Abstract
Description
The invention relates to medicine and pharmacology and relates to the creation of funds based on high molecular weight heparin with anticoagulant, antiplatelet, fibrinolytic effect on the body.
Most often in medicine, drugs based on heparin and its derivatives are used as anticoagulant agents.
Heparin is a natural anticoagulant factor and refers to direct-acting anticoagulants, i.e. directly affects blood coagulation factors. Heparin has not only an anticoagulant effect, but to some extent it can activate the fibrinolytic properties of blood and improve coronary blood flow. The introduction of heparin into the body is accompanied by a certain decrease in the content of cholesterol and lipoproteins in the blood serum.
The anticoagulant effect of heparin is manifested invitro and invivo when administered intravenously, intramuscularly and subcutaneously. Heparin acts quickly, but relatively briefly.
Heparin is used in the prevention and treatment of various thromboembolic diseases and their complications, for example, to prevent or limit thrombosis in acute myocardial infarction, thrombosis and embolism of the main veins and arteries, brain vessels and eyes.
В.А. However, it has a number of negative properties, such as thrombocytopenia and immediate allergic reactions (urticaria, angioedema or bronchospasm), with an overdose the possibility of hemorrhage. [Clinical Biochemistry / Ed. V.A. Tkachuk. 2nd ed., Rev. and add. M.: GEOTAR-MED, 2004. 512 p.].
2007.07.10].Known anticoagulant agents of animal origin, representing a fragment of collagen with M.M. about 12 kDa, containing 10.0-12.0% carbohydrates, 6.0-7.5% sulfate groups, micro and macro elements [RF Patent 2302250, A61K 35/56, publ. 2007.07.10].
These agents have a different mechanism of anticoagulant action than the mechanism of action of one VMG, however, the method of obtaining them is complex and long and, in addition, their antiplatelet and anticoagulant effects are insufficient.
27.07.2012).Also known anticoagulant containing heparin and leucine (RF Patent No. 2448717 IPC A61K 31/727, publ. 27.07.2012).
However, this anticoagulant, having a high anticoagulant activity, causes bleeding under certain conditions. In addition, the disadvantage is the need to dilute eikonol with vegetable oil and oral administration.
The basis of the present invention is the task of increasing the effectiveness of the target product by increasing anticoagulant and fibrinolytic activities.
%:The problem is solved in that the anticoagulant containing heparin and leucine, characterized in that it additionally contains arginine, with the following ratios of components, in wt. %:
We have for the first time established that the combined use of leucine, arginine with heparin in the claimed modes makes it possible to unobviously increase the effectiveness of the target product by increasing anticoagulant and fibrinolytic activities. An analysis of the known literature indicates that the claimed technical solution meets the criterion of "novelty."
Comparison of the claimed method with the known shows that they for the specialist do not follow explicitly from the prior art, which indicates that the claimed technical solution meets the criterion of "inventive step".
The claimed technical solution is industrially applicable, because Currently, all components of the claimed solution are produced by domestic industry.
The invention is illustrated by the following examples.

%:Under sterile conditions, to 2.5 g of arginine add 2.0 g of leucine and 95.5 g of heparin with an activity of 120 IU / g, obtaining the composition in the following ratio of components, wt. %:
2.Next, to the resulting composition add sterile saline to 1 kg to obtain a composition of 2.
3.Example 3
%:Under sterile conditions, 1.9 g of leucine and 95.7 g of heparin with an activity of 100 IU / g are added to 2.4 g of arginine with the composition in the following ratio of components, wt. %:
3.Next, to the resulting composition add sterile saline to 1 kg to obtain a composition of 3.
Example 4. The effectiveness of the known anticoagulant (control) and the resulting compositions 1-3 were studied as follows.
табл.1).When a well-known anticoagulant and the obtained compositions 1-3 were added to rat blood plasma, the following results were obtained (see Table 1).
65,5-75%).Thus, the claimed technical solution in comparison with the known one allows to increase anticoagulant activity by 80-83%, fibrin-depolymerization activity - by 80-85% and total fibrinolytic activity - by 65.5-75%).
