Abstract
Denne metoden gjelder nye prinsipper og fremgangsmåter for identifisering av smaksspesifikke gener, herunder gener involvert i oppfatningen av saltsmak, spesielt oppfatningen av saltsmak hos mennesker, men også gener involvert i oppfatningen av søt, bitter, umami og sursmak, og gener involvert i andre smakscelle- eller smaksreseptorrelaterte aktiviteter, som f.eks. fordøyelsesfunksjon og fordøyelsesrelaterte sykdommer, smakscelleomsetning, immunregulering i munnhule og fordøyelseskanal og stoffskifteregulering som f.eks. ved diabetes og sykelig overvekt, gener som identifiseres ved disse metodene og analyser for identifisering av smaksmodulatorer (forsterkere eller blokkerere) og potensielle terapeutiske midler basert på disse genene. Disse forbindelsene kan potensielt anvendes i modulering (forsterking eller blokkering) av smaksoppfatning, spesielt oppfatningen av saltsmak og som potensielle terapeutiske midler. I tillegg gjelder denne oppfinnelsen nye metoder for identifisering av smaksspesifikke gener som kan brukes som markører for forskjellige smakscelletyper, herunder celler for søt, bitter, umami, sur, saltsmak og andre smaker hos pattedyr samt analyser som måler aktiviteten i den søte, bitre, umami eller sure reseptoren i nærvær av disse genene for å identifisere modulatorer av søt, bitter, umami og sur smak og for å identifisere terapeutiske midler spesifikt for behandling av fordøyelses- eller metabolismerelaterte sykdommer, smakstap og munninfeksjoner. Oppfinnelsen gir også spesifikke metoder for rensing, anrikning, isolering eller merking av ønskede smakscellesubtyper eller -avstamninger som f.eks. søt, umami, bitter, salt, sur, fet eller stamceller osv, f.eks. ved bruk av FACS, magnetiske perler eller andre seleksjonsmetoder som renser, anriker, merker eller eliminerer som f.eks. ved bruk av merkede cytotoksinceller som uttrykker eller ikke uttrykker ett eller flere smaksspesifikke gener. 1
