Description
The invention relates to the field of medical biotechnology, in particular, to the production and use of cell lines used for cell technology in medicine, among which the creation of biomedical cell products - antitumor vaccines, diagnostic kits and test systems for the development of new drugs and new therapeutic approaches, testing the activity of various pharmaceuticals.
- 2016. - Т. 20(6). - С. 945-948. DOI 10.18699/VJ16.215).The progress of modern biomedical science and biotechnology is largely due to the use of in vitro cultured cells of various origins. The method of cell culture is unique and at the same time is widely used at present, both in practical and in scientific research. It became possible to use animal cells to obtain genetically engineered drugs, substitution therapy, the production of drugs for immunoprophylaxis, diagnosis and treatment largely depends on the stability and reproducibility of the properties of the cells used. The basis for any safe bio-product should be a standardized and certified cell culture. Currently, the use of such cell lines is an essential condition for research or the development of new technologies, as well as to ensure stable and safe production of biotechnological preparations (Menzorov A.G., Serov OL.Why do we need collections of cell lines // Vavilovsky Journal of Genetics and selection. - 2016. - T. 20 (6). - S. 945-948. DOI 10.18699 / VJ16.215).
Однако рак почки относится к группе новообразований, чувствительных к иммунотерапевтическому воздействию (Cho Y.H., Kim M.S., Chung H.S., Chang E.Novel immunotherapy in metastatic renal cell carcinoma // Investig Clin Urol. - 2017 - Vol. 58(4). - P. 220-227. doi: 10.411 l/icu.2017.58.4.220).Kidney cancer is one of the leading growth rates among oncourological diseases. According to statistics, in 2016, 108.9 cases of malignant neoplasms of the kidney were recorded in Russia per 100,000 thousand of the population, compared with 78.5 in 2011 (Edited by A.D. Kaprin, V.V. Starinsky, G.V. Petrova The state of oncological care for the population of Russia in 2016. - M.: P. Herzen MNIIO - branch of FSBI “NIIRTS” Ministry of Health of Russia, 2017. - ill. - 236 pp. ISBN 978-5-85502 -231-5). The course of this disease is such that in 25-50% of patients at the time of diagnosis, metastases are already being determined, in about half of patients, the disease will acquire a systemic character at different times after surgical treatment. The biological feature of renal cell carcinoma is the ability to metastasize to any organs and tissues of the body. Traditional types of systemic treatment prescribed for common forms of tumors are ineffective in patients with renal cell carcinoma. However, kidney cancer belongs to the group of neoplasms sensitive to immunotherapeutic effects (Cho YH, Kim MS, Chung HS, Chang E. Novel immunotherapy in metastatic renal cell carcinoma // Investig Clin Urol. - 2017 - Vol. 58 (4). - P . 220-227. Doi: 10.411 l / icu.2017.58.4.220).
Neoantigens in cancer immunotherapy // Science. - 2015. - Vol. 348, Issue 6230. - P. 69-74. DOI: 10.1126/science. 4971). Н., Sckisel G.D., Murphy W.J. The role of antigen-specific and non-specific immunotherapy in the treatment of cancer // J Immunotoxicol. - 2012. - Jul-Sep; 9(3). - P. 248-58. - doi: 10.3109/1547691X.2012.685527). (Amin A., Dudek A.Z., Logan T.F. et al. Survival with AGS-003, an autologous dendritic cell-based immunotherapy, in combination with sunitinib in unfavorable risk patients with advanced renal cell carcinoma (RCC): Phase 2 study results // J Immunother Cancer. 2015 Apr 21; 3:14. doi: 10.1186/s40425-015-0055-3).Active immunotherapy includes: 1) non-specific immunotherapy (inducing an active inflammatory process, non-specific vaccination, local and systemic use of BCG, the use of levamisole, interferons α and γ, tumor necrosis factor, interleukins, colony-stimulating factors, high doses of antiestrogens), 2) specific immunotherapy with immunization with tumor antigens (vaccine therapy), 3) gene therapy. Currently, a search is underway for tumor-associated antigens (OAAs), the use of which as targets for immunotherapy would bring these methods to a whole new level (Schumacher TN, Schreiber RD Neoantigens in cancer immunotherapy // Science. - 2015. - Vol. 348, Issue 6230. - P. 69-74. DOI: 10.1126 / science. 4971). The process of creating and selecting tumor cell lines capable of stably producing high levels of OAA is the basis for the production of antitumor cell vaccines, including dendritic cell (DC) vaccines. The creation of antitumor vaccines using DC is a promising direction in the treatment of cancer, as it allows you to activate the protective system of the patient’s body, using the natural ways of recognizing tumor antigens and their subsequent elimination. Tumor antigens obtained from destroyed tumor cells are more suitable for loading DK, since in this case the whole set of tumor-associated and tumor-specific antigens enters the cell (Baldueva I.A. Immunotherapy of malignant neoplasms: a look into the future. Allergology and Immunology - 2014 . - No. 4. - S. 266-268). Autologous and allogeneic whole or lysed tumor cells are used as OAA carriers, each tumor cell line having a specific unique set of OAAs, and expanding the spectrum of cell lines allows you to create a wider pool of these antigens that can become available for such antigen-presenting cells? as a DC in the process of in vitro manipulation when creating antitumor vaccines, which allows the most effective induction of antitumor immunity (Monjazeb AM, Hsiao NN, Sckisel GD, Murphy WJ The role of antigen-specific and non-specific immunotherapy in the treatment of cancer // J Immunotoxicol. - 2012. - Jul-Sep; 9 (3) .- P. 248-58. - doi: 10.3109 / 1547691X.2012.685527). DC vaccines have been successfully used to treat patients with kidney cancer, both as monotherapy and as part of combination antitumor therapy (Amin A., Dudek AZ, Logan TF et al. Survival with AGS-003, an autologous dendritic cell-based immunotherapy, in combination with sunitinib in unfavorable risk patients with advanced renal cell carcinoma (RCC): Phase 2 study results // J Immunother Cancer. 2015 Apr 21; 3:14. doi: 10.1186 / s40425-015-0055-3).
The technical result of the invention is the expansion of the arsenal of cell lines used to create whole-cell and genetically engineered antitumor vaccines, which makes it possible to increase the effectiveness of treatment and increase life expectancy in the treatment of malignant neoplasms.
In addition, the obtained new cell line can be used to test the activity of various pharmaceuticals, create diagnostic kits, test systems and experimental models for the development of new drugs and new therapeutic approaches.
