Foreliggende oppfinnelse vedrører enzymholdige vaskemiddelblandinger, nærmere bestemt blandinger som omfatter lipase. The present invention relates to enzyme-containing detergent mixtures, more specifically mixtures comprising lipase.
Vaskemiddelblandinger som omfatter lipase er kjent. F.eks. åpenbarer GB-patent nr. 1.372.034 lipase fra Pseudomonas i spesi-fikke ikke-ionisk-holdige vaskemiddelblandinger for bløtlegging av tekstiler. Detergent compositions comprising lipase are known. E.g. GB Patent No. 1,372,034 discloses lipase from Pseudomonas in specific non-ionic detergent compositions for soaking textiles.
USP 3,950,277 also describes compositions for soaking textiles. The described mixtures include lipase and lipase activators, and several types of lipase extracted from microorganisms and other sources are mentioned. Those mentioned as particularly preferred are Amano CE, Amano M-AP, Takeda 1969-4-9, and Meito MY-30 lipases, but there is no indication of the form in which the lipase is to be processed or used.
USP 4.011.169 (NL 74 08763) beskriver anvendelse av en lignende type av enzymer for fremstilling av additiver for vaskemidler (vaskemiddelblandinger). USP 4,011,169 (NL 74 08763) describes the use of a similar type of enzymes for the production of additives for detergents (detergent mixtures).

EP 258 068 gives a detailed description of lipases extracted from organisms of the strain Thermomyces/Humicola, and certain uses thereof.
It is also assumed that in certain areas a lipase-containing, granular detergent mixture containing approx. 37% detergent active substances including 5% non-ionic detergent, and where the rest is essentially an anionic detergent, approx.
16% zeolitt, ca. 60 LU/g lipase, pluss protease og andre vanlige vaskemiddel-additiver. 16% zeolite, approx. 60 LU/g lipase, plus protease and other common detergent additives.
Further examples of known lipase-containing detergent mixtures can be found in JA application no. 63-078000 (1988) which discloses the properties and use of Pseudomonas lipase, including use in a lipase-containing system based on 10-40% surfactant (e.g. eg sodium C14-C18-α-olefin sulphonate), as well as other conventional detergent ingredients.
Teknikkens stand inkluderer også EP-patentsøknad, publ. The state of the art also includes EP patent application, publ.
nr. 268 456, som i forbindelse med tabell 10(b) viser en eksperi-mentell vaskeløsning som inneholder lipase og ca. 1 mikrogram/ml natriumdodecylsulfat. no. 268 456, which in connection with table 10(b) shows an experimental washing solution containing lipase and approx. 1 microgram/ml sodium dodecyl sulfate.
The state of the art also includes Research Disclosure 29056, June 1988, which discloses lipase in connection with the detergent compositions according to EP 179 533.
One problem in connection with the lipase-containing detergent compositions according to the state of the art is that there is a strong tendency for many detergent-active materials to inhibit the action of lipase.
It is an aim of the present invention to provide detergent compositions in which the activity of the added lipase is relatively less inhibited.
I henhold til foreliggende oppfinnelse tilveiebringes det en enzymholdig vaskemiddelblanding for vasking av tekstiler, idet blandingen er karakterisert ved at den omfatter: According to the present invention, an enzyme-containing detergent mixture is provided for washing textiles, the mixture being characterized in that it comprises:
1) 1-60 vekt% av et overflateaktivt system bestående av: 1) 1-60% by weight of a surfactant system consisting of:
a) minst 50 vekt% et primært alkoholsulfat med en kjedelengde i området C12 til ci8 °9 b) 0-50 vekt% av et overflateaktivt middel valgt fra gruppen bestående av andre anionisk overflateaktive a) at least 50% by weight of a primary alcohol sulfate with a chain length in the range C12 to c18 °9 b) 0-50% by weight of a surfactant selected from the group consisting of other anionic surfactants
midler enn primære alkoholsulfater, ikke-ionisk overflateaktive midler, kationiske overflateaktive agents than primary alcohol sulphates, non-ionic surfactants, cationic surfactants
midler og blandinger derav; og agents and mixtures thereof; and
2) lipase enzyme in an amount in the range of 50 to 30,000 (LU) lipase units per grams of the surface-active system or of the detergent mixture, together with usual detergent auxiliaries.
In the present description, lipase units are defined as they are defined in EP 258 068.
In the most practical embodiments, the ingredients in the detergent mixtures also include e.g. conventional detergent additives such as e.g. one or more of those mentioned below.
It has been discovered that in such systems as described here, the lipase enzyme shows surprisingly little inhibition. One reason for this surprisingly good performance of lipase in this system is that at least one member of the class of primary alcohol sulfates, i.e. sodium dodecyl sulfate, is well known and used in laboratory studies as a denaturant for proteins, including enzymes. When compared with a lipase-containing surfactant based on α-olefin sulfonate, the compositions according to the present invention show stronger detergent activity combined with suitably low levels of lipase inhibition.
It is also surprising that the (known) building effect of the primary alcohol sulphate and its high calcium affinity when compared with the highly inhibitory LAS surfactants does not lead to any greater degree of lipase inhibition.
The primary alcohol sulphates used in these mixtures can e.g. be based on coconut-primary-alcohol-sulfates or tallow-primary-alcohol-sulfates or intermixtures of these and/or analogues having alkyl chains originating from other sources.
What is also allowed in the mixtures are proportions of other surface-active types, especially anionic, non-ionic or cationic detergents, in small or smaller quantities, e.g. up to 10%, possibly up to 20% or up to 35%, sometimes up to 50%, of the surfactant system, e.g. linear alkylbenzenesulfonates (LAS) or (branched chain) alkylbenzenesulfonates (ABS), which alone have been found to show an unacceptably high degree of lipase inhibition.
In certain examples of the mixtures according to the invention, e.g. particularly such as are illustrated in the following examples 11-13, the alkylbenzene sulphonates (LAS or ABS) may be present in an amount of up to approx. 66%, or in some cases up to approx. 70% or even 75%, calculated on the surface-active system, including the primary alcohol-sulphate.
E.g. some of the compositions may comprise a surface-active system which essentially consists of a mixture of PAS with ABS and/or LAS. The proportion of ABS or LAS in such a mixture can e.g. be up to approx. 75%, e.g. up to approx. 70%, often somewhat lower than e.g. about. 66% or 60%, by weight.
A preferred surfactant system for use in the present invention is based on a mixture (e.g. approx. 1:1 w/w) of <C>12<-C>14-coco-alkyl-0<3 <C> 16<-C>18 tallow alkyl sulfates.
The lipolytic enzyme can usefully be added in the form of a granular mixture of lipolytic enzyme with carrier material.
Den tilsatte mengde av lipolytisk enzym kan velges innen vide grenser, nemlig 50 til 30.000 LU/g av granulær vaskemiddelblanding, f.eks. ofte minst 100 LU/g, meget nyttig minst 500 LU/g, noen ganger fortrinnsvis over 1000, over 2000 LU/g eller over 4000 LU/g eller mer, således meget ofte innen området 50-4000 LU/g og muligens innen området 200-1000 LU/g. The added amount of lipolytic enzyme can be chosen within wide limits, namely 50 to 30,000 LU/g of granular detergent mixture, e.g. often at least 100 LU/g, very useful at least 500 LU/g, sometimes preferably above 1000, above 2000 LU/g or above 4000 LU/g or more, thus very often within the range 50-4000 LU/g and possibly within the range 200-1000 LU/g.
Spesielt egnet er f.eks. de følgende, kommersielt tilgjengelige lipase-preparater: Novo Lipolase, The lipolytic enzyme can be selected from a large selection of lipases, especially the lipases described in e.g. the following EP patents: 214 761, 258 068 and in particular lipases which show immunological cross-reactivity with antisera against lipase from Thermomyces lanuginosus ATCC 22070, EP 205 208 and 206 390), and in particular lipases which show immunological cross-reactivity with antisera against lipase from Chromobacter viscosum was lipolyticum NRRL B-3673, or against lipase from Alcaligenes PL-679, ATCC 31371 and FERM-P 3783, and also the lipases described in the publications WO 87/00859 and EP 204 284. Particularly suitable are e.g. the following commercially available lipase preparations: Novo Lipolase,
Amano lipases CE, P, B, AP, M-AP, AML og CES, og Meito lipases MY-30, OF og PL, også esterase MM, Lipozym, SP225, SP285, Saiken lipase, Enzeco lipase, Toyo Jozo lipase og Diosynth lipase (varemerker). Amano lipases CE, P, B, AP, M-AP, AML and CES, and Meito lipases MY-30, OF and PL, also esterase MM, Lipozym, SP225, SP285, Saiken lipase, Enzeco lipase, Toyo Jozo lipase and Diosynth lipase (trademarks).
Genetic modification of the enzymes can be achieved by extracting a suitable lipase gene, e.g. the gene for lipase from Thermomyces lanuginosus or from a mutant thereof, and introduction and expression of the gene or its derivative in a suitable producer organism such as e.g. an Aspergillus. The techniques described in WO 88/02775 and EP patent applications, publ. Nos. 243 338 and 268 452 can be used and adapted.

Particularly suitable are e.g. the washing processes, which are carried out at low temperatures of approx. 60-65°C or lower, e.g. about. 30-35°C or lower.
Oppfinnelsen er ytterligere illustrert ved de følgende The invention is further illustrated by the following
eksempler, som også viser ytelsen i form av reflektans ved 460 nm på test-tøystykker tilsmusset med lipid og deretter vasket, etter 5 vaskeomganger med flere forskjellige lipase-holdige vaskemiddelblandinger. examples, which also show the performance in terms of reflectance at 460 nm on test cloths soiled with lipid and then washed, after 5 washing cycles with several different lipase-containing detergent mixtures.
Eksempler 1- 5 Examples 1-5
In each example, a detergent mixture was used to wash a test piece of cotton soiled with a fat mixture based on olive oil (up to 10-15% by weight calculated on the test piece) after three pre-washes. The detergent compositions used corresponded to that detailed above, except that different surfactant systems were used (each in an amount of 9% by weight) as follows:
Eks. 1 Kokos-alkylsulfat (Elfan 280 fra Akzo) Ex. 1 Coconut alkyl sulfate (Elfan 280 from Akzo)
Eks. 2 Talg-alkylsulfat (Sulphopon T55 fra Henkel) Ex. 2 Tallow alkyl sulphate (Sulphopon T55 from Henkel)
Eks. 3 50:50 kokos-alkyl-/talg-alkylsulfater Ex. 3 50:50 coconut alkyl/tallow alkyl sulfates
Eks. 4 25:25:50 kokos-alkyl-talg-alkyl-DOB113 Ex. 4 25:25:50 coconut-alkyl-tallow-alkyl-DOB113
Eks. 5 DOB113 (lineært alkylbenzensulfonat alene, for Ex. 5 DOB113 (linear alkylbenzene sulfonate alone, for
sammenligning). comparison).
I ovennevnte blanding betyr "D0B113" sulfoneringsproduktet av Shell Alkylbenzen DOB113.<®>In the above mixture "D0B113" means the sulfonation product of Shell Alkylbenzene DOB113.<®>
Eksemplene 6-10 tilsvarer eksemplene 1-5, bortsett fra vaske-temperaturen. Examples 6-10 correspond to examples 1-5, except for the wash temperature.
In each example, isothermal washing was carried out at 30°C or 60°C for 30 minutes with and without lipase enzyme, with pre-dissolved detergent mixtures used in an amount of 4 g/l. The ratio cloth/washing water was uniformly 10:1, containing 50:50 soiled: unsoiled quantity.
In each experiment, one of two lipases was used: Lipolase from Novo as specified above, or lipase from Pseudomonas gladioli, used in the form of a conventional purified culture preparation (600 LU/mg) with a lipase activity equivalent to that found in Lipolase.
The results are reproduced as average reflectance at 4 60 nm on 5 pieces of cloth after 5 soiling/washing cycles.
Examples 1 to 5 were carried out at 60°C, Examples 6 to 10 were carried out at 30°C.
The results show an increased benefit due to the lipolytic activity. The general trend is to show that lipase in the presence of an LAS-type detergent gives a significantly smaller advantage (ex. 5, 10) than the average for the other examples (primary alcohol sulfates and mixtures thereof), which are similarly preferred for application.
Eksemplene 11 til 13 Examples 11 to 13
The following detergent compositions in the form of bars very well support the lipolytic action of lipase.
Stoffer i små mengder og vann opptil 100% i hvert tilfelle. Substances in small quantities and water up to 100% in each case.
The mixtures are composed in a conventional manner for detergents in the form of bars.
