В настоящее время для контроля микробной чистоты продукции в пищевой, фармацевтической, косметической промышленности, а также в медицине перспективным является использование методов "быстрой" микробиологии. Одним из таких методов является биолюминесцентный метод определения микробной обсемененности анализируемого образца по количеству внутриклеточного микробного АТФ, отличающийся от традиционного метода подсчета колоний на чашках экспрессностью, простотой исполнения, высокой воспроизводимостью. Метод определения внутриклеточного АТФ основан на способности фермента светляковой люциферазы [1] катализировать реакцию окисления D-люциферина в присутствии соли магния, О2 и АТФ до оксилюциферина с выделением кванта света. Интенсивность свечения при фиксированном содержании D-люциферина и соли магния в реакционной смеси пропорциональна содержанию АТФ в анализируемом образце, т.е. зависит от содержания в нем жизнеспособных клеток микроорганизмов [2]
В работе [3] принятой за аналог, предложен биолюминесцентный способ количественного определения микроорганизмов в жидких образцах (воде, молоке, крови, моче). Согласно данному способу к образцу, исходно содержащему 1000-5000 кл/мл, добавляют смесь 0,1% Тритона Х-100 и апиразы картофеля (0,01-0,1 ед/мл) и выдерживают 5-20 мин, после чего в ту же кювету вносят NRB-реагент (0,05-2% водный раствор смеси этоксилированных аминов и их четвертичных аммониевых солей), или водный раствор кислоты, или водный раствор основания от 1 до 10 объемов.Currently, to control the microbial purity of products in the food, pharmaceutical, cosmetic industries, as well as in medicine, the use of “fast” microbiology methods is promising. In work [3] adopted as an analogue, a bioluminescent method for the quantitative determination of microorganisms in liquid samples (water, milk, blood, urine) was proposed. Для определения концентрации микроорганизмов, согласно данному способу, образец исследуемой ткани гомогенизируют в физиологическом растворе и полученный гомогенат, содержащий исходно 103-105 кл/мл, инкубируют в воздушном термостате при 37оС в течение 5 ч. Гомогенат обрабатывают смесью растворов Тритона Х-100 в концентрации 0,05-0,08% и периодата натрия в концентрации 1-5 мМ, выдерживают 1-3 мин, затем добавляют диметилсульфоксид при объемном соотношении 1: (4-9).
