Abstract
Description
The invention relates to the field of biology, ecology and cardiology and can be used to control the parameters of the heartbeat of biological test objects.
119-120).A known method and device for determining the frequency of heartbeats in daphnia, based on the change in light flux and measuring the electrical signal from the photosensor (see Kolupaev B.I., Andreev A.A., Samoilenko Y.K. Optical method for recording heart rhythm in daphnia . - Hydrobiological Journal, vol. XIII, issue 3, 1977, p.119-120).
However, the method cannot unambiguously determine the heart rate, since it is not possible to control the area through which the light flux passes.
2003. №3. С.59-62).A known method of measuring the heart rate of daphnia, in which the radiation of the light source is focused by a lens into the region of the heart of daphnia, placed in a channel on a transparent table, into which the water habitat of daphnia is poured. A microscope is used to direct radiation into the region of the heart of daphnia. Part of the radiation was directed to a photodetector, the signal from which goes through an amplifier and an analog-to-digital converter to a computer, and after recording the oscillograms of Daphnia heart oscillations, the spectrum of the detected signal is calculated using the fast Fourier transform method (see Usanov D.A., Shishkin G.G. , Skripal A.V., Usanov A.D. Effect of low-intensity alternating magnetic fields on the heart rate of daphnia waves // Biomedical Technologies and Radioelectronics. 2003. No. 3. P. 59-62).
The disadvantage of this method is the lack of reliability of registration of fluctuations of the heart, and not other organs of daphnia, as well as the inability to determine the amplitude of the heartbeat of daphnia.
G01N 33/18).There is a method for determining the frequency and amplitude of Daphnia heartbeats from an external auto-detection signal when a part of the radiation reflected from a controlled object is returned to a semiconductor laser resonator, in which, after calculating the spectrum of the autodyne signal, the oscillation period of the Daphnia heart is determined by the frequency of the main signal, and the beat amplitude Daphnia heart (see Usanov D.A., Skripal A.V., Vagarin A.Yu., Skripal A.V., Potapov V.V., Shmakova T.T., Mosiyash S.S. Laser autodyne inter ferrometry of dynamic parameters of bioobjects // Letters to ZhTF. 1998. V.24. Issue 5. P.39-43 and RF patent for invention №2155335, IPC G01N 33/18).
However, this method is very time-consuming and cannot unambiguously determine the parameters of the heartbeats, because it is not possible to control the area through which the light flux passes.
The closest in technical essence to the proposed solution is a method for controlling the frequency of a daphnia heartbeat, in which the radiation of a light source is focused by a lens into the area of a daphnia heart, placed in a channel on a transparent table, into which the water habitat of daphnia is poured. To direct radiation to the heart of Daphnia, a microscope with a video camera is used, the signal from the video camera is transmitted through an analog-to-digital converter to a computer, and the heart rate of Daphnia is calculated using a specially developed video image analysis program that includes visual calculation of the number of periods of movement of the controlled area per unit time (see Lyapina E.P., Lyapin M.N., Golovko E.M., Usanov A.D., Usanov D.A. Daphnia as an experimental model for studying the features of action homeopathically x drugs).
However, this method is very time-consuming and does not allow measurements at a high heart rate.

An example of a practical implementation of the method
As a biological object, freshwater Daphnia crustaceans (Daphnia magna Straus), cultivated under standard laboratory conditions, were used. In the experiments, individuals of 0.7-1.5 mm in size were used.
AVI.A single daphnia from an aquarium culture was placed on a transparent plate and fixed on a microscope stage. After pointing the Daphnia heart region using a Biolam-10 microscope, video was recorded by a Sony camcorder onto a computer hard drive using a Matrox Marvell G400 video capture card that supports full-format video input at 30 frames per second, and the software for capturing video images supplied with it AVI file format.
3.Frame-by-frame processing of the sequence of video frames was carried out; for this, the borders of the heart of Daphnia were identified on each frame. The area limited by the selected boundaries was determined by counting the ratio of the number of points of the selected daphnia heart to the total number of points in the image. The dependence of the area in each frame on time, having a quasiperiodic character, is shown in Fig.3.
The daphnia heart rate was determined by the average value of the period of the indicated dependence and was ~ 6.2 Hz, and the amplitude was determined by the average value of the amplitude of the indicated dependence, which was 0.12 mm. The accuracy of calculating the amplitude and frequency can be improved by increasing the recording time of the image of the heart of daphnia.
Thus, the proposed method allows reliably and with high accuracy to determine the heart rate of daphnia, and also provides the ability to measure the amplitude of the heart rate of daphnia.
