Синонимы: Иомезан, Иомесан, Jometan, Kontal, Lintex, Niclosamidum, Radeverm, Teniarene, Vermitin и др.Phenasal (Phenasalum), 5,2'-Dichloro-4'-nitrosalicylanilide or N- (2-chloro-4-nitrophenyl) -3-chlorosalicylamide. Synonyms: Iomezan, Iomezan, Jometan, Kontal, Lintex, Niclosamidum, Radeverm, Teniarene, Vermitin, etc.
FENASAL is a non-toxic substance (class IV), its preparations do not possess embryotropic, carcinogenic properties. FENASAL has anti-cestodic activity, it is a protoplasmic poison, completely denaturing the structure of helminth cells.
The disadvantages of the drug include the laborious synthesis of the intermediate product FENASAL 5-chloro-salicylic acid, for which chlorine gas is used, and the completeness of the reaction is controlled by the weight gain of the reaction mixture.
Previously, the authors synthesized N- (4-methylphenyl) -3,5-dibromosalicylamide, similar in structure to FENASALU.
2006. №1. С.57-59].This compound, codenamed VIVALIN, was tested on farm animals - sheep, cattle (cattle) in the All-Russian Research Institute of Helminthology named after K.I.Skryabin (VIGIS, Moscow) [Mikhaylitsyn F.S., Trusov S.N., Sevbo D.P., Koshevarov N.I., Arkhipov I.A. Assessment of the latitude of the anthelmintic effect of the drug VIVALIN (VIG-08) // Honey. parasitology and parasitar. illnesses. 2006. No1. S.57-59].
As a result, it was found that VIVALIN possesses not only anti-cestodose activity (in case of sheep monysiosis), comparable to FENASAL, but also anti-trematode activity in cattle fascioliasis (83.5% effectiveness), in which FENASAL is not used. VIVALIN (prototype) is the closest in structure and activity to compound 1 (C-1), it is obtained in one step by the interaction of n-toluidine with 3,5-dibromosalicylic acid in toluene in the presence of phosphorus trichloride.
Its disadvantages include the fact that VIVALIN, like FENASAL, does not have a nematodicidal effect against sheep nematododiruses.
The objective of the invention is to expand the spectrum of anthelmintic action, increase activity and expand the arsenal of means of influence on a living organism.

The indicated amount is an amount sufficient to exhibit a noticeable therapeutic, prophylactic or alleviating effect. The specific effective amount for the subject will depend on the body weight, nature and severity of the condition to be treated. For correct comparison with the base drug, the proposed tool was asked at a dose of 100 mg / kg.
Obtaining N- (3,4-dichlorophenyl) -2-acetoxy-3,5-dichlorobenzamide. 7.0 g of N- (3,4-dichlorophenyl) -2-hydroxy-3,5-dibromobenzamide (obtained by the method described in [J.Pharm. Sci. 1961. V.50. P.831-837], are mixed, 20 ml of acetone, 10 ml of acetic anhydride and 5 ml of pyridine and the resulting solution are left for 1 hour at room temperature. At the end of the exposure, a heavy precipitate precipitates from the reaction mass, which is filtered off, after adding 100 ml of water to the reaction mass After drying for 12 hours at 65 ° C, 7.0 g of a solid are obtained which, after crystallization from ethyl acetate, has a melting point of 170-171 ° C. The thus obtained m benzamide (C-1) called mitranox, which is a white to creamy powder, readily soluble in dioxane, moderately in alcohol, toluene, insoluble in water, was directly used to test anthelmintic efficacy (examples 3, 4).
1680 см-1.After crystallization of the indicated sample C-1 from alcohol, an analytical sample is obtained — small plate crystals with a creamy white tint, mp 172-172.5 ° C. TLC on Sorbfil PTSX-P-A-UV plates in the toluene-hexane-ethyl acetate-acetic acid glacial system (4: 1: 0.5: 0.2) is chromatographically homogeneous. PMR - C-1 spectrum is fully consistent with its structure. The IR spectrum in the KBr disk corresponds to formula 1, NH amide 3320 cm -1; ν CO acetyl 1760 cm-1 ;. ν CO amide 1680 cm -1.
2,40; N 3,71.The molecular weight of 377.06. Example 2. The toxicity of the drug was tested on nonlinear white mice with a body weight of 18-22 g. The substance was injected into the stomach using a special probe once in the form of a suspension with starch paste. At least 30 mice were used for each experiment; animals were monitored for 2 weeks from the moment of administration of the substance.
The toxicity of the claimed compounds is comparable with the toxicity of vivalin (class IV), but lower than that of FENASAL.
Evaluation of the anti-cestodic activity of C-1 (Mitranox) was carried out on 28 sheep spontaneously invaded by moniesia (Moniezia expansa and M. benedeni). (Table 1).
According to the results of coproovoscopic studies using the flotation method, animals spontaneously invaded by moniesia were divided into 4 equivalent groups according to the principle of analogues, 7 sheep each.
Овцы четвертой группы препарат не получали и служили контролем (табл.1).The animals of the first group were given mitranox at a dose of 100 mg / kg, and the second, 50 mg / kg. The third group for comparison, as an analogue, received FENASAL at a dose of 100 mg / kg. All preparations were given orally once with water using a rubber bottle, taking into account the weight of the animals. Sheep of the fourth group did not receive the drug and served as a control (Table 1).
The effectiveness of the preparations was taken into account according to the results of quantitative studies of fecal samples of animals of all groups before and 10 days after the administration of the drug to sheep.
As a result, mitranox at a dose of 50 and 100 mg / kg showed respectively 75.8% and 100% effectiveness against moniesia of sheep.
Example 4. Evaluation of the nematodicidal activity of mitranox was carried out on 36 sheep, spontaneously infested with strongilata of the digestive tract, which were divided into 3 experimental and control groups. The animals of the 1st and 2nd groups were given mitranox at a dose of 100 and 50 mg / kg, respectively. Group 3 sheep received phenasal at a dose of 100 mg / kg. All preparations were administered once orally with water from a bottle, taking into account the weight of the animals. Sheep of the 4th group served as a control and did not receive the drug.
В результате митранокс в дозе 100 и 50 мг/кг показал 100 и 95,4%-ную эффективность, а фенасал против кишечных нематод активности не проявил (табл.2).As a result, mitranox at a dose of 100 and 50 mg / kg showed 100 and 95.4% effectiveness, and phenasal did not show activity against intestinal nematodes (Table 2).
Ранее аналогичным образом испытывался препарат вивалин - прототип (сравнение с фенасалом).The calculation of the effectiveness of the preparations was carried out according to the type of “control test”, which shows the percentage reduction in the infection of treated animals compared with animals of the control group (untreated). In accordance with the Guidelines approved by the World Association for the Progress of Veterinary Parasitology [Wood J.B., Amaral N.K., Bairden K., et al. // Vet Parasitol. 1995. V. 58. No. 1/2. P.181-213]. During the experiment, the clinical condition of the animals was also taken into account, which testified to their cure and the absence of toxic effects. The results were processed statistically using the computer program Excel. Previously, the drug vivalin was tested in a similar way - a prototype (comparison with phenasal).
The test results of the inventive mitranox agent for moniesiosis and strongilitis of the intestinal tract of sheep indicate that the drug is highly effective in tested doses against moniesia and nematodes. 10 days after the introduction of mitranox, all animals were completely freed from moniesia, as evidenced by the absence of moniesia eggs in the feces of sheep.
М.benedeni. During helminthological dissection of the small intestine, three selectively killed sheep of the control group found, on average, 2.3 specimens. Moniezia expansa and 1.5 copies. M. benedeni. Contamination of sheep in the control group did not change significantly during the experiment. The number of moniesia eggs on average g / feces of sheep at the beginning of the experiment was 260.4 ± 15.1 specimens. and at the end of the experiment -267.8 ± 16.1 ind. (P> 0.05). The basic drug FENASAL, used in accordance with the instructions at a dose of 100 mg / kg, showed 99.9% efficiency.
Thus, the results of the evaluation of the anthelmintic effect of the claimed mitranox agent show that the drug is not inferior to the drug FENASAL in cestodocidal action, but also has a nematodicidal effect. Mitranox may find application in medicine and veterinary practice.
