Для продления контакта препарата с роговицей и сокращения числа инстилляций в состав капель введено полимерное пролонгирующее вещество - низкомолекулярный декстран с относительной молекулярной массой 30000-40000 дл или сополимер N-винилпирролидона с 2-метил-5-винилпиридином (далее - сополимер) с молекулярной массой 29000-40000 дл. Декстран представляет собой гидрофильный полисахарид, который медленно проникает через сосудистые стенки и при введении в кровеносное русло длительно циркулирует в нем, таким образом повышает эффективность офтальмологического препарата, пролонгируя терапевтическое действие. Сополимер также пролонгирует действие препарата, а также обладает иммуномодулирующим действием. Опыт поставлен на мышах обоего пола линии F1 (CBA×C57B1) весом 16-18 г. Катаракты индуцировали облучением мышей гамма-излучением 137Cs в дозе 4,0 Гр (наиболее адекватная модель старческим катарактам). Через 18 недель после радиационного воздействия одной группе животных - в количестве 92 производили профилактическое закапывание нового лекарственного средства в течение 7 недель два раза в день по 2 капли в оба глаза. Вторая группа животных в количестве 60 служила контролем; третью группу животных в количестве 60 лечили тауфоном (4% раствор 2-аминоэтансульфоновой кислоты).The experiment was performed on mice of both sexes of the F1 line (CBA × C57B1) weighing 16-18 g. В качестве теста повреждения роговицы исследована частота образования аберрантных митозов через 24, 72, 168 и 240 часов после облучения. Новое лекарственное средство закапывали по 2-3 капли в оба глаза 5 раз через каждые 10 мин сразу после облучения. Курс повторяли ежедневно в течение 10 суток. Методика заключалась в следующем: после эпибульбарной анестезии трепаном диаметром 6,0 мм производился надрез передних слоев стромы роговицы, после чего слой этот под уровень кромки надреза удаляется вместе с эпителием. Такая методика позволяла следить не только за скоростью эпителизации, но и за регенерацией стромы роговицы.
