Description
Opis wynalazkuDescription of the invention
Przedmiotem wynalazku jest sposób wytwarzania N-(triarylometylo)hydroksylamin przedstawionych wzorem ogólnym 1, w którym A oznacza fenyl. The invention relates to a process for the preparation of N- (triarylmethyl) hydroxylamines represented by the general formula 1 in which A is phenyl.
N-(triarylometylo)hydroksylaminy znajdują szerokie zastosowanie w syntezie organicznej jako równoważniki syntetyczne hydroksylaminy, a zwłaszcza znajdują zastosowanie do syntezy N-(triarylometylo)nitronów, 2-[N-(triarylometylo)hydroksylamino]alkanonitryli, 2-[N-(triarylometyIo)hydroksylamino]alkanoli i ich pochodnych z ortogonalnie zabezpieczonymi grupami funkcyjnymi.N- (triarylmethyl) hydroxylamines are widely used in organic synthesis as synthetic equivalents of hydroxylamine, and especially for the synthesis of N- (triarylmethyl) nitrons, 2- [N- (triarylmethyl) hydroxylamino] alkanonitriles, 2- [N- (triarylmethyl) hydroxylamino] alkanols and their derivatives with orthogonally protected functional groups.
The so far known method of producing N- (triarylmethyl) hydroxylamines consists in reacting anhydrous hydroxylamine with a suitable triarylmethyl halide, for example with triphenylmethyl chloride, in a single-phase system in an organic solvent, most often ether or tetrahydrofuran, resulting in a product concentrate composed of which also includes by-products, mainly N, O-bis (triarylmethylation), which due to similar physicochemical properties are difficult to separate from the main product. Although a method of purifying N- (triarylmethyl) hydroxylamines is known, which consists in precipitation of the hydrochloride, in most cases it leads to the decomposition of the product due to the high lability of the C-N bond in these compounds.
The process according to the invention consists in reacting one mole part of a triarylmethyl halide of the general formula II, in which A has the above-mentioned meanings, with at least one mole part of a hydroxylamine salt in the presence of at least one mole part of a strong base, especially sodium hydroxide or potassium, the reaction is carried out at a temperature of 250-330 K, preferably at a temperature of about 273 K, in a water-immiscible organic solvent, preferably in benzene, toluene or xylene, until the reactants react, and then a solution of N- ( triarylmethyl) hydroxylamine from which the pure N- (triarylmethyl) hydroxylamine is optionally isolated.
The main technical and operational advantage resulting from the application of the method according to the invention is the preparation, with good yield, of N- (triarylmethyl) hydroxylamine solutions of a purity sufficient for direct use as a synthetic N-equivalent of hydroxylamine, from which the product can be isolated in a crystalline form, if necessary. An additional advantage of using the method according to the invention is the unique simplicity of synthesis, because there is no need to use any additional operations and technological processes, such as extraction, chromatography, salt precipitation, distillation and others.
Przedmiot wynalazku przedstawiony jest w przykładach wykonania. The subject matter of the invention is illustrated in the following examples.
NMR (CDCI3, δ [ppm], J [Hz]): 7,32 m (6H, ArH orto, J=nieoznaczono), 7,24 m (6H, ArH meta, J=nieoznaczono), 7,15 m (3H, ArH para, J=nieoznaczono), 4,56 s, poszerzony (2H, NH). Krystaliczną N-(trifenylometylo)hydroksylaminę otrzymuje się przez odparowanie roztworu pod zmniejszonym ciśnieniem z łaźni o temperaturze nie przekraczającej 300 K.Example 1. 27.8 g, 0.10 mol of triphenylmethyl chloride is dissolved in 100 g of toluene, and the solution obtained in this way is added in portions with vigorous stirring to 100 cm 3 of a 2M hydroxylamine solution obtained by dissolving hydroxylamine hydrochloride in water and dropwise addition to 0.20 mol of a 5M solution of sodium hydroxide in water, at a temperature of about 273 K, then continued stirring and kept alkaline by dropwise addition of 5M NaOH (about 2% excess in total), then allowing the reactants to react completely to the same temperature for 24 hours. Then as much diethyl ether is added as needed to dissolve the partially precipitated product, the organic layer is dried with anhydrous sodium sulfate and a solution of N- (triphenylmethyl) hydroxylamine is obtained, the identity of which is confirmed by the NMR spectrum taken from a sample of 0.10 cm 3 which was evaporated under reduced pressure. pressure from a bath temperature below 300 K, then the crystalline residue was dissolved in CDCl3. NMR (CDCl3, δ [ppm], J [Hz]): 7.32 m (6H, ArH ortho, J = not determined), 7.24 m (6H, meta ArH, J = not determined), 7.15 m ( 3H, ArH para, J = undetermined), 4.56 s, broadened (2H, NH). Crystalline N- (triphenylmethyl) hydroxylamine is obtained by evaporating the solution under reduced pressure from a bath temperature not exceeding 300 K.
