HOECHST AKTIENGESELLSCHAFT, a Joint Stock Company existing under the laws of the Federal Republic of Germany, of D-6230 Frankfurt am Main 80, Federal Republic of Germany do hereby declare the invention, for which we pray that a patent may be granted to us, and the method by which it is to be performed, to be particularly described in and by the following statement:-

- I - (Followed by 1A)

r» r r~ o f-

- 1A -

HUJLlflST 'P <11 '81 .ABj'aUl'

Description

~ Pharmaceutical and feedstuff premixes

In livestock farming, in particular intensive livestock farming, premixes play an important role in the adminis-5 tration of pharmaceuticals and feed additives. Premixes are concentrated mixtures of the pharmaceutical, or of the feed additive, with one or more carriers. Before they are administered to the animals, they are extended by the user to the use concentration. To avoid separation of the 10 mixtures, and thus inhomogeneity in the end mixture, during transport and mixing at the user's, the physical properties of the premix must meet high demands. The properties of the premix, mainly flowability, mixability, separation behavior of the mixture, dust count and 15 tendency to caking under pressure are determined by the carrier, or carriers, which generally make up more than 90 % of the premix. Selection of the suitable carrier is therefore of crucial importance for the quality of the premix.

20 The manufacturer of a stable premix will strive to optimize the range of demands with regard to quality, having had the problem that, for example, admixing coarse cereal particles improve the flowability properties, but . adversely affect the stability of the mixture (IFF-Dienst

— 25 No. 221, Institut fur Futtermittelforschung [Institute of Feedstuffs Research], Braunschweig-Thune, January/February 1990).

This is so because separation of the mixture can take place when the carrier is composed of particles of 30 different sizes. This separation process of the mixture is favored the greater the difference in particle size is, since the small particles are capable of falling through the interstices of the large particles. The predominant school of thought therefore proposes to 35 produce premixes of carriers and active substances having

245526

- 2 -

the same small particle size. The tendency of the mixture to separate can be prevented in this manner, however, other negative properties such as reduced flowability and increased dust count, will have to be accepted.

5 Surprisingly, it has been found that coarsely-particulate carriers show no greater separation of the mixture than finely-particulate ones. Compared with premixes with finely-particulate carriers, premixes with coarsely-particulate carriers show markedly better physical 10 properties. The flowability is improved, there is no measurable development of dust, and caking under pressure is reduced.

This means that the use of coarsely-particulate carriers can solve the problems which occur in the manufacture of 15 premixes in a satisfactory manner.

The invention relates to an active compound preparation which comprises one or more active compounds (pharmaceutical additives and feed additives) and coarsely-particulate cereal bran as carrier. This active compound 20 preparation is a pharmaceutical or feedstuff premix.

Moreover, the invention relates to a process for manufacturing this active compound preparation which comprises intimately mixing the active compound and the carrier.

A suitable coarsely-particulate cereal bran is preferably 25 cereal bran having a particle size fraction > 250 ^m of at least 90 %, in particular durum and aestivum wheat middlings bran, such as coarsely-particulate wheat middlings bran WG 12.

The term active compound embraces pharmaceutical addi-30 tives and feed additives. These are known in principle and are described, for example, in the Arzneimittelgesetz ,r. [Pharmaceutical Act] (§2) and in the Futtermittelgesej^t*1 °' vV

[Feedstuffs Act] (§2). ^

r ^

*

- 3 -

245526

compounds which can be added to the carrier are, for example, coccidiostats, anthelmintics, antibiotics, minerals, vitamins and performance promoters. Performance promoters are to be understood as meaning those compounds which affect the growth, metabolism, body weight, tissue composition and/or feed conversion, for example sulazuril, halofuginon, salinomycin sodium and flavophos-pholipol, in particular sulazuril.

The examples which follow are intended to illustrate the invention, particularly in its preferred embodiments. The invention is furthermore determined by the contents of the patent claims. Unless otherwise indicated, percentages are by weight.

1. Test for separation behavior of the mixture

The separation behavior of the mixture was tested by allowing the homogeneous premix to run along a vibration channel and forming a poured cone. Samples were taken from the tip and the foot of the cone and analyzed. Deviations from the desired value of the homogeneous premix are indicators for separation of the mixture during transport along the vibration channel or during free fall.

In a plough-share mixer, 99 parts of carrier were mixed with one part of active compound (sulazuril). After 5, 10 and 15 minutes, samples were taken from different locations of the mixer and analyzed.

245526

- 4 -

Content of active compound sulazuril (INN):

10

Sampling site

5 min

10 min

15 min

a b

a b

a b

1

0.98

0.99

1.00

1.01

0.99

1.01

2

0.99

0.99

0.99

1.00

0.99

0.99

3

1.00

1.01

0.99

1.01

0.99

1.00

a) Fine wheat middlings bran (70 % < 250 nm)

b) WG 12

15

20

Separation of the mixture of the transport along vibration channel:

Vibration channel: length 45 cm, pouring height: 15 cm Active compound content: Fine wheat middlings bran WG 12

Tip of the cone Foot of the cone

1.03 1.02

1.00 0.99

Separation of the mixture after tamping: 250 ml tamping cylinder, 1250 strokes

25

Active compound

Surface

Bottom content Fine wheat middlings bran WG 12

1.03 1.01

1.02 0.99

Neither during transport along the vibration channel nor during tamping, does the use of coarse wheat middlings bran result in separation of the premix.

2.
