Abstract
Description
The present invention relates to medicine, in particular to new preparations of magnesium based on the natural mineral bischofite, used in balneological practice.
- 272 с.].The bischofite mineral is hexahydrate magnesium chloride of the formula MgCl 2 × 6H 2 O; its deposits were discovered in vast areas of the Caspian Basin and the Volga monocline at depths of 1000-2000 m. bischofite brine. The resulting brine is diluted with water, standardized in density to a value of 1.31-1.33 g / ml and get 33-35% aqueous bischofite solution corresponding to VFS 42-2950-97. The specified bischofite solution is approved by the Ministry of Health of the Russian Federation for medical use as an anti-inflammatory drug [Order of the Ministry of Health of the Russian Federation of 04.23.98 No. 133]. On the basis of a standardized bischofite solution, a number of drugs have been developed, in particular, the Polycatan drug, anti-inflammatory and stimulating regenerative processes in the mucous membranes [VFS 42-2952-97; RF patent No. 2053774 (1996). In addition, the high efficiency of the use of bischofite and medicines based on it in dental practice, dermatology, with inflammatory diseases of the upper respiratory tract, sensorineural hearing loss, hypertension and a number of other diseases [Spasov A.A. Magnesium in medical practice. - Volgograd: Ed. Otrok LLC, 2000. - 272 p.].
А.Б. I Всероссийской конф. However, to date, the main area of medical use of bischofite is balneology, when diluted bischofite solutions in the form of baths and compresses are used to treat diseases of the musculoskeletal system [Bischofite in the treatment of diseases of the joints / Under. ed. A.B. Zborovsky. - Sat thesis. doc. I All-Russian Conf. - Volgograd, 1993, 42 p.].
Based on a bischofite solution purified from technogenic impurities [RF Patent No. 2442593 (2012). Petrov V.I., Spasov A.A., Ozerov A.A., Sysuev B.B. // Method for the purification of bischofite (IPC 7 A61K 33/14, C01F 5/00, C22B 1/00)], a new generation of balneological products - a bischofite solution [TU 9318-004-01896777-2010] have been developed and produced at the Volgograd State Medical University and Polycathan ointment [TU 9318-001-01896777-2010] as therapeutic agents for diseases of the musculoskeletal system, including inflammatory diseases of the joints, peripheral nervous system, muscles and skin.
In addition to an aqueous solution of bischofite in recent years, crystalline bischofite and agents based on it [Pat. RF №2221551 (2004). Ziaztdinova Yu.A., Zhelnina L.N. Balneological treatment and prophylactic agent “Bishol” (IPC 7 A61K 9/08, A61K 33/06, A61K 35/78); Pat. RF №2316333 (2008). Tikhonov S.N., Shishlyakov K.N. Biologically active additive to the product for health-improving cosmetic panto-magnesium baths and a product for health-improving cosmetic panto-magnesium baths based on it (IPC 7 A61K35 / 32, A61K 33/06, A61Q 19/10)]. At present, the production of crystalline bischofite has been mastered at a number of enterprises in the Volgograd region (LLC GRANTREID, CJSC NIKOMAG). Crystalline bischofite is obtained by evaporating at atmospheric pressure water from concentrated bischofite coming from a well and containing, in addition to crystallization, an additional 15-25% water. With this heat treatment in a concentrated bischofite solution, the oxidation of ferrous impurities to ferric impurities (the main inorganic technogenic impurity in the extracted bischofite), hydrolysis of ferric chloride to basic salts and hydroxide, and also partial hydrolysis of magnesium chloride itself are intensified. Moreover, all insoluble impurities present in the initial concentrated bischofite also remain in the final product. When dissolving the crystalline bischofite thus obtained in water, solutions often have a yellow or tan color and high turbidity, which makes their medical use difficult.
The aim of the present invention is to develop a new balneological preparation based on crystalline bischofite, which provides more pure aqueous solutions and has increased convenience of individual use.
Помимо кристаллического бишофита пакет может содержать пищевой зеленый и (или) желтый краситель и ароматизатор для улучшения органолептических свойств получаемого раствора, например масло эфирное мяты, мелиссы, сосны, эвкалипта или пихты [ТУ 9151-001-99535663-07].This goal is achieved by using a metered crystalline bischofite, placed in a perforated bag of water-insoluble polymer material. When such a bag is placed in the water inside it, the fast dissolution of magnesium chloride begins and a highly concentrated solution is formed, the density of which significantly exceeds the density of the water surrounding the package. Due to the indicated difference in density under the influence of gravitational forces, bischofite solution is rapidly naturally filtered through perforations, while all insoluble impurities remain inside the package, which ensures high purity of the resulting balneological solution. The mass of crystalline bischofite placed in the bag is determined by the required value of the final concentration of magnesium chloride for individual balneological use by patients, based on an average bath volume of 150 l. In addition to crystalline bischofite, the package may contain food green and / or yellow coloring and flavoring to improve the organoleptic properties of the resulting solution, for example, essential oil of peppermint, lemon balm, pine, eucalyptus or fir [TU 9151-001-99535663-07].

The use of packaged forms of crystalline bischofite or other inorganic salts for balneological purposes is not known.
The following examples illustrate the invention.
Example 1. A perforated bag with crystalline bischofite net weight of 300 g. In a rectangular bag of size 20 × 20 cm, made of low-pressure polyethylene with a thickness of 4-40 microns, with regularly spaced circular openings on the entire surface with a diameter of 0.3-1.0 mm in increments of 1 cm, placed 300 g of crystalline bischofite [Salt of the "Ancient Sea". Means balneological. TU 9369-004-93524115-2014. CJSC NIKOMAG, Volgograd]. The open portion of the bag is sealed using a PFS-300 Pulse Sealer (PRC). In order to protect from moisture in the air during storage, the resulting bag is placed in a second larger polymer bag without perforation and sealed in a similar manner.
Example 2. A perforated bag with crystalline bischofite net weight of 500 g. Produced in the same way, but using 500 g of crystalline bischofite and a perforated bag measuring 25 × 30 cm
7,4-7,5.Example 3. The study of the dissolution process of bischofite. In a steel cylindrical container with a volume of 200 l, equipped with an ES-8300 steel paddle mixer located on its central axis (ZAO NPO ECROS, Russia), pour 150 l of tap water and, at a temperature of 20-22 ° C and intensive stirring, place perforated a bag containing 300 g or 500 g of crystalline bischofite, so that the bag is located at a distance of 1/2 radius from the inner wall of the tank and is completely immersed in a liquid to a depth of not more than 10-20 cm from its surface. Samples of about 100 ml are taken from the surface layer of the fluid for 10 minutes at intervals of 1 minute. Quantitative determination of magnesium is carried out complexometrically, as described for magnesium sulfate seven-water [State Pharmacopoeia of the USSR. Tenth Edition. - M .: Medicine, 1968. - S. 401-402] in terms of magnesium chloride, six-water: 5 ml of ammonia buffer solution are added to 50 ml of sample and titrated with a 0.05 M solution of Trilon B in the presence of the Eriochrome Black T indicator (Aldrich) before the formation of blue-blue staining; 1 ml of Trilon B solution corresponds to 0.01016 g of magnesium chloride of six-water or 0.00122 g of elemental magnesium. The analysis results are presented in table 1 and figure 1. The pH value of the obtained bischofite aqueous solution is 7.4-7.5.
As follows from the data presented in Figure 1, when using a perforated packet containing 300 g of crystalline bischofite, 90% of it passes into solution after 5.5 minutes from the beginning of the dissolution process (the calculated maximum concentration of bischofite in water is 1.94 g / l taking into account its 97% purity according to TU 9369-004-93524115-2014). When using a perforated bag containing 500 g of crystalline bischofite, the same degree of recovery of 90% is achieved by the 5th minute of the dissolution process, which is probably due to the lower ratio of the mass of the contents of the bag to its surface area (500 g / 25 × 30 × 2 cm 2 = 0.33 g / cm 2 ) compared with a bag containing 300 g of crystalline bischofite (300 g / 20 × 20 × 2 cm 2 = 0.38 g / cm 2 ). The aqueous bischofite solutions obtained in both cases are colorless, transparent, and have no specific odor. The residual contents of the packets are a small amount of a yellow-brown solid, insoluble in water.
Thus, a new convenient balneological preparation based on crystalline bischofite, providing a high purity bischofite solution, has been proposed.
