Abstract
Fremgangsmåte for fremstilling av en renset dimer av en benavledet faktor, som omfatter underkastelse av et inklusjonslegeme av en benavledet faktor fremstilt ved hjelp av genetisk konstruksjon for følgende trinn a) til. e) i rekkefølge; a) trinnet med behandling av et inklusjonslegeme av en benavledet faktor med et denaturerende middel for å fremstille en oppløst monomer; b) trinnet med behandling av den oppløste monomer med en refoldende oppløsning for å fremstille en dimer; c) trinnet med underkastelse av den refoldende dimer for ultrafiltrering og utbytting av oppløsningsmiddel; d) trinnet med underkastelse av dimeroppløsningen fremstilt ovenfor for isoelektrisk utfelling; og e) trinnet med underkastelse av den isoelektrisk utfelte dimer for revers-fasekromatografi.A process for the preparation of a purified dimer of a bone-derived factor, which comprises subjecting an inclusion body to a bone-derived factor produced by genetic engineering for the following steps a) to. e) in order; a) the step of treating an inclusion body of a bone-derived factor with a denaturing agent to produce a dissolved monomer; b) the step of treating the dissolved monomer with a refolding solution to produce a dimer; c) the step of subjecting the refolding dimer to ultrafiltration and solvent exchange; d) the step of subjecting the dimer solution prepared above for isoelectric precipitation; and e) the step of subjecting the isoelectrically precipitated dimer for reverse phase chromatography.
