Abstract
Przedmiotem wynalazku jest sposób otrzymywania nanocząstek lipidowych, syntetyzowanych na bazie oleju z mikroglonów morskich (Schizochytrium) oraz lipidów pozyskiwanych z okrzemek (Halamphora), charakteryzujących się wielkością cząstek w zakresie 100 – 300 nm. Sposób otrzymywania nanocząstek lipidowych syntetyzowanych na bazie oleju z mikroglonów morskich (Schizochytrium) oraz lipidów pozyskiwanych z okrzemek (Halamphora) metodą HSH polega na tym, że ogrzewa się mieszaninę zawierającą stearynian glicerolu (Imwitor 900K) w ilości 1 – 5 g korzystnie 2,5 g, bromek cetylotrimetyloamoniowy (CTAB) w ilości 0,1 – 0,8 g korzystnie 0,4 g, lipidy wyekstrahowane wcześniej z okrzemek Halamphora w ilości 0,2 – 1 g korzystnie 0,6 g, olej z mikroglonów morskich (Schizochytrium) w ilości 0,4 – 1,2 g korzystnie 0,8 g i glicerol w ilości 5 – 15 g korzystnie 10 g, do temperatury powyżej 75°C przy ciągłym mieszaniu z prędkością 400 – 600 rpm korzystnie 600 rpm do uzyskania jednolitej konsystencji, następnie mieszaninę poddaje się prehomogenizacji przy prędkości 8 000 – 24 000 rpm korzystnie 13 500 rpm przez 5 – 20 sekund korzystnie 10 sekund, dalej do zhomogenizowanej próbki dodaje się 1 ml roztworu wodnego Tween 80 podgrzanego do temperatury 40°C o stężeniu 10 – 60% wag., korzystnie 30% wag. i miesza się z prędkością 400 – 600 rpm korzystnie 600 rpm przez 0,5 – 2 minut korzystnie przez 1 minutę do uzyskania jednolitej konsystencji, dalej próbkę poddaje się dwuetapowej homogenizacji właściwej: w pierwszym etapie przy prędkości 13 500 – 24 000 rpm korzystnie 20 500 rpm przez 5 – 15 sekund korzystnie 10 sekund; w drugim etapie przy prędkości 8 000 – 13 500 rpm korzystnie 9 500 rpm przez 5 – 15 sekund korzystnie 10 sekund, po czym tak powstałą dyspersję dodaje się do 14 – 14,9 ml korzystnie 14,3 ml roztworu wodnego Tween 80 o stężeniu 10 – 60% wag. korzystnie 30% wag. przy ciągłym mieszaniu z prędkością 350 – 550 rpm, korzystnie 350 rpm i miesza do momentu ochłodzenia układu do temperatury pokojowej korzystnie 25°C i do otrzymania nanocząstek lipidowych typu NLC, charakteryzujących się wielkością cząstek w zakresie 100 – 300 nm.
