Abstract
Description
The invention relates to medical microbiology, and in particular to methods for laboratory diagnosis of shigellosis (Boyd dysentery) and can be used in scientific and practical laboratories involved in the indication and identification of bacterial dysentery pathogens.
1984). To identify shigella in the test material, a method is known which consists in the phased isolation of the pure shigella culture from the test material, the study of its properties and final identification by a serological method (agglutination reaction). The final result becomes possible on the 4th day with typical forms and on the 6-7th day with atypical forms of the pathogen (Guidelines for microbiological diagnosis of diseases caused by enterobacteria. Moscow, USSR Ministry of Health, 1984).
The strain Bacteriophagum shigellosum BI is stored in the official depository collection of the Republic of Uzbekistan (Republican collection of microorganisms) at the Research Institute of Epidemiology, Microbiology and Infectious Diseases of the Republican Scientific Center for the Control of Infectious and Parasitic Diseases (NIIEMIZ RSCBI) under the Ministry of Health of the Republic of Uzbekistan following morphological and physiological characters.
It belongs to the fifth morphological group. Head size 75 ± 15 , appendix - 160 ± 40 . The type of nucleic acid is DNA, and its molecular weight is 6000 kdaltons.
Nutrient agar (according to FS 42-188 BC-88). On the Shigella boydii 1 lawn, the size of the negative colonies was 2.0 ± 0.2 mm in diameter. The edge of the colonies is even, the bottom is transparent.
Nutrient broth (according to FS 42-224 BC-88). With a seeding dose of 1 x 10 7 microbial cells per 1 ml of medium and a multiplicity of infection of 1:10, suspension is clarified and nx 10 8 corpuscles per 1 ml of medium accumulate.
A single development cycle is characterized by a minimum latent period of 19 minutes. Productivity - 78 corpuscles per 1 infected cell.
Serological properties. Antiphase serum neutralization rate constant 390 ml / min.
The purpose of the invention is to obtain a strain of a specific bacteriophage for the identification of Shigella boydii 1 bacteria that are released on the territory of Uzbekistan and other regions of the Union, i.e. phage with strict specificity for the indicated group of shigella and suitable for their accelerated laboratory diagnosis.
The proposed strain of Bacteriophagum shigellosum B 1 is new and is not described in the patent literature.
П р и м е р. PRI me R. On a dense nutrient medium (nutrient agar), a strip of fresh (no more than 1 day) broth culture or suspension of microbes in the condensation liquid of a tri-sugar agar column is applied with a bacteriological loop. A drop of this bacteriophage is applied to the culture strip with a Pasteur pipette or bacteriological loop. Sowing incubated for 18 hours at 37 ° C.
If the test culture is Shigella boydii 1, a clear area of specific lysis is observed on a strip at the site of application of the bacteriophage. The presence of a lysis is a diagnostic sign for identifying the culture, which is confirmed by a serological route (agglutination reaction), after which an answer is issued. The use of the diagnostic phage Bacteriophagum shigellosum B 1 can reduce the time of laboratory diagnostics by at least 1 day due to the reduction of the diagnostic process (stage of biochemical research - "motley row"). An economic effect is also achieved through the targeted use of expensive agglutinating serums.
