Abstract
Description
The invention relates to methods for the isolation of biologically active substances from plant materials, in particular to methods for the isolation of phytoecdysteroids, namely β-ecdysone ecdysterone). Ecdysterone is an insect molting hormone used in medicine and agriculture.
A known method of producing ecdysteroids from plants of the genus Rhaponticum [Patent of the Russian Federation N 2063763, A 61 K 35/78, 1996], which consists in the extraction of raw materials with 70% ethanol, removal of the accompanying substances from the primary extract by a series of sequential liquid-phase extraction, purification of the resulting product column adsorption chromatography on alumina and its subsequent crystallization. The disadvantages of this method are the use of significant volumes of ethanol in the first stage of extraction, large volumes of organic solvents in subsequent multiple liquid-liquid extractions, multi-stage process time.
Three methods are also known for producing ecdysterone from a plant of the genus Serratula. According to one of them [Kholodova Yu.D., Baltaev U., Volovenko V.O. et al. Phytoecdysteroids Serratula xerathomoides. - "Chemistry of Natural Compounds", 1979, p.171.] Ecdysterone is isolated from the inflorescences of Serratula xerathomoides by grinding air-dried leaves, followed by extraction from them with methanol. The obtained extract was evaporated, diluted with water, extracted repeatedly with hexane, additional distillation of methanol, extraction with butanol, its evaporation, elution with a mixture of chloroform-methanol on an alumina column were carried out. The disadvantage of this method is the low yield of ecdisterone (0.26% of the air-dry weight of the raw material), as well as the multi-stage and long process time (115 hours for ten operations).
According to another method [USSR Author's Certificate N 1146050, A 61 K 35/78, 1985], the primary extraction of the raw material is carried out with methanol, while the vacuum circulation unit is evaporated in the extractor and then the associated substances are removed by liquid extraction and the product is purified by liquid adsorption column chromatography on oxide aluminum. The method includes nine sequential operations: three extractions, three residues, one filtration operation, one dissolution operation and column adsorption chromatography. The implementation time of the method 66 - 70 hours. The disadvantages of this method are the use of liquid-liquid extraction operations, which require multiple repetitions and a minimum of three times the volume of extractant to the extracted volume for each extraction operation, the duration, complexity and multi-stage process, as well as the significant energy consumption of the method due to three operations of evaporation and the duration of the vacuum circulation installation.
According to the method [Patent of the Russian Federation N 2063763, A 61 K 35/78, 1996], adopted as a prototype, to obtain ecdysterone, air-dried leaves of plants of the genus Serratula are crushed and extracted with water at a temperature not exceeding 100 o C for 1 hour. The extract obtained is passed through a layer of a solid hydrophobic sorbent, for example, activated carbon, diasorb, polysorb, while simultaneously separating the accompanying substances and purifying the target product. Then the product is extracted with the sorbent with an organic solvent. The disadvantage of this method is the low yield of the target product (0.49%), the use of large volumes of organic solvents, as well as distilled water. In a typical example of the description of the invention, to obtain 1 g of ecdysterone, 8 l of distilled water, 4 l of 20% ethyl alcohol (which is not reregulated), 8 l of a mixture (1: 1) ethyl acetate-ethanol, 200 g of activated carbon are consumed.
The aim of the invention is to simplify the method while increasing the yield of the product, increasing the productivity of the process, as well as reducing the specific consumption of organic solvents and energy consumption.
The method is as follows.
Dry leaves of plants of the genus Serratula coronata are extracted with water (at a temperature of 45-100 o C), the resulting extract is filtered, and the filtrate (transparent yellow) is concentrated to 0.5 of the original volume by evaporation, freezing of water with ice separation or other method. Extracted from the obtained concentrate with ethyl acetate, the extract was evaporated (at a temperature of 40-65 o C) to 0.25 of the original volume of the extract, cooled and the precipitated crystals of ecdysterone were filtered off.
