Abstract

Przedmiotem zgłoszenia jest biopreparat, sposób jego wytwarzania oraz zastosowanie do stymulowania wzrostu papryki słodkiej (Capsicum annuum L.) w uprawach szklarniowych i pod osłonami. Biopreparat otrzymany jest na bazie szczepu bakterii z gatunku Serratia liquefaciens oznaczonego kodem Dv065 wyizolowanego z ciała stonki kukurydzianej Diabrotica virgifera (LeConte), wytworzony przez formulację szczepu Dv065 i zastosowany do stymulowania wzrostu papryki słodkiej (Capsicum annuum L.), w uprawach pod osłonami. Formulacja obejmuje proces otoczkowania szczepu Dv065 w alginianie sodu zawierającym wyciąg wodny z pancerzyków krewetek. Biopreparat do stymulowania wzrostu roślin zawiera wyselekcjonowany szczep Dv065, należący do gatunku Serratia liquefaciens, pozyskany z ciała owada Diabrotica virgifera, wykazujący określone właściwości biochemiczne, takie jak: wysoka aktywność enzymów fosfatazy, lipazy i proteazy, zdolność do produkcji amoniaku oraz tolerancja na zasolenie do 4% NaCl. Sposób wytworzenia biopreparatu polega na tym, że wyselekcjonowany szczep Dv065, pozyskany z ciała owada Diabrotica virgifera poddaje się formulacji w zmodyfikowanym procesie otoczkowania w alginianie sodu, w następujący sposób: w pierwszym etapie przygotowuje się zawiesinę wodną szczepu Dv065 o koncentracji bakterii w zakresie od 4 x 106 do 4 x 108 jtk/ml; w drugim etapie procedurę otoczkowania w alginianie sodu modyfikuje się poprzez dodanie 0,5 g suchych pancerzy krewetek, uwodnionych w wodzie destylowanej przez 2 godziny, homogenizowanych do uzyskania gładszej formy, a z powstałego homogenatu oddziela się części stałe przez sedymentację, dalej w trzecim etapie przygotowuje się wodny roztwór alginianu sodu w zakresie stężeń 0,5% - 4%, dalej roztwór sterylizuje się, a następnie studzi do temperatury pokojowej, następnie, w czwartym etapie przygotowuje się wodny roztwór chlorku wapnia (CaCl2) w zakresie stężeń 2% - 7%, dalej wysterylizuje się i studzi do temperatury pokojowej, w piątym etapie do roztworu alginianu sodu dodaje się zawiesinę bakterii o koncentracji w zakresie od 4 x 106 do 4 x 108 jtk/ml w proporcji 30:1; gdzie następuje otoczkowanie bakterii, poprzez zamykanie ich w kapsułkach powstałych z usieciowanego alginianu sodu osiągniętego przez wkraplanie roztworu alginianu połączonego zawierającego wyciąg z pancerzyków krewetek, z przygotowaną zawiesiną bakterii, do roztworu chlorku wapnia, a powstałe kapsułki biopreparatu zawierające żywe komórki szczepu Dv065 w otoczce z alginianu sodu i wyciągiem z pancerzyków krewetek, następnie otrzymały biopreparat pozostawia się w roztworze chlorku wapnia w temperaturze 4°C, przez okres do sześciu miesięcy od dnia wytworzenia. Biopreparat zawierający wyselekcjonowany szczep Dv065, pozyskany z ciała owada Diabrotica virgifera do zastosowania do stymulacji wzrostu papryki słodkiej (Capsicum annuum L.).The subject of the application is a biopreparation, a method of its production and its use for stimulating the growth of sweet pepper (Capsicum annuum L.) in greenhouse and covered crops. The biopreparation is obtained on the basis of a strain of bacteria of the Serratia liquefaciens species designated with the code Dv065 isolated from the body of the corn rootworm Diabrotica virgifera (LeConte), produced by the formulation of the Dv065 strain and used for stimulating the growth of sweet pepper (Capsicum annuum L.) in covered crops. The formulation comprises the process of encapsulating the Dv065 strain in sodium alginate containing an aqueous extract of shrimp shells. The biopreparation for stimulating plant growth contains the selected strain Dv065, belonging to the species Serratia liquefaciens, obtained from the body of the Diabrotica virgifera insect, exhibiting specific biochemical properties, such as: high activity of phosphatase, lipase and protease enzymes, the ability to produce ammonia and tolerance to salinity up to 4% NaCl. The method of producing the biopreparation consists in that the selected strain Dv065, obtained from the body of the Diabrotica virgifera insect, is subjected to formulation in a modified process of encapsulation in sodium alginate, as follows: in the first stage, an aqueous suspension of the Dv065 strain is prepared with a bacterial concentration in the range of 4 x 106 to 4 x 108 cfu/ml; in the second stage, the sodium alginate encapsulation procedure is modified by adding 0.5 g of dry shrimp shells, hydrated in distilled water for 2 hours, homogenized to a smoother form, and the solid parts are separated from the resulting homogenate by sedimentation, then in the third stage, an aqueous solution of sodium alginate is prepared in the concentration range of 0.5% - 4%, the solution is then sterilized and then cooled to room temperature, then, in the fourth stage, an aqueous solution of calcium chloride (CaCl2) is prepared in the concentration range of 2% - 7%, then sterilized and cooled to room temperature, in the fifth stage, a suspension of bacteria with a concentration in the range of 4 x 106 to 4 x 108 cfu/ml in a ratio of 30:1 is added to the sodium alginate solution; where the bacteria are encapsulated by enclosing them in capsules made of cross-linked sodium alginate achieved by dripping a combined alginate solution containing an extract of shrimp shells with a prepared suspension of bacteria into a calcium chloride solution, and the resulting capsules of the biopreparation containing live cells of the Dv065 strain in a sodium alginate shell and shrimp shell extract, then received the biopreparation are left in a calcium chloride solution at 4°C for up to six months from the date of production. A biopreparation containing a selected Dv065 strain, obtained from the body of the Diabrotica virgifera insect, to be used to stimulate the growth of sweet pepper (Capsicum annuum L.).
