Abstract
1. Способ обнаружения антигенов чумного микроба, включающий нанесение на твердую подложку исследуемого материала в количестве 1-2 мкл, промывание подложки после полного впитывания материала 0,01 М фосфатным буфером pH 7,2, блокирование свободных участков связывания раствором инертного белка, промывание подложки 0,01 М фосфатным буфером, помещение подложки в раствор меченных противочумных IgG и визуальное обнаружение антигенов чумного микроба: при формировании окрашенных пятен определяют наличие в исследуемом материале антигенов чумного микроба, а при отсутствии пятен - отсутствие антигенов чумного микроба, отличающийся тем, что подложку помещают в 0,067 М фосфатный буфер, содержащий 0,14 М NaCl, на 20-30 мин, после чего ее высушивают при комнатной температуре и затем помещают в раствор противочумных IgG (раствор готовят из расчета 500 мкг сухого белка на 1 мл 0,01 М фосфатного буфера) на 30 мин при температуре 37°C, после чего подложку отмывают 0,01 М фосфатным буфером и блокируют свободные участки, при этом в качестве инертного белка для блокирования свободных участков используют 1%-ный раствор казеината натрия, в который помещают подложку на 20-30 мин при температуре 20-25°C, затем наносят исследуемый материал, после чего помещают подложку в раствор меченых противочумных IgG на 40-60 мин при комнатной температуре, а в качестве меченных противочумных IgG используют противочумные IgG, меченные дисперсным красителем, затем подложку извлекают, высушивают при комнатной температуре и визуально определяют наличие или отсутствие антигенов чумного микроба. ! 2.
