevention of fatty hepatosis in pets.
To solve this problem, it is proposed to use a complex composition in which L-ornithine-L-aspartate, used as a feed additive, acts as an active hepatoprotector.
As excipients, the following can be used: lemon flavor - 0.015 g, mint flavor - 0.005 g, aspartame - 0.05 g, anhydrous citric acid - 0.5 g, mannitol - 1.430 g.
All components of the composition are selected taking into account their metabolic role and complex effects on all pathogenetic links underlying the development of fatty hepatosis in domestic animals. The manufacture of the test compositions is illustrated by the following examples.
The object of clinical trials were domestic animals (domestic dogs, weighing 10-32 kg) with the first signs of loss of appetite and weight loss. It is during this period that the risk of intensive involvement in the energy processes of fat reserves of one's own body is highest.
Example 1. Evaluation of the effectiveness of a hepatoprotective composition in domestic animals with clinically significant signs of fatty hepatosis
Оценка эффективности проведена по результатам клинических наблюдений (таблица 1) и биохимических исследований сыворотки крови (таблица 2).Taking into account the clinical condition of the animals, the hepatoprotective compositions were applied by forced feeding. The course of application is once a day for 5 days. Efficacy was evaluated based on the results of clinical observations (table 1) and biochemical studies of blood serum (table 2).
Signs of a pronounced improvement in the clinical condition began to appear in domestic animals of the first experimental group after 2 hours, while in animals of the second group a similar effect developed after an hour. On the second day, all domestic animals of the second group were fed more actively than those from the first group. In general, the clinical effect was more pronounced in domestic animals of the second group. Animals of both experimental groups on the 3rd day showed a pronounced tendency to increase motor activity without reducing body weight. On the contrary, in pets of the control group during this period, there was a worsening of the clinical condition, a decrease in activity and a progressive decrease in body weight.
Biochemical research. The initial state. Low rates of reserve alkalinity, within 15-16 mmol/l, indicated acidification of the body (metabolic acidosis). A low glucose level indicated the development of an energy-deficient state. The increased content of triglycerides is about the involvement in the energy metabolism of fat reserves of one's own body. Pronounced deviations of biochemical parameters indicated a violation of the metabolic functional activity of the liver.
Repeated biochemical studies (on day 10) showed pronounced positive changes in protein, carbohydrate and lipid metabolism in domestic animals of both experimental groups. First of all, the triglyceride content decreased to the upper level of the physiological norm. Indicators of reserve alkalinity and glucose increased to average physiological values. The indicator of total protein content decreased to the level of average physiological values. Thus, the use of hepatoprotective compositions in progressive fatty hepatosis provided the clinical effect of normalizing not only lipid, but also protein and carbohydrate metabolism.
Example 2 Prophylactic Efficacy of Hepatoprojective Compositions Under Production Test Conditions
The experimental groups included pets, at the first signs of weight loss and loss of appetite.
3.Procedure for conducting clinical trials. Hepatoprotective compositions according to prescription 1 were used on domestic animals of the first group, and according to prescription 2 on animals of the second group. Multiplicity 1 time in 5 days. Duration 40 days. Method of application - individual irrigation of feed. Domestic animals consumed it with great desire for food. The results of clinical observations and the condition of the animals are presented in Table 3.
Thus, in the course of clinical trials, it was found that the use of hepatoprotective compositions, during the period of intensive involvement in the energy metabolism of lipids of one's own body, provides a rapid recovery of the metabolic function of the liver and effectively prevents the development of fatty hepatosis in pets.
