Abstract

FIELD: biotechnology. SUBSTANCE: strain Corynebacterium tuberculosis ВГНКИ N209R-ДЕП is obtained for accelerated identification of caseous lymphadenitis pathogen at bacteriological, allergic and serological diagnosis of infection in sheep. Molecular properties of strain DNA: spectral data: A 260/280 = 1.9, A 260/230 = 2.3, concentration is 3333 mcg/ml, G + C content = 45.1 mole%. Based on homology nucleotide sequence of DNA this strain is concerned to the group II. For diagnosis allergen prepared from this strain is administrated in subtail wrinkle to sheep by intracutaneous route. Results are recorded after 24 hr. Inflammatory reaction appears in the place of diagnosticum administration in sick animals. This reaction is absent in health animals. Method ensures to detect additionally up to 80% infected animals as compared with conventional way (by palpation of surface lymphatic nodes). EFFECT: improved method of diagnosis.

Description

The invention relates to microbiology, epizootology, and can be used in the practice of bacteriological laboratories for the manufacture of antigens in the reaction of blast transformation of lymphocytes and serological reactions, as well as in biological enterprises for the manufacture of allergen.

The causative agent of caseous lymphadenitis in small cattle has a large variability of phenotypic properties, therefore bacteriological diagnosis of infection is often difficult due to the inability to identify the species.

Bergi's determinant of bacteria (1980) provides data on genotypic properties based on the radiation of strains from foreign collections of microorganisms. This determinant indicates the content of G + C in the DNA of this type of pathogen of infection of 67.5 mol. and pathogenic for humans and animals of the representatives of the genus Corynebacterium 57-60 mol.

To date, in the collections of microorganisms available in Russia, there are no reference strains of C. pseudotuberculosis.

The strain C. pseudotuberculosis was obtained by Zabolotnykh M.V. Kolychev N.M. Trofimov I.G. on 06.10.1987 from the purulent contents of an abscess in a sheep’s liver belonging to the Olginsky state farm of the Poltava region of the Omsk region.

Morphological signs are as follows.

In smears from pure cultures, microorganisms are represented as gram-positive, thick with rounded ends of rods or ovoids 1.07 + 0.04 μm long, 0.649 + 0.02 μm thick.

The physiological signs are as follows.

On blood tellurite agar, the strain restores metallic tellurium, so the colonies become black; dull, brittle, move along the surface of the medium. The maximum size they attain after 2-3 days of incubation at 37 ° C

On MPa, under the same conditions and at the same time, R-form colonies are formed in gray-white. Their edges are even, the top is slightly raised, dry, the surface is dull, rough. The presence of yellow pigment is possible.

On the BCH, a slight turbidity of the medium, an abundant granular precipitate at the bottom of the tube; on the surface of the broth is a continuous film, which, when shaken, breaks into a uniform suspension, a parietal ring. On the third day of cultivation, the maximum amount of exotoxin is formed, which coincides with a decrease in the pH of the medium.

The strain does not reduce nitrates, ferments glucose, lactose, sucrose, maltose and to a small extent sorbitol. Contains catalase, does not contain urease and gelatinase. Intraperitoneal infection of guinea pigs at a dose of 10 MEM / ml causes damage to internal organs, hemorrhagic foci in the lungs, adhesive pleurisy, necrotic nodules in the spleen, abscessed orchitis.

II. In the DNA-DNA hybridization reaction with several tens of Corynebacterium pseudotyberculosis strains, strain 209 R is assigned to group II by the level of homology of the nucleotide DNA sequences.

The strain Corynebacterium pseudotuberculosis was assigned registration number N 209 R-DEPT in the collection of microorganisms of the laboratory for the control and standardization of drugs against tuberculosis, pseudo- and paratuberculosis of VGNKI veterinary drugs.
