Abstract
Description
The invention relates to the field of agriculture, in particular to poultry farming, and in particular to the processing of hatching quail eggs.
Modern poultry farming faces a number of urgent problems that require immediate solutions. One of these is the increase in the output of conditioned chickens, which largely depends on the period and conditions of storage of hatching eggs.
When carrying out the incubation of eggs, the main goal is to obtain the maximum possible number of healthy young animals by reducing the mortality of embryos and maintaining the optimal level of their metabolism during critical periods of incubation.

When processing by the aerosol method, at the moment of cooling the heated eggs, not only the solution, but also air is absorbed, while a significant part of the solution simply drains from the eggs; with insufficient moisture, the surface of the shell does not fully come into contact with the solution, as a result, part of the biologically active substances planned for delivery to the egg does not enter it.
The introduction of biologically active substances in ovo requires large labor (introducing a solution with a needle into each egg) or material (purchase of expensive equipment) costs. In this case, the integrity of the shell is damaged, which, if the rules of sanitation are not adequately observed, can lead to damage to the hatching eggs.
When using the method of immersing heated eggs in a cold solution, the solution contacts the entire surface of the shell, the maximum possible amount of a solution of biologically active substances without air is sucked into the egg, and the integrity of the shell is not violated.

There are very few published data on the stimulation of avian embryogenesis with solutions of vitamin B 2 (riboflavin).
М. Riboflavin-Binding Protein induces early death of chicken embryos. / Lee С.М., White H.B. - J. Nutr. - 1996 - 126: pp. 523-528].Foreign scientists have found that maternal riboflavin-binding protein (RfBP) contributes to the deposition of riboflavin in bird eggs. Chickens genetically unable to synthesize RfBP produce riboflavin-deficient eggs. Embryos in such eggs die around day 13 of incubation with severe hypoglycemia and impaired fatty acid oxidation. An injection of free riboflavin prior to incubation allows these embryos to survive and hatch normally. At the same time, the presence of RfBPB in excess can reduce the availability of riboflavin to the chick embryo, since this protein presumably absorbs a small amount of riboflavin, usually available to the embryo in the early stages of development [Lee C.M. Riboflavin-Binding Protein induces early death of chicken embryos. / Lee S.M., White H.B. - J. Nutr. - 1996 - 126: pp. 523-528].
-2010. -Т. 12. - №2. - С. 269-275].Other investigators have studied the effect of in ovo administration of high doses of riboflavin (0, 300, 1500 and 3000 µg/egg) to hatching Ross 308 broiler eggs before incubation and on day 6. They found that the injection of riboflavin into the egg before the start of incubation led to an increase in embryonic mortality in the early stages of embryogenesis. The use of riboflavin at a dose of 3000 μg / egg reduced the fattening performance of broiler chickens [Glodek K. Injection of large doses of riboflavin, which is administered inovo, on the friability of chickens and the appearance of chicken broilers / K. Glodek, M.V. Lis, B. Plitich, A. Mazur, J.V. Nedzyulka // Biology of creatures. -2010. -T. 12. - No. 2. - S. 269-275].
Он участвует в окислительно-восстановительных реакциях, является коферментом многих ферментов, участвующих в процессах клеточного дыхания, обмена и активации других витаминов, метаболизма липидов, лекарственных средств и ксенобиотиков, формирования структуры белка, синтеза гормонов и передачи информации между клетками [https://stylab.ru/directory/constituents/vitamins/b2/].At the same time, vitamin B 2 is of the greatest importance for birds from all B vitamins. It participates in redox reactions, is a coenzyme of many enzymes involved in the processes of cellular respiration, metabolism and activation of other vitamins, lipid metabolism, drugs and xenobiotics, formation protein structure, hormone synthesis and information transfer between cells [https://stylab.ru/directory/constituents/vitamins/b2/].
Brown W.O. При инъекции витамина В2 в яйцо он полностью использовался эмбрионом [https://www.activestudy.info/vliyanie-kormleniya-ptic-na-ix-produktivnost/].With its lack in the feed of laying hens, the hatchability of eggs is greatly reduced, and the hatched chicks suffer from paralysis. According to a study by Tretyakova E.I., with a lack of riboflavin in the diet of poultry, changes in the physicochemical parameters of protein and yolk are observed in eggs. Brown WO observed the direct transfer of riboflavin from the egg to the chicken embryo, the largest amount was absorbed by the embryo on the 14-15th day of incubation, and the mortality of embryos with a lack of vitamin B 2 was the highest at this time. When vitamin B2 was injected into an egg, it was completely used by the embryo [https://www.activestudy.info/vliyanie-kormleniya-ptic-na-ix-produktivnost/].
The closest to the claimed solution is the work of Dmitrieva O.S. [Dmitrieva O.S. The effect of riboflavin on the body weight and eyes of chicken embryos in antenatal ontogenesis // Proceedings of the Velikiye Luki State Agricultural Academy - 2017. - No. 4. - S. 2-7]. The eggs heated in the incubator were placed in a 0.001% solution of vitamin B 2 at room temperature for 20 minutes, then the eggs were incubated under a stable incubation mode. In this method, only the effect of riboflavin on the increase in the mass of the eyeball and the chick embryo as a whole was monitored. The disadvantage of this method is that the eggs were processed prior to incubation and the solution got into the outer liquid protein, and not directly to the embryo. Exposure of eggs in the riboflavin solution for 20 minutes is time-consuming to produce and excessive, since the eggs cool quite quickly (within 3 minutes), and after cooling the solution no longer penetrates the egg.
The technical objective of the claimed invention is to increase the hatchability of eggs and the output of conditioned young animals, by increasing the viability of the embryos.
The technical result of the method for increasing the viability of quail embryos includes incubation, processing eggs on the 13.5th day of incubation by immersion in a container with a 0.03% riboflavin solution having a temperature of 10-16°C for 3 minutes, followed by placing the eggs back into the incubator until the end incubation.
The proposed method allows the solution of riboflavin to enter directly into the bloodstream of the developing embryo and during the period of greatest need for this vitamin. So, on the 13.5th day, allantois, consisting of a network of blood vessels, covers almost the entire inner surface of the shell (with the exception of the air chamber), performing both the respiratory function and the function of transporting shell mineral substances to form the bone structure of the embryo. When heated eggs are immersed in a 0.03% riboflavin solution, which has a temperature of 10-16 ° C, the entire contents of the egg are cooled, it is compressed and absorbed through the pores of the shell of the solution surrounding the egg, where it is absorbed by the allantois vessels.
The method is carried out as follows.
Quail eggs are incubated at 37.6°C and 51% humidity. On the 13.5th day of incubation, the eggs are removed from the incubator and immediately in the trays are immersed in a container with a 0.03% riboflavin solution having a temperature of 10-16°C for 3 minutes. The egg trays are then removed from the container and placed back into the incubator to continue incubation.
Examples of specific implementation of the method
The experiments were carried out on eggs of quails of the Pharaoh breed at the SibNIIP-branch of the Federal State Budgetary Scientific Institution "Omsk ARC". Eggs are obtained from one parent flock, the collection period is not more than 3 days. Groups were formed according to the principle of analogues, taking into account the shelf life and weight of eggs.
The purpose of the experiment is to establish the optimal time for quail eggs to stay under water without worsening the results of incubation. Three groups of 200 eggs each were formed, the eggs were immersed in water immediately after being removed from the incubator on the 13.5th day of incubation, the immersion time was 1, 3 and 5 minutes, the water temperature was 10-16°C.
The water temperature range is chosen with the expectation that at 10°C it is allowed to store hatching eggs without harm to the embryo. A solution temperature of more than 16°C is undesirable, since when an egg heated to 37.6°C is immersed, its temperature becomes the same with the water temperature much faster and the effect of absorption of the solution into the egg quickly stops.
2,02%.On the 13.5th day of incubation, quail embryos are already actively absorbing oxygen from the environment, so immersion under water for a long time can be detrimental to them. The data in Table 1 indicate that the duration of egg treatment on the 13.5th day of incubation by immersion up to 3 minutes does not reduce the results of incubation. Increasing the time spent incubating eggs under water up to 5 minutes, relative to the group with a 3-minute immersion, led to a decrease in hatchability of eggs by 6.86%; hatching of young animals - by 6.57% due to an increase in the amount of incubation waste of the category "frozen 4-15.5 days" - by 1.01%, "suffocating" - by 3.03%, "weak and crippled" - by 2, 02%.
The purpose of the experiment is to determine the optimal concentration of the riboflavin solution. Formed 4 groups of 100 eggs each: control - not treated; experimental groups 1, 2 and 3 were treated on the 13.5th day of incubation by immersion for 3 minutes in riboflavin solutions at a concentration of 0.01; 0.03 and 0.05%, respectively, the temperature of the solution is 10-16°C.
В результате исследований установлено, что максимальный положительный эффект оказал раствор рибофлавина в концентрации 0,03% (2 опытная группа) (таблица 2).As a result of the research, it was found that the riboflavin solution at a concentration of 0.03% (experimental group 2) had the maximum positive effect (table 2).
14,99%.Thus, the treatment of eggs on the 13.5th day of incubation with a solution of riboflavin at a concentration of 0.03% made it possible to increase, in comparison with the control, the hatchability of eggs and the hatching of young animals by 14.45 and 14.99%.
Embryos of the control group lacked riboflavin and the introduction of its solution into the eggs contributed to the optimization of lipid metabolism, redox reactions and tissue respiration.
An increase in the concentration of the solution to 0.05% led to a decrease in the hatchability of eggs and the hatching of young animals relative to the 2nd experimental group by 13.64 and 13.48%, which indicates an excess and possible hypervitaminosis of riboflavin for embryos.
The purpose of the experiment was to determine the optimal age of embryos for egg treatment with 0.03% riboflavin solution. Formed 4 groups of 300 eggs each: control - not treated; experimental group 1 - the egg heated for 2 hours at 38°C was treated by immersion for 20 minutes 1 hour before incubation; groups: 2, 3 and 4 - at 11.5, 13.5 and 15.5 days of incubation immediately after removal from the incubator - by immersion for 3 minutes, the temperature of the solution was 10-16°C.
При проведении погружения яиц в раствор до инкубации разница с контрольной группой по выводимости яиц и выводу молодняка составила всего 2,25 и 1,97% (таблица 3).When the eggs were immersed in the solution before incubation, the difference with the control group in terms of egg hatchability and young hatchlings was only 2.25 and 1.97% (Table 3).
An increase in the positive effect was obtained when eggs were immersed in a solution of riboflavin during the period of intensive development of allantois vessels after 11 days of incubation.
The processing of eggs on the 13.5th day of incubation made it possible to increase the hatchability of eggs and the hatching of young animals by 8.00 and 7.35% relative to the control group by reducing the amount of incubation waste of the category "frozen 4-15.5 days", "weak and crippled".
When processing eggs on days 11.5 and 15.5 of incubation, the effect was less pronounced, however, experimental groups 2 and 4 also exceeded the control in hatching eggs and hatching young animals by 6.47 and 5.84%; 6.18 and 5.34%, respectively, to a greater extent due to a decrease in the amount of incubation waste of the categories “suffocating” and “weak and crippled”.
7,35%.The results of the studies showed that the proposed method for increasing the viability of quail embryos by treating hatching eggs with a solution of riboflavin in the second half of incubation (on day 13.5) is effective, since the vitamin enters the bloodstream of the embryo immediately, which leads to optimization of lipid metabolism, oxidative - recovery reactions and tissue respiration, due to which the hatchability of eggs increases by 8.00%, the hatchability of young animals - by 7.35%.
