Для получения паратуберкулезного аллерго-диагностикума предлагается новый хорошо изученный и охарактеризованный штамм Micobacterium avium subspecies paratuberculosis VIEV, депонированный во Всероссийской государственной коллекции патогенных и вакцинных штаммов микроорганизмов-возбудителей инфекционных болезней животных ФГБНУ ФНЦ ВИЭВ РАН под номером В-1402. 7.1. Проведено полногеномное секвенирование штамма Mycobacterium avium subspecies paratuberculosis VIEV на платформе Illumina NovaSeq с использованием технологии парных прочтений. Биоинформатический анализ включал фильтрацию и тримминг прочтений с использованием программ fastqc (0.11.9) и trimmomatic (0.39). Сборка генома осуществлялась с помощью программы Unicycler (0.4.8). Полученные контиги проверялись на таксономическую принадлежность с помощью программ kraken2 (2.1.2) и blastn (2.5.0). Биоинформатический анализ подтвердил принадлежность вышеупомянутого штамма к виду Mycobacterium avium subspecies paratuberculosis.7.1. Whole-genome sequencing of the Mycobacterium avium subspecies paratuberculosis VIEV strain was carried out on the Illumina NovaSeq platform using paired-read technology. Для получения туберкулопротеинов использовали: штаммы Micobacterium avium subspecies paratuberculosis VIEV и штамм M. avium subspecies paratuberculosis №19698 АТСС, которые параллельно и раздельно культивировали на глицериново-картофельной среде, при температуре 39°С, в течение 30 суток. После окончания периода культивирования указанных микобактерий на глицериново-картофельной среде (вся поверхность картофельного клина покрыта сплошным слоем микобактерий) осуществляли два последовательных пересева колоний на мясо-пептонный глицериново-картофельный бульон (культивирование при температуре 39°С в течение 30 суток).To obtain tuberculoproteins, we used the following strains: Micobacterium avium subspecies paratuberculosis VIEV and M. avium subspecies paratuberculosis strain No. 19698 ATCC, which were cultivated in parallel and separately on a glycerol-potato medium at a temperature of 39°C for 30 days. Второй пересев на мясо-пептонный глицериново-картофельный бульон характеризовался сплошной пленкой, который и использовали для посева на синтетическую среду. Культуры микобактерий штамм Micobacterium avium subspecies paratuberculosis VIEV и штамм M. avium subspecies, paratuberculosis №19698 АТСС характеризовались сплошным ростом на поверхности синтетической питательной среде (Культивирование при температуре 39°С в течение 55 суток в бутылях). После окончания периода культивирования штаммов микобактерий на синтетической среде, проводили стерилизацию посевов при температуре 120°С в течение 30 минут и последующим охлаждением до температуры 18-20°С, в течение 4 часов. Жидкость из бутылей фильтровали через фильтр-полотно получая культуральный фильтрат. Раздельно проводили обработку культурального фильтрата микобактерий Micobacterium avium subspecies paratuberculosis VIEV и культурального фильтрата микобактерий М. avium subspecies paratuberculosis №19698 АТСС, согласно последовательности, представленной ниже. Обработка вариантов культурального фильтрата проводилась 50% раствором трихлоруксусной кислоты при перемешивании на мешалке в течение 5-10 минут, после чего проводили отстаивание в течение 16-18 часов при 4-6°С. В течение указанного периода происходило взаимодействие трихлоруксусной кислоты и протеинов культуральных фильтратов. После этого сливали надосадочную жидкость, а осадок (туберкулопротеин) центрифугировали при 3000 об/мин. в течение 20-25 минут, с последующим удалением надосадочной жидкости. Полученный осадок вариантов туберкулопротеина растворяли дистиллированной водой из расчета 1:5 и подщелачивали 10% раствором нашатырного спирта до рН 7,2-7,4. Растворы туберкулопротеинов подкисляли 10% раствором уксусной кислоты до рН 4,0-5,0 и смешивали с равным количеством насыщенного раствора сернокислого аммония, перемешивали три раза с интервалом 10-15 минут и отстаивали в течение 18 часов при 6°С. После этого проводили центрифугирование при 3000 об/мин. в течение 20-30 минут, надосадок сливали, а осадок растворяли в дистиллированной воде в соотношении 1:5. После этого подщелачивали 10% раствором нашатырного спирта до рН 7,4-7,6 и проводили диализ в проточной водопроводной воде в течение 4 суток, после этого подщелачивают до рН 7,2-7,4 10% раствором аммиака, центрифугировали при 3000 об/мин. в течение 30 минут и фильтровали через стерилизующий фильтр. Таким образом, были получены растворы туберкулопротеинов микобактерий из Micobacterium avium subspecies paratuberculosis VIEV и Micobacterium avium subspecies paratuberculosis штамм №19698 АТСС.The second transfer to meat-peptone glycerin-potato broth was characterized by a continuous film, which was used for sowing on a synthetic medium. The cultural filtrate of mycobacteria Micobacterium avium subspecies paratuberculosis VIEV and the cultural filtrate of mycobacteria M. avium subspecies paratuberculosis No. 19698 ATCC were processed separately, according to the sequence presented below. Treatment of variants of the culture filtrate was carried out with a 50% solution of trichloroacetic acid with stirring on a stirrer for 5-10 minutes, after which settling was carried out for 16-18 hours at 4-6°C. During this period, interaction between trichloroacetic acid and proteins in culture filtrates occurred. After this, the supernatant liquid was discarded, and the sediment (tuberculoprotein) was centrifuged at 3000 rpm. for 20-25 minutes, followed by removal of the supernatant. The resulting sediment of tuberculoprotein variants was dissolved with distilled water at a ratio of 1:5 and alkalinized with a 10% ammonia solution to pH 7.2-7.4. Solutions of tuberculoproteins were acidified with a 10% solution of acetic acid to pH 4.0-5.0 and mixed with an equal amount of saturated ammonium sulfate solution, stirred three times at intervals of 10-15 minutes and left for 18 hours at 6°C. After this, centrifugation was carried out at 3000 rpm. for 20-30 minutes, the supernatant was poured off, and the sediment was dissolved in distilled water in a ratio of 1:5. After this, they were alkalized with a 10% ammonia solution to pH 7.4-7.6 and dialyzed in running tap water for 4 days, after which they were alkalized to pH 7.2-7.4 with a 10% ammonia solution, centrifuged at 3000 vol. /min. for 30 minutes and filtered through a sterilizing filter. Thus, solutions of mycobacterial tuberculoproteins were obtained from Micobacterium avium subspecies paratuberculosis VIEV and Micobacterium avium subspecies paratuberculosis strain No. 19698 ATCC.
В полученных растворах определили концентрацию туберкулопротеинов. Биологическую активность туберкулопротеинов определяли по Международному стандарту ППД для птиц на морских свинках, сенсибилизированных микобактериями птичьего типа.The concentration of tuberculoproteins in the resulting solutions was determined. Полученные растворы туберкулопротеинов разводили растворителем для микобактериальных аллергенов до стандартной рабочей концентрации (2500МЕ) помещали во флаконы, закрывали резиновыми пробками и закатывали алюминиевыми колпачками, и стерилизовали при температуре 120°С в течение 30 минут. Активность микобактериальных аллергенов осуществлялось на морских свинках. Морских свинок весом 350-400 грамм сенсибилизировали культурой М. paratuberculosis в ланолин-вазелиновом адъюванте. Культуру вводили подкожно в область холки и крестца из расчета 0,5 см3 адъюванта, содержащего 5 мг микобактерий.The activity of mycobacterial allergens was carried out on guinea pigs. Определение специфической активности осуществлялось через 35 дней после сенсибилизации. У сенсибилизированных и контрольных свинок боковые поверхности справа и слева депилировали и внутрикожно в объеме 0,1 см3 инъецировали туберкулопротеины, справа полученные из штамма Micobacterium avium subspecies paratuberculosis VIEV, а слева из штамма Micobacterium avium subspecies paratuberculosis №19698 АТСС. Результаты учитывали через 24 часа после введения аллергенов.Determination of specific activity was carried out 35 days after sensitization.
