Description
The utility model relates to the field of medicine and biotechnology and can be used to select colonies of microorganisms from solid nutrient media for the collection of necessary biomass and further identification using the MALDI-TOF mass spectrometry method.
In connection with the rapid pace of development of modern microbiology and the emergence of new methods for the exact species identification of microorganisms, new methods of sample preparation and selection of colonies of microorganisms for subsequent identification are also required. One of the most significant and promising methods from the point of view of quick and high-quality identification is the MALDI-ToF mass spectrometry method. In this case, the research material is a pure culture of bacteria collected directly from solid nutrient media. In this case, a pure culture can be represented as a single colony, as well as a bacterial mass, if the colonies are small in size.
Identification of most microorganisms is possible without preliminary preparation of material for research, however, there is a fairly wide group of microorganisms for which sample preparation procedures are necessary, due to the peculiarities of their structure. These include microorganisms such as slowly growing non-tuberculous mycobacteria, streptomycetes, actinomycetes and other members of the Actinomycetales family. The microorganisms described above during cultivation on solid nutrient media often give rise in the form of point and small colonies with a diameter of up to 2 mm; therefore, their identification is rather difficult due to insufficient biomass of bacteria necessary for identification. Also, in the process of sample preparation of biomaterial, it is necessary to minimize the ingress of components of nutrient media into the microbial mass, since they reduce the quality of identification.
Standard devices for collecting microbial cultures from culture media are bacteriological loops of various diameters and spatulas of various sizes. The disadvantage of these devices is that they do not allow to collect the biomass volume of microorganisms necessary for identification, giving small point colonies and can capture fragments of dense nutrient media.
The purpose of creating a useful model is to develop a device for collecting point and small colonies of microorganisms from the surface of dense nutrient media, which will allow you to collect the necessary biomass for further identification using the MALDI-ToF mass spectrometry method.
