Invention prototype
The closest in purpose and set of essential features is the method of using platelet autoplasma in caesarean section. To obtain platelet-rich autoplasma, 15 ml of venous blood was taken into test tubes.
Purpose of the invention
The aim of the invention is to activate the stimulation of tissue regeneration and the healing process of a postoperative wound on the uterus during caesarean section. This goal is achieved due to the ability of platelets to secrete substances that stimulate the process of tissue regeneration and regulate both angiogenesis and inflammatory reactions.
The essence of the invention
The method proposed as an invention is as follows:
е. To achieve this goal, blood taken from the central vein of the puerperal is centrifuged at a speed of 580g in 9-ml tubes containing sodium citrate for 8 minutes, which leads to the separation of red and white blood cells from platelet-rich plasma. The two Endoret® (PRGF®) fractions are separated from the rest of the blood components using an extraction device (PTD: *plasma transfering device). Further, immediately before use, in the 2nd fraction of Endoret® (PRGF®) is 2 ml of the plasma layer above the buffy coat, add 50 µl (0.05 cm3) of the activator PRGF®-Endoret® (calcium chloride) per 1 cm3 of plasma. The amount of activator is measured with an insulin syringe calibrated per µl and then added to the plasma. Endoret® clot forms in 5-8 minutes. The formed clot is distributed along the postoperative suture on the uterus, then it is fixed during suturing with the vesicouterine fold, i.e. with peritonization.
Endoret® (PRGF®) contains a mixture of autologous growth factors that are derived from both plasma and platelets.
Platelets secrete substances that stimulate tissue regeneration and regulate both angiogenesis and inflammatory responses. The most important among all other factors are PDGF: platelet-derived growth factor, TGF-β: beta-transforming growth factor, bFGF: basic fibroblast growth factor, VEGF: vascular endothelial growth factor, EGF: epidermal growth factor or angioprotein-1. They simultaneously release antibacterial molecules and special growth factors that affect the mobilization of stem cells from the bone marrow or peripheral niches.
Calcium acts as a cofactor in the Endoret® (PRGF®) activation process, which allows the conversion of plasma fibrinogen to fibrin, resulting in the formation of a gel or clot with important biological functions. Fibrin is an excellent matrix for containing and locating cells and acts as a temporary scaffold while certain tissue is being regenerated, as well as a permanent growth factor secretion system. Thus, it is a biocompatible and autogenous sponge filled with growth factors and cytokines, allowing their progressive release over several weeks.
The angiogenesis action of growth factors is decisive for the start of the regeneration process. One of the main properties of growth factors is their anti-inflammatory and antibacterial properties.
