Abstract

Przedmiotem zgłoszenia jest sposób otrzymywania matrycy kolagenowej z kości wołowych, w którym kości wołowe oczyszcza się, demineralizuje, sterylizuje i liofilizuje, który charakteryzuje się tym, że obejmuje następujące etapy: a) kości wołowe oczyszcza się z tkanki miękkiej i tnie na kawałki o objętości 0,25 - 16 cm<sup>3</sup>, b) surowiec przygotowany w etapie a) myje się wodą odsącza i osusza, c) osuszony surowiec poddaje się procesowi usunięcia tłuszczy w procesie ekstrakcji CO<sub>2</sub> fazie nadkrytycznej, d) materiał uzyskany w etapie c) poddaje się demineralizacji w roztworze kwasu solnego o stężeniu 0,05 - 1 mol/dm<sup>3</sup> przez okres 24 godzin, e) materiał pozbawiony składników mineralnych uzyskany w etapie d) w postaci surowej matrycy kolagenowej myje się wodą dejonizowaną, f) surową matrycę kolagenową poddaje się dezynfekcji w roztworze H<sub>2</sub>O<sub>2</sub> przez okres od 1do 4 godzin, przy stosunku objętościowym ilości materiału do wody 1:50, g) zdezynfekowaną matrycę zamraża się do temperatury nie niższej niż – 40°C, h) zamrożony materiał poddaje się liofilizacji. Otrzymaną matrycę kolagenową można umieścić się w roztworze substancji wzbogacającej o stężeniu od 0,5 do 5% v/w. Według sposobu otrzymuje się matrycę kolagenową w formie porowatych bloczków o średnicy porów 50 - 500 mikrometra. Matryce kolagenową stosuje się do regeneracji ubytków kostnych oraz tkanek miękkich przyzębia.The subject of the application is a method for obtaining a collagen matrix from beef bones, in which the beef bones are cleaned, demineralized, sterilized and freeze-dried, which is characterized by the following steps: a) beef bones are cleaned of soft tissue and cut into pieces with a volume of ,25 - 16 cm<sup>3</sup>, b) the raw material prepared in stage a) is washed with water, drained and dried, c) the dried raw material is subjected to the process of fat removal in the CO<sub>2</sub> extraction process supercritical phase, d) the material obtained in stage c) is subjected to demineralization in a hydrochloric acid solution with a concentration of 0.05 - 1 mol/dm<sup>3</sup> for a period of 24 hours, e) material devoid of mineral components obtained in stage d) in the form of a raw collagen matrix, it is washed with deionized water, f) the raw collagen matrix is disinfected in a H<sub>2</sub>O<sub>2</sub> solution for a period of 1 to 4 hours, at a volume ratio amount of material to water 1:50, g) the disinfected matrix is frozen to a temperature not lower than - 40°C, h) the frozen material is freeze-dried. The obtained collagen matrix can be placed in a solution of an enriching substance with a concentration of 0.5 to 5% v/w. According to the method, a collagen matrix is obtained in the form of porous blocks with a pore diameter of 50 - 500 micrometers. The collagen matrix is used to regenerate bone defects and periodontal soft tissues.
