Abstract
Ujawniono biosensor do oznaczania enzymu metaloproteinazy-1, z wykorzystaniem króliczego, specyficznego przeciwciała przeciwko ludzkiej metaloproteinazie-1. Biosensor składa się ze szkiełka (1) oraz czterech warstw, na które nałożona jest siatka polimeru (6) dzieląca sensor na partycje. Spodnią warstwę biosensoru stanowi chrom (3), na który nałożone jest złoto (2), następnie tiolowy linker (4) oraz królicze, specyficzne, zimmobilizowane przeciwciało przeciwko ludzkiej metaloproteinazie-1 (5).

Opis wynalazku
Przedmiotem wynalazku jest biosensor do oznaczania enzymu metaloproteinazy-1 (MMP-1).
Metaloproteinazy macierzy zewnątrzkomórkowej (ang. matrix metalloproteinases, MMPs) jest to grupa zależnych od cynku enzymów proteolitycznych, należących do endopeptydaz, których podstawową funkcją jest uczestniczenie w fizjologicznych i patologicznych procesach przebudowy i degradowania składników macierzy zewnątrzkomórkowej (ang. extracellular matrix, ECM): kolagenu, laminy, proteoglikanów, czy fibronektyny, co przekłada się na ułatwienie migracji komórek. Jednak ECM nie jest tylko biernym rusztowaniem tkanki, ale jest także rezerwuarem licznych czynników wzrostu, które uwolnione przez MMP mogą wpływać na komórki. Poza ECM substratami MMP są różnego rodzaju molekuły znajdujących się na powierzchni komórek. Mogą one uczestniczyć w procesie formułowania receptorów komórkowych oraz w procesach immunologicznych. Dlatego funkcji MMP nie można upraszczać tylko do prostej degradacji ECM. W swoim składzie zawierają atom cynku (Zn2+), który pełni rolę katalityczną i strukturalną w cząsteczce enzymu.
MMP są syntetyzowane w postaci nieaktywnych zymogenów (proMMP). Aktywacja MMP odbywa się przez usunięcie prodomeny, w wyniku czego następuje odsłonięcie miejsca aktywnego enzymu. MMP mogą być aktywowane przez inne proteazy lub czynnikami chemicznymi. Także niskie pH oraz podwyższona temperatura prowadzą do aktywacji MMP. Aktywność metaloproteaz jest precyzyjnie regulowana na poziomie transkrypcji, translacji oraz poprzez endogenne inhibitory, takie jak a2-makroglobulina i tkankowe inhibitory metaloproteinaz (TIMP). Istotą wynalazku jest biosensor umożliwiający specyficzne oznaczanie metaloproteinazy-1 techniką powierzchniowego rezonansu plazmonów w wersji Imaging. Biosensor składa się ze szkiełka, korzystnie typu BK7 oraz czterech warstw, na które nałożona jest siatka polimeru dzieląca sensor na partycje. Spodnią warstwę biosensora stanowi chrom, na który nałożone jest złoto, korzystnie o grubości 50 nm, a następnie tiolowy linker, korzystnie cysteamina. Warstwę wierzchnią biosensora stanowi królicze, specyficzne, zimmobilizowane przeciwciało przeciwko ludzkiej metaloproteinazie-1.
Dzięki zastosowaniu rozwiązania według wynalazku uzyskuje się bardzo szybką analizę badanego materiału, w tym: osocza, moczu, śliny, homogenatów tkanek na stężenie metaloproteinazy-1.
Przedmiot wynalazku uwidoczniony jest na rysunku, na którym Fig. 1 przedstawia przekrój pionowy przez biosensor, a Fig. 2 przedstawia zakres prostoliniowy krzywej kalibracyjnej enzymu MMP-1. Biosensor składa się ze szkiełka (1) oraz czterech warstw. Spodnią warstwę biosensora stanowi chrom (3), na który nałożone jest złoto (2) korzystnie o grubości 50 nm, następnie tiolowy linker (4) oraz królicze, specyficzne, zimmobilizowane przeciwciało przeciwko ludzkiej metaloproteinazie-1 (5). Sensor na partycje dzieli siatka polimeru (6).
Sensor działa współpracując z urządzeniem do pomiarów techniką powierzchniowego rezonansu plazmonów w wersji Imaging (SPRI). Sensor kładzie się na pryzmacie urządzenia. Odpowiednie przygotowanie sensora polega na zimmobilizowaniu na warstwie złota (2), monowarstwy tiolu (4) np. cysteaminy, a na niej utworzenie monowarstwy króliczego, specyficznego przeciwciała przeciwko ludzkiej MMP-1 (5) w zakresie stężeń 50- 300 ng/ml- z plateau otrzymanej krzywej odczytuje się stężenie tego
PL 235 043 B1 przeciwciała przy którym następuje maksymalne wysycenie powierzchni biosensora. Na poszczególne pola sensora zawierające dużą liczbę miejsc aktywnych np. 12, nakłada się wzorzec MMP-1 tak, aby uzyskać stężenia 0,05; 0,1; 0,25; 0,5; 0,75; 1,0 ng/ml. Stężenia te odpowiadają prostoliniowemu odcinkowi krzywej kalibracyjnej MMP-1. Na wolne pola nakłada się próbki osocza i moczu pochodzących od zdrowych ludzi jak i pacjentów z nowotworem pęcherza.
Nierozcieńczane próbki moczu od osób zdrowych i pięciokrotnie rozcieńczone próbki pochodzące od pacjentów z nowotworem pęcherza, nanoszone są na powierzchnie sensora na 10 minut. Objętość przenoszonego roztworu wynosi ok. 3 pl. Powierzchnia sensora jest przemywana co najmniej sześciokrotnie buforem HBS-ES oraz wodą. Mierzy się sygnał SPRI. Stężenie MMP-1 odczytuje się z krzywej kalibracyjnej.
P r z y k ł a d
Oznaczenie stężenia MMP-1 w próbkach osocza i moczu pochodzące od pacjentów z nowotworem pęcherza i od osób zdrowych.
Z próbek moczu od pacjentów z nowotworem pęcherza otrzymano następujące wartości intensywności sygnału SPRI dla poszczególnych miejsc aktywnych:

Claims (1)
1. Biosensor do oznaczania enzymu metaloproteinazy-1 (MMP-1) składający się ze szkiełka (1) oraz czterech warstw, na które nałożona jest siatka polimeru (6) dzieląca sensor na partycje, przy czym spodnią warstwę biosensora stanowi chrom (3), na który nałożone jest złoto (2) korzystnie o grubości 50 nm, następnie tiolowy linker (4), a biosensor jest znamienny tym, że jego warstwę wierzchnią stanowi królicze, specyficzne, zimmobilizowane przeciwciało przeciwko ludzkiej metaloproteinazie-1 (5).
