Рекомбинантная плазмидная ДНК pYP2, кодирующая ферментативно-активную часть полипептида липазы, белка пищеварительного тракта пчелиной огневки длиной в 504 а.к., характеризующаяся следующими признаками:- имеет молекулярную массу 3,212 мегадальтон ( 4,942 т. п. о.);- кодирует ферментативно-активную часть полипептида липазы, белка пищеварительного тракта пчелиной огневки длиной в 504 а.к.;- состоит из:- BamHI/HindIII - фрагмента ДНК плазмиды pQE30 (3,424 т. п. о.)[4];- BamHI/HindIII - фрагмента гена Yp2 Galleria mellonella, кодирующего ферментативно-активную часть полипептида липазы, белка пищеварительного тракта пчелиной огневки (1,551 т. п. о.);- содержит:- в качестве генетического маркера ген bla β-лактамазы, определяющий устойчивость трансформированных плазмидой pYP2 клеток к ампициллину;- нуклеотидную последовательность, кодирующую полигистидиновый тракт в рамке считывания гена Yp2, состоящий из 6 молекул аминокислоты гистидина, для последующей очистки с помощью аффинной хроматографии на сорбенте Ni-NTA-агароза либо аналогов;- уникальные сайты узнавания рестрикционными эндонуклеазами, имеющие следующие координаты:XhoI - 2; EcoRI - 89; BamHI - 148;SmaI - 914; HindIII - 1670; NheI - 1789.Recombinant plasmid DNA pYP2 encoding the enzymatically active part of a lipase polypeptide, a bee moth digestive tract protein of 504 a.k. length, characterized by the following features: - has a molecular weight of 3.212 megadaltons (4.942 bp); - encodes enzymatically the active part of the lipase polypeptide, a protein of the digestive tract of the bee moth, 504 a.k. long; - consists of: - BamHI / HindIII - DNA fragment of plasmid pQE30 (3.424 bp) [4]; - BamHI / HindIII - fragment the Yp2 gene of Galleria mellonella encoding the enzymatic active part of the lipase polypeptide, on the digestive tract of the bee moth (1.551 bp); - contains: - the bla β-lactamase gene, which determines the resistance of ampicillin transformed by plasmid pYP2, as the genetic marker; - the nucleotide sequence encoding the polyhistidine tract in the reading frame of the Yp2 gene , consisting of 6 histidine amino acid molecules, for subsequent purification by affinity chromatography on a sorbent of Ni-NTA agarose or analogues; - unique recognition sites by restriction endonucleases having the following coordinates: XhoI - 2; EcoRI - 89; BamHI - 148; SmaI - 914; HindIII - 1670; NheI - 1789.
