Abstract
Prezenta inventie se refera la un procedeu de obtinere a calivreinei, care consta în aceea ca supernatantul obtinut prin tratarea pancreasului de porc cu apa ce contine 1,5% fenol, prin macerare la 37 degree C, timp de 72h si adus la pH=4,5 cu acid acetic,se dializeaza în apa timp de 58...72h, se aduce la pH=9...9,5 si apoi se trateaza cu solutie 5% rivanol ce are acelasi pH într-un raport de 70:0,8 apoi precipitatul obtinut se trateaza cu solutie 4% acetat de amoniu cu pH=4,5 într-un raport 2:30, extractul obtinut se purifica prin dializa timp de 24h, în solutie 4,5 % de acetat de amoniu la pH=4,5, iar calicreina se separa din acest dializat prin procedee cunoscute.The present invention relates to a process for obtaining calivreine, which consists in that the supernatant obtained by treating the pork pancreas with water containing 1.5% phenol by maceration at 37 ° C for 72h and brought to pH = 4 , 5 with acetic acid, dialysed in water for 58 ... 72h, adjusted to pH = 9 ... 9.5 and then treated with 5% rivanol solution having the same pH in a ratio of 70: 0.8 then the obtained precipitate was treated with 4% ammonium acetate solution pH = 4.5 in a ratio of 2:30, the obtained extract was purified by dialysis for 24h in 4.5% ammonium acetate solution at pH = 4.5, and the calicrein is separated from this dialysate by known procedures.
