Description

Opis wynalazkuDescription of the invention

Wynalazek dotyczy zastosowania nanocząstek srebra jako środka fungistatycznego do ochrony roślin bobowatych (Fabaceae), szczególnie grochu siewnego (Pisum sativum L.) przed Didymella pinodes. The invention relates to the use of silver nanoparticles as a fungistatic agent for protecting legumes (Fabaceae), especially field peas (Pisum sativum L.) against Didymella pinodes.

Jednak częste stosowanie tych samych fungicydów może prowadzić do wykształcania odporności przez D. pinodes, co zostało zaobserwowane w Australii [2], a same fungicydy i ich pozostałości stanowią zagrożenie dla środowiska i zdrowia zwierząt i ludzi [3].Legume plants, especially field peas, are attacked by the pathogenic fungus Didymella pinodes, which causes ascochyta blight (pea blight), one of the most serious fungal diseases of peas. Symptoms of this disease appear initially on leaves and stems, then on pods and seeds, leading to underdevelopment, discoloration, and shriveling [1]. The pathogen can survive for many years on heavily infected seed material, as well as in the soil (on crop residues). The Mycosphaerella pinodes fungus, in its perfect stage, Didymella pinodes, causes pea yield reductions of over 30%, and in exceptional cases, even 100%. It was shown that infection of plants within 10-12 nodes resulted in a 20-25% reduction in seed number and a 14-17% increase in the proportion of small seeds, compared to the yield from healthy plants. Pea cultivars and breeding lines are not resistant to this disease, and the search for more resistant genotypes has not yet resulted in the development of varieties fully resistant to ascochyta blight. Therefore, current pea protection against this disease relies on appropriate agricultural practices, including delaying sowing when climatic conditions permit, maintaining appropriate crop rotation, and using a range of fungicides. However, frequent use of the same fungicides can lead to the development of resistance in D. pinodes, as observed in Australia [2], and the fungicides themselves and their residues pose a threat to the environment and to animal and human health [3].

W ostatnich latach coraz szersze zastosowanie w zwalczaniu chorobotwórczych bakterii i grzybów znajdują nanocząstki metali/tlenków metali, w tym nanocząstki srebra (ang. silver nanoparticles, AgNPs), pierwiastka znanego z antyseptycznych właściwości [4]. Istotny wpływ na toksyczność AgNPs wobec bakterii i grzybów ma też metoda ich syntezy [5, 6].In recent years, metal/metal oxide nanoparticles, including silver nanoparticles (AgNPs), an element known for its antiseptic properties, have been increasingly used to combat pathogenic bacteria and fungi [4]. The bactericidal and fungicidal effects of AgNPs include: disintegration of the microorganism's cell wall, destruction of the integrity and selectivity of the cell membrane, disruption of mitochondrial respiration, overproduction of reactive oxygen species (ROS), lipid peroxidation, DNA damage, transcription, translation, and disruption of protein synthesis (including enzymatic proteins) and cell wall components. The effectiveness of AgNPs against microorganisms depends on the physicochemical properties of the nanoparticles (size, shape, surface-to-volume ratio, type of coating/shielding substances, stability/aggregation) and their concentration, as well as the properties of the microorganisms they target. The method of their synthesis also has a significant impact on the toxicity of AgNPs towards bacteria and fungi [5, 6].

Istotną zaletą tak wytwarzanych AgNPs jest możliwość uzyskiwania nanocząstek o wysokiej skuteczności w zwalczaniu patogennych mikroorganizmów, a jednocześnie mniej toksycznych od nanocząstek wytwarzanych chemicznie [7-9].The desire to reduce the use of chemicals in agriculture and energy inputs is driving increased interest in producing AgNPs as potential nanopesticides obtained through "green chemistry" methods, including plant extracts and metabolites produced by microorganisms. Biological synthesis of AgNPs is considered rapid, simple, and environmentally friendly due to the lower consumption of chemicals (including toxic ones) and energy inputs compared to classical chemical and physical methods. A significant advantage of AgNPs produced in this way is the ability to obtain nanoparticles that are highly effective in combating pathogenic microorganisms, while also being less toxic than chemically produced nanoparticles [7-9].

In recent years, the possibilities of using various species of lactic acid bacteria (LAB: Lactobacillus acidophilus, L. casei, L. bulgaricus, L. plantarum, L. lactis, L. ruminis, L. paracasei, L. bravis) present, among others, in dairy products, for the synthesis of AgNPs have been investigated, which is in line with the idea of sustainable agriculture [10]. LAB metabolic products secreted outside the cells [11, 12], as well as exo-polysaccharides of the bacterial capsid, are both factors reducing silver ions Ag+ to Ag0 and stabilizing the resulting silver nanoparticles [13]. The obtained AgNPs are small (5-40 nm), spherical and exhibit strong antibacterial activity against pathogenic gram-positive bacteria, e.g. Staphylococcus aureus, Listeria monocytogenes [14] or gram-negative bacteria, e.g. Klebsiella pneumoniae [11], as well as both types of bacteria [12, 15, 16]. The bactericidal effect depends on the species/strain of LAB producing AgNPs, the concentration of the nanoparticles themselves and the species of bacteria they affect.

Niektóre AgNPs, wytwarzane za pomocą LAB, wykazują (w stężeniu od kilku do kilkudziesięciu mg/dcm3) działanie fungistatyczne wobec Pythium aphanidermatum, Fusarium oxysporum, Phytophthora parasitica [14]; Aspergillus flavus, Aspergillus ochraceus, Penicillum expansum [17]; Candidia albicans [10]; Saccharomyces cerevisiae [15]. Działanie takie w w/w pracach analizowano w badaniach in vitro w testach dołkowych na szalkach z pożywką glukozowo-ziemniaczaną (PDA) i kulturami zawiesinowymi patogennego grzyba. Some AgNPs produced by LAB exhibit (at concentrations ranging from several to several dozen mg/ dcm3 ) fungistatic activity against Pythium aphanidermatum, Fusarium oxysporum, Phytophthora parasitica [14]; Aspergillus flavus, Aspergillus ochraceus, Penicillum expansum [17]; Candidia albicans [10]; Saccharomyces cerevisiae [15]. Such activity was analyzed in the above-mentioned works in in vitro tests in wells on plates with glucose-potato broth (PDA) and suspension cultures of the pathogenic fungus.

Jak dotąd brakuje informacji i danych naukowych, zarówno z badań in vitro jak i z in vivo, dotyczących aktywności fungistatycznej/grzybobójczej AgNPs wytwarzanych za pomocą LAB wobec D. pinodes. So far, there is a lack of information and scientific data, both from in vitro and in vivo studies, regarding the fungistatic/fungicidal activity of AgNPs produced by LAB against D. pinodes.

Przedmiotem wynalazku jest zastosowanie nanocząstek srebra (AgNPs) pozyskanych za pomocą bakterii kwasu mlekowego do wytwarzania środka fungistatycznego do ochrony roślin bobowatych (Fabaceae), szczególnie grochu siewnego (Pisum sativum L.) przed Didymella pinodes. The subject of the invention is the use of silver nanoparticles (AgNPs) obtained with the help of lactic acid bacteria for the production of a fungistatic agent for the protection of legumes (Fabaceae), especially field pea (Pisum sativum L.) against Didymella pinodes.

The use according to the invention consists in using silver nanoparticles (AgNPs) obtained with the help of lactic acid bacteria to produce a preparation for protecting legumes (Fabaceae), especially field peas (Pisum sativum L.) against Didymella pinodes in the form of an aqueous suspension.

The method according to the invention involves pre-sowing seed dressing with an AgNPs suspension, which involves briefly soaking the seeds in the AgNPs suspension and then drying them. Advantages of using AgNPs according to the invention include: short seed dressing period, low concentration of AgNPs used (up to 100 ppm), high fungistatic efficacy against D. pinodes, protection of peas against D. pinodes infection carried with the seed, and no negative impact on plant yield parameters. Additionally, seed dressing with the fungistatic agent according to the invention can be performed several months before sowing without affecting seed viability and vigor. However, in this case, extending the seed soaking time (up to 90 minutes) or increasing the AgNPs concentration (for 30-60 minutes) to 150 mg/ dcm3 is inadvisable, as these changes result in decreased seedling vigor.

Przykład 1Example 1

Wytwarzanie nanocząstek srebra za pomocą bakterii kwasu mlekowego Lactobacillus curvatus MEVP1 [OM736187], wyizolowanych z serwatki wg metody opisanej wcześniej [18], przeprowadzono zgodnie z międzynarodowym zgłoszeniem patentowym nr EP22460029 z dnia 30.06.2022 i opisem w publikacji Railean i in. [19]. The production of silver nanoparticles using the lactic acid bacteria Lactobacillus curvatus MEVP1 [OM736187], isolated from whey according to the method described earlier [18], was carried out in accordance with international patent application No. EP22460029 dated June 30, 2022 and the description in the publication by Railean et al. [19]. The entire disclosure of this invention should be considered as a reference to the present application and is incorporated herein by reference. The bacteria were cultured in MRS medium in a Biostat A bioreactor (Sartorius Stedim Biotech, Germany) at 26°C for 48 hours, after which the bacterial cells were eliminated by centrifugation and the post-culture medium was combined with AgNO3 (to 5 mM final concentration) for 1 minute and centrifuged, separating the supernatant (with silver nanoparticles) from the pellet (containing silver chloride nanoparticles). The supernatant was incubated in the dark at 40°C for 36 hours to complete AgNP synthesis. After centrifugation, the pellets were repeatedly purified to remove residual silver nitrate and low-molecular-weight metabolites by washing the pellet with water and centrifugation, and finally, by five days of dialysis (3 kDa cutoff, Spectrumlab, USA). The obtained silver nanoparticles (bio-AgNPs) were characterized by a crystalline structure, the presence of only silver (no chlorine), a spherical shape, a diameter in the range of 5-30 nm (24.06 ± 8.43 nm), the presence of organic compounds on the surface, and resistance to agglomeration (for 3-7 days).

Aktywność fungi statyczną AgNPs wobec D. pinodes określono na podstawie dwóch testów. Zawiesinę AgNPs przygotowano poprzez dyspersję nanocząstek srebra w wodzie redestylowanej i poddanie jej działaniu ultradźwięków (310 W, w wodnej łaźni ultradźwiękowej, POLSONIC Pałczyński Sp. J. Warszawa, Polska) przez 30 minut bezpośrednio przed użyciem. The fungistatic activity of AgNPs against D. pinodes was determined based on two tests. The AgNPs suspension was prepared by dispersing silver nanoparticles in redistilled water and subjecting it to ultrasonication (310 W, in an ultrasonic water bath, POLSONIC Pałczyński Sp. J. Warsaw, Poland) for 30 minutes immediately before use.

W pierwszym z testów - metodzie zatrutych podłoży, wykorzystano jednozarodnikowe kultury grzyba D. pinodes (szczep 165/T; NCBI: NC_029396.1) pochodzące z kolekcji Katedry Entomologii, Fitopatologii i Diagnostyki Molekularnej UWM w Olsztynie. Pobrane 6-cio milimetrowe krążki grzybni D. pinodes hodowano na pożywce PDA z dodatkiem przygotowanej zawiesiny AgNPs (do końcowego stężenia 10, 100, 150 i 200 mg/dcm3). W celu weryfikacji efektywności nanocząstek w ograniczeniu wzrostu badanego gatunku grzyba zastosowano fungicyd Amistar 250 S.C. (substancja czynna: azoksystrobina). 1A.In the first test, the poisoned media method, single-spore cultures of the D. pinodes fungus (strain 165/T; NCBI: NC_029396.1) from the collection of the Department of Entomology, Phytopathology and Molecular Diagnostics, University of Warmia and Mazury in Olsztyn were used. Six-millimeter mycelial discs of D. pinodes were cultured on PDA medium with the addition of a prepared AgNP suspension (to final concentrations of 10, 100, 150, and 200 mg/ dcm3 ). To verify the effectiveness of the nanoparticles in limiting the growth of the tested fungal species, the fungicide Amistar 250 SC (active substance: azoxystrobin) was used. The experiment was conducted for 14 days at 22°C, with access to daylight. After 7 and 14 days, mycelium growth was measured in two perpendicular directions, and the degree of growth inhibition was determined. It was shown that AgNPs suspension at concentrations of 100, 150, and 200 mg/ dcm3 reduced D. pinodes mycelium growth by approximately 50%. Increasing the AgNPs concentration from 10 to 100 mg/ dcm3 increased the percentage of mycelium growth inhibition three-fold, while further increases in the nanoparticle concentration (to 150-200 mg/ dcm3 ) had no significant effect, as shown in Fig. 1A.

1B.In the second test, to determine the minimum inhibitory concentration (MIC) of the fungistatic agent produced according to the invention against D. pinodes, a survival test was performed with resazurin dye (whose color changes in the presence of living cells are a consequence of their respiratory activity) in multi-well plates. AgNPs were added to wells containing 100 μL of liquid potato glucose broth (PDB) to obtain concentrations of 500, 250, 125, 62.5, 31.25, and 15.62 mg/ dcm3 . 10 μL of D. pinodes spores were added to the wells at a concentration of 2 x 106 spores/ cm3 (density determined using a Brucker hemocytometer). Resazurin dye (Tokio Chemical Industry, Japan, CAS RN: 62758-13-8, cat. no. R0203) was dissolved (10 mg) in 3 mL of sterile water, and 10 μL of the prepared solution was added to each well. Incubation was carried out for 4 days at 22°C (day/night, under laboratory conditions). The MIC value was shown to be 62.5 mg/ dcm3, as shown in Fig. 1B.

The use of AgNPs suspension to protect peas against D. pinodes consisted in soaking the seeds of the general-purpose narrow-leafed pea variety 'Nemo' for 30-60 minutes in AgNPs suspensions prepared at a concentration of 50 and 100 mg/ dcm3 (below and above the MIC value shown in Example 1), added to the seeds in a volume-to-weight ratio of 1:1. Then the suspension was removed and the seeds (without rinsing with water) were left to dry at room temperature with natural air circulation and access to daylight for 48 hours.

2.The effect of the invention on pea resistance to D. pinodes was analyzed in an artificial infection test on seedlings and during plant vegetation in pot culture (greenhouse experiment). In the first case, control seeds - untreated and treated in seed dressing (MAXIM 025 FS, Syngenta) with fludioxonil as the active substance, and treated with AgNPs suspensions - were germinated in moist filter paper rolls (Eurochem BDG, Tarnów, Poland) for 5 days (in the dark in an ILW 115-T STD chamber, Pol-Eko-Aparatura), after which properly developed seedlings were placed in glass tubes with 10 mL of redistilled water so that only the root was immersed in the water. After two days, the epicotyl was coated with spore-forming mycelium of D. pinodes (spore concentration: 1 x 107 CFU), the seedlings were covered with a transparent foil for 24 hours (to ensure adequate humidity for the developing fungi), and 14 days after infection, the degree of seedling infection, their length, and fresh and dry weight were determined. It was shown that seedlings grown from seeds treated with AgNPs exhibited high resistance to D. pinodes – higher than seedlings from seeds treated with fungicide, as shown in Fig. 2.

In the second experiment, dry seeds soaked in a suspension of AgNPs at a concentration of 50 mg/ dcm3 for 1 hour and seeds treated in MAXIM 025 FS (with fludioxonil as the active ingredient) were sown into pots (6 seeds per pot) filled with 10 liters of a mixture of garden soil and calcined sand in a 2:1 volume ratio, to a depth of 1 cm. Each seed sown into the pot was dosed with 5 mL of D. pinodes mycelium with spores at a concentration of 1 x 107 CFU and covered with soil. Fifty days after infection, plant infection was assessed (including using qPCR). The plants were grown to full maturity (140 days from sowing), after which morphometric analyses were performed, taking into account the following yield structure elements: plant height, height of first pod deposition, number of pods per plant, number of seeds per pod, number of seeds per plant, 1000-seed weight.

Najwyższy poziom infekcji (25,3%) stwierdzono w wariancie kontrolnym, w którym gleba została potraktowana zarodnikami D. pinodes, podczas gdy zaprawianie AgNPs znacząco obniżyło porażenie roślin (indeks porażenia 8,1 ± 3,4%) w stopniu zbliżonym do zaobserwowanego na roślinach wyrosłych z nasion traktowanych zaprawą nasienną (5,3 ± 1,2%). Powyższe wyniki potwierdzono metodą qPCR (fig. 3) - w kontrolnych roślinach infekowanych wykazano obecność 707,5-882,5 pg DNA D. pinodes (w obu seriach badań), podczas gdy w roślinach z nasion zaprawionych AgNPs poziom patogennego DNA był wielokrotnie niższy (96-107 pg DNA).The highest level of infection (25.3%) was found in the control variant in which the soil was treated with D. pinodes spores, while AgNPs treatment significantly reduced plant infection (infection index 8.1 ± 3.4%) to a degree similar to that observed in plants grown from seeds treated with seed dressing (5.3 ± 1.2%). The above results were confirmed by qPCR (fig. 3) - in control infected plants the presence of 707.5-882.5 pg of D. pinodes DNA was shown (in both series of tests), while in plants from seeds treated with AgNPs the level of pathogenic DNA was many times lower (96-107 pg DNA).

Pomiary wysokości dojrzałych roślin (w momencie zbioru), osadzania pierwszego strąka, liczby strąków z rośliny, liczby nasion w strąku i masy 1000 nasion nie wykazały statystycznie istotnych różnic pomiędzy roślinami kontrolnymi a rozwijającymi się z nasion zaprawianych zaprawą nasienną i środkiem fungistatycznym wg wynalazku (wodna zawiesiną AgNPs w stężeniu 50 mg/dcm3).Measurements of the height of mature plants (at the time of harvest), the deposition of the first pod, the number of pods per plant, the number of seeds per pod and the weight of 1000 seeds did not show any statistically significant differences between the control plants and the plants developing from seeds treated with the seed dressing and the fungistatic agent according to the invention (aqueous suspension of AgNPs at a concentration of 50 mg/ dcm3 ).
