Изобретение относится к области биотехнологии растений и может быть использовано для массового получения качественного посадочного материала бельвалии сарматской (Bellevalia sarmatica (Pall, ex Georgi) Woronow), растения, занесенного в Красную Книгу РФ и находящегося под угрозой исчезновения (статус редкости - 2 - сокращающиеся в численности) [2]. Полученные растения могут использоваться для пополнения интродукционных коллекций ботанических садов и возвращения в природные места обитания, а также для ландшафтного озеленения. Изобретение позволяет получить большое количество выровненного экологически чистого посадочного материала.The invention relates to the field of plant biotechnology and can be used for the mass production of high-quality planting material of Sarmatian Belvalia (Bellevalia sarmatica (Pall, ex Georgi) Woronow), a plant listed in the Red Book of the Russian Federation and endangered (rare status - 2 - decreasing in number) [2]. Бельвалия сарматская - многолетнее луковичное растение. Высота до 20 до 30 см. Листья ремневидные, сосредоточены у основания стебля. Цветет многоцветковой кистью, цвет серо-бурый, цветоножки - поникающие, при созревании плода удлиняются. Плод - продолговатая коробочка с 2-4 семенами. Цветет - весной. Размножается семенами, зацветает на 5-й год после посева. Изобретение представляет собой способ размножения растений бельвалии сарматской (Bellevalia sarmatica (Pall, ex Georgi) Woronow) методом культуры in vitro. включающий стерилизацию семян, высаживание на питательные среды, собственно размножение (Кр=7.8), последующее укоренение и адаптацию микролуковиц к условиям ex vitro. В качестве первичных эксплантов использовали семена.Seeds were used as primary explants.
Оптимальная система стерилизации: 1.) фунгицид - 30 минут: 2.) 70% этанол - 2 минуты; 3.) 7% гипохлорит кальция - 20 минут.Optimal sterilization system: 1.) fungicide - 30 minutes: 2.) 70% ethanol - 2 minutes; 3.) 7% calcium hypochlorite - 20 minutes.
На этапе собственно микроразмножения исследовали влияние регуляторов роста и их концентраций на коэффициент размножения. Наибольшие значения морфометрических показателей были получены на питательных средах, содержащих 10 мг/л 6-БАП с добавлением 0.1 мг/л НУК. Дальнейшее увеличение концентрации регуляторов роста не дало положительного эффекта (фиг. 1). Максимальный коэффициент размножения В. sarmatica, равный 7,8±1.95, достигается при культивировании на питательной среде, содержащей 6-БАП (10 мг/л) и НУК (0,1 мг/л).At the stage of micropropagation proper, the effect of growth regulators and their concentrations on the reproduction rate was investigated.
