Abstract
En karotenogen biosyntetisk genklynge er blitt isolert fra Pantoea stewartii stamme DC413, hvor den genetiske organiseringen av klyngen er crtE-idi-crtX-crtY-crtI-crtB-crtZ. Genene som finnes innen denne klyngen koder for geranylgeranyl pyrofosfat (GGPP) syntetase (CrtE), isopentenylpyrofosfat isomerase (Idi), zeaxantin glukosyl transferase (CrtX), lysopen syklase (CrtY), fytoen desaturase (CrtI), fytoen syntase (CrtB), og (-karoten hydroksylase (CrtZ). Genklyngen, genene og deres produkter er nyttige for omdannelsen av farnesyl pyrofosfat til karotenoider. Vektorer som inneholder disse DNA-segmenter, vertceller som inneholder vektorene og fremgangsmåter for å produsere disse enzymer ved rekombinant DNA teknologi i transformerte vertsorganismer blir utgreiet.A carotenogenic biosynthetic gene cluster has been isolated from Pantoea stewartii strain DC413, where the genetic organization of the cluster is crtE-idi-crtX-crtY-crtI-crtB-crtZ. The genes found within this cluster encode geranylgeranyl pyrophosphate (GGPP) synthetase (CrtE), isopentenyl pyrophosphate isomerase (Idi), zeaxanthin glucosyl transferase (CrtX), lysopenic cyclase (CrtY), phytoene desaturase (CrtI), phyto (-carotene hydroxylase (CrtZ). The gene cluster, genes and their products are useful for the conversion of farnesyl pyrophosphate to carotenoids. Vectors containing these DNA segments, host cells containing the vectors and methods for producing these enzymes by recombinant DNA technology in transformed host organisms being paid off.
