Abstract
Description
The invention relates to biotechnology and is a thermophilic strain of unicellular green alga Chlorella vulgaris N, intended for the production and processing of biomass.
A known strain of unicellular green algae Chlorella vulgaris LARG-3, currently used for the same purpose. Temperature optimum growth of this strain was 38 ° C. At the optimum cultivation temperature is 45-50 strain productivity DIA g / m 2 per day. The strain is grown in industrial-type apparatus in Central Asia under sunlight. In the summer months, daytime temperatures are often considerably higher than 38 ° C, therefore, requires significant energy costs for cooling the suspension of algae in the cultivators. If the LARG-3 strain is cultivated without cooling, then its productivity first decreases, and then the biomass growth stops and cell death sets in. Therefore, in the industrial cultivation of this strain in sunlight in Central Asia, significant energy costs are needed to cool and maintain the temperature of cultivation of a suspension of algae in industrial-type apparatuses.
The aim of the invention is a thermophilic strain of unicellular green algae Chlorella vulgaris N, having a higher temperature optimum cultivation of 42 about With, resulting in reduced energy costs for cooling the cell suspension in apparatus for industrial cultivation, while increasing productivity to 52-56 g asv / m 2 per day.
Штамм деионизирован в ВКПМ под N А-29.It proposed a thermophilic strain of the unicellular green algae Chlorella vulgaris N with a temperature optimum of about 42 C. The strain obtained by mutation selection using nitrosomethylurea of wild strain 132. The strain VKPM in deionised under N A-29.
Morphological signs. The cells are round, spherical, with a diameter of 7-9 microns. On an agarized mineral medium, Tamiyya forms dense round colonies of dark green color, the surface of the colonies is smooth, the edge is even. With a natural change of day and night, the duration of the development cycle is within a day. The cell is normally divided into 16, 32 and 64 autospores. The optimal cultivation temperature is 42 ° C superheat Migrates to 45 ° C for several hours. The strain is photo-resistant, grows well under illumination of 250-300 W / m 2 PAR on the beam-receiving surface of the cultivator.
2).Physiological properties. The strain is autotrophic, grows on the mineral environment of Tamiyya, does not require carbon-containing compounds for growth. In semi-industrial installations it grows well on mineral media, for the preparation of which mineral fertilizers are used. As a source of nitrogen, potassium nitrate or urea can be used. The strain is stored in test tubes on a Tamiyya agar medium. Does not lose properties when stored at room temperature or in a refrigerator at a minimum brightness and a temperature not below 5 ° C. The strain highly productive. In a pilot plant in the summer months at 41-42 о С, the productivity is 52-56 g asv / m 2 per day. At the same time the prototype (strain LARG-3) at 38 ° C the productivity is 45-50 g DIA / m 2 per day (Table. 2).
Below are examples of the growth rate of the proposed strain depending on the cultivation conditions.
