Abstract
Description
The invention relates to organic chemistry, specifically to the synthesis of ortho- and para-aminophenols, which are important raw materials for the chemical and pharmaceutical industries. Thus, ortho-aminophenol is used as a dye for fur, in photography as a developer, and in the synthesis of certain drugs. Para-aminophenol is used as a component of dyes for fur, a black-and-white developer in photography, and in the synthesis of drugs, in particular paracetamol.
II).A two-stage method for producing ortho- and para-aminophenols is known. At the first stage, the nitration of phenol is carried out, at the second stage, the corresponding nitrophenol is reduced in the presence of iron filings in an acidic environment (A.E. Chichibabin “Basic Principles of Organic Chemistry”, vol. II).
Нитрофенолы, в свою очередь, можно получить нитрованием фенола [US 3663261 (1973), US 3517075 (1971), US 3510527 (1971)], гидроперекиси кумола [US 3388173 (1968)], из нитрохлорбензола [US 3263011 (1966)], окислением нитрозофенола [пат. СРР 53445 (1971), RU 2076095 (1997)].Nitrophenols, in turn, can be obtained by nitration of phenol [US 3663261 (1973), US 3517075 (1971), US 3510527 (1971)], cumene hydroperoxide [US 3388173 (1968)], from nitrochlorobenzene [US 3263011 (1966)], oxidation of nitrosophenol [pat . CPP 53445 (1971), RU 2076095 (1997)].
So, when phenol is nitrated with a nitrating mixture consisting of nitric, sulfuric acids and nitrogen oxides [US 3663261 (1973), US 3517075 (1971)] or liquid nitrogen oxides in nitric acid [US 3510527 (1971)], a mixture of ortho- and para-nitrophenols is obtained with a yield of 17 and 73%, respectively. The disadvantage of this method is the use of toxic nitrogen oxides.
When cumene hydroperoxide is nitrated with a mixture of nitric acid and nitrogen oxides in chloroform, a mixture of ortho- and para-nitrophenols is also obtained with yields of 30 and 7%, respectively [US 3388173 (1968)]. The disadvantages of this method, along with the use of toxic nitrogen oxides, is the use of unstable cumene hydroperoxide, as well as the need to separate ortho- and para-nitrophenols by steam distillation.
According to [US 3263011 (1966)], when 4- or 2-nitrochlorobenzenes are heated in an autoclave in a 2-4% aqueous solution of alkali at 160°C with continuous air blowing, the corresponding nitrophenol is obtained. The disadvantage of this method for obtaining nitrophenols is the process under pressure, as well as the non-selectivity of nitration of chlorobenzene, which requires the separation of ortho- and para-isomers by steam distillation.
Para-nitrophenols can also be obtained by oxidation of para-nitrosophenol. So, according to [US Pat. CPP 53445 (1971)] para-nitrosophenol is oxidized with nitric acid, and according to [RU 2076095 (1997)] - with 33% hydrogen peroxide in the presence of Na 2 WO 4 ⋅2H 2 O catalyst. in the second, an expensive and toxic catalyst is used.
Para-nitrosophenol can be reduced to para-aminophenol in ethyl acetate in the presence of a Pd/C catalyst at a hydrogen pressure of 2.0–4 atm and a temperature of 20–50°C [RU 2461543 (2012)]. The disadvantage of the method is the process under pressure, the use of an expensive catalyst (Pd), as well as environmentally unsafe solutions of concentrated acids (HCl, HNO 3 ) and alkalis.
A known method for producing aminophenols by photooxidation of aromatic amines with oxygen by irradiation with visible light in an aqueous organic solvent in the presence of active and stable photosensitizers, which are substituted phthalocyanines [RU 2567552 (2015)]. The yield of p-aminophenol is 70%. However, this method involves the use of complex special equipment - a laser or LEDs.
Максимальный выход пара-аминофенола (72%) достигается при продолжительности окисления анилина 15 ч, при этом их реакционной смеси выделен также орто-аминофенол (13%) (Заявка на патент РФ №2021130569).The closest to the claimed method for obtaining ortho- and para-aminophenols is a two-stage process, in the first stage of which aniline is oxidized with ammonium persulfate in an alkaline medium at 40-60 ° C, in the presence of 0.01-0.03 wt% phthalocyanine oxidation catalyst (phthalocyanine cobalt, iron (II), iron (III), manganese, nickel or zinc), followed by cooling the reaction mixture, isolation of intermediate 4(2)-aminophenyl sulfates. By hydrolysis of the latter in an acidic medium at 85-95°C, a mixture of ortho- and para-aminophenols is obtained, from which ortho-aminophenol is isolated by extraction with chloroform. Para-aminophenol remains in the aqueous phase. Depending on the duration of aniline oxidation, the yield and ratio of the formed ortho- and para-isomers varies. The maximum yield of para-aminophenol (72%) is achieved when the duration of aniline oxidation is 15 h, while ortho-aminophenol (13%) is also isolated from the reaction mixture (RF Patent Application No. 2021130569).
The problem to be solved by the present invention is to increase the yield of target compounds.
1, п. 6).The problem is solved by the proposed one-pot method for obtaining ortho- and para-aminophenols. Aniline is oxidized with ammonium persulfate (PSA) or potassium (PSK) or sodium (PSN) and hydrogen peroxide as the second oxidizing agent, in an alkaline medium at a molar ratio of Aniline:NaOH:PSA (PSK, PSN):H 2 O 2 equal to 1:5:1.5:2.0 at 40-50 ° C for 2-18 hours. Then the reaction mixture is cooled to room temperature, hydrochloric acid is hydrolyzed oh at 85-95 ° C and a mixture of o- and p-aminophenols is obtained, from which o-aminophenol (o-AF) is isolated by extraction with chloroform (yield up to 30%), p-aminophenol (p-AF) remains in the aqueous phase (yield up to 75%). It should be noted that the duration of aniline oxidation (2–15 h) affects the ratio of ortho- and para-aminophenols isolated after hydrolysis of intermediate 4(2)-aminophenyl sulfates (table). Thus, during the oxidation of aniline for 15 h, only one isomer, para-aminophenol, was isolated from the reaction mixture in a 75% yield (Table 1, p. 6).
The essence of the claimed technical solution is confirmed by examples of a specific implementation.
