Abstract
Procedeu de obtiere a unei celulaze pure, caracterizat prin aceea ca consta prin folosirea ca materie prima a unor nevertebrate marine care dupa o omogenizare fina urmata de înghet-dezghet se supun la trei extractii succesive cu apa hidromodul 1:3 greutate/volum timp 48, 24, 24h fiecare la temperatura de 2...5 degree C dupa care urmeaza o separare a fazei lichide în care se introduce celuloza 20g/1000 cm3 lichid sau carboximetilceluloza (1:20 greutate/colum) si dupa o pauza de 30 minute la 4 degree C se izoleaza prin centrifugare, celulaza fixata se extrage cu fosfat disodic 0,2 M (100 ml/40 g) la temperatura de 20 degree C în doua etape de 24 h fiecare si cu NaCl 5% 100 ml/40 g, de asemenea în 2 faze de 20 h fiecare în lichidele reunite se adauga sulfat de amoniu 60% saturatie si se separa precipitatul format dupa o sedimentare de o ora la 4 degree C, apoi precipitatul se reia cu apa 1: greutate/volum si se dializeaza 10h fata de un curent de apa distilata, iar pentru activare enzima se trateaza cu o solutie de ZnCl2 în concentratie de 1 niu gZn la 2+ la 27,5 mg proteina.
