Description
Fremgangsmåte ved fremstilling av Procedure in the manufacture of
2-benzimidazolcarbaminsyreestere. 2-benzimidazolcarbamic acid esters.
The present invention relates to a new process for the production of alkyl esters of certain benzimidazolecarbamic acids, especially from cyanamide or cyanamide salts.
Alkylestere av 2-benzimidazolcarbaminsyrer av den generelle formel: Alkyl esters of 2-benzimidazolcarbamic acids of the general formula:
hvor R er alkyl med 1-4 carbonatomer, og X er hydrogen, halogen, where R is alkyl with 1-4 carbon atoms, and X is hydrogen, halogen,
nitro eller alkyl med 1-4 carbonatomer, er nyttige som fungi-nitro or alkyl of 1-4 carbon atoms, are useful as fungi-
cider. The compounds where X is hydrogen and R is alkyl with 1-4
The above compounds are also useful as starting materials in the preparation of dialkyl esters of l-carboxy-2-benzimidazolcarbamic acids of the general formula:
hvor X er som ovenfor angitt, og R og R^, som er like eller for-skjellige, er alkyl med 1-4 carbonatomer. where X is as indicated above, and R and R, which are the same or different, are alkyl with 1-4 carbon atoms.
Disse forbindelser er omtalt i U.S. patent nr. 2.933.504 ogThese compounds are disclosed in U.S. Pat. patent no. 2,933,504 and
er meget nyttige som fungicider.are very useful as fungicides.
U.S. patent no. 3-010,968 describes a method for the preparation of 2-benzimidazolcarbamic acid esters of formula I by reacting thiourea with dimethylsulphate to form 2-methylthiopseudo-ureasulphate. This reaction product is then reacted with an alkyl chloroformate and a base to form an acylated 2-methylthiopseudourea which is then further reacted with an o-phenylenediamine in the presence of a protonic acid to form the desired product.
The present method has several advantages over the above-mentioned method according to the patent. A particular advantage is that commercially available, cheap, cyanamide salts or cyanamide solutions of technical quality can be used instead of the relatively more expensive thiourea materials. Furthermore, the present method does not require the handling or disposal of unpleasant methyl mercaptan materials which are by-products of this type of process. Furthermore, the present method can be carried out batchwise or continuously with ordinary reaction vessels.
From Belgian patent 666,795 it is known to prepare compounds of formula I by reacting cyanamide with an alkyl chloroformate in a suitable organic solvent, and then adding o-phenylenediamine to obtain the product. The present method differs from this, firstly, in that the reaction is carried out in an aqueous medium, which is of course cheaper, but more importantly, in the second step of the present method, a regulated pH is used. These differences mean that with the present method significantly higher yields are obtained than according to the Belgian patent.
According to the present invention, the compounds of formula I are prepared in two steps by first reacting cyanamide or a cyanamide salt with an alkyl chloroformate, after which the reaction product is reacted in the second step with an o-phenylenediamine, and the method is characterized by the fact that the reaction in the first step takes place in a the substantially neutral to basic aqueous medium at a temperature between 0° and 105°C to form an alkyl cyanocarbamate salt of the formula: where R is as above, M is an alkali metal or an alkaline earth metal, and m is the valence of M, as in second step is reacted with an o-phenylenediamine of the general formula:
hvor X er som ovenfor angitt, i et surt vandig medium ved en temperatur på 40 - 130°C inntil felningen av den ønskede 2-benzimidazol-carbaminsyreester er fullstendig, hvorefter det dannede produkt utvinnes fra reaksjonsblandingen. where X is as stated above, in an acidic aqueous medium at a temperature of 40 - 130°C until the precipitation of the desired 2-benzimidazole-carbamic acid ester is complete, after which the formed product is recovered from the reaction mixture.
Fremgangsmåten ifølge oppfinnelsen kan oppsummeres ved følg-ende ligninger: The method according to the invention can be summarized by the following equations:
(Trinn 1)(Step 1)
hvor R er alkyl med 1-4 carbonatomer, X er hydrogen, halogen, nitro eller alkyl med 1-4 carbonatomer, M er et alkalimetall eller et jordalkalimetall, og m er valensen av M, og H+ kan være avledet av en hvilken som helst syre. where R is alkyl of 1-4 carbon atoms, X is hydrogen, halogen, nitro or alkyl of 1-4 carbon atoms, M is an alkali metal or an alkaline earth metal, and m is the valence of M, and H+ may be derived from any acid.
Trinn 1 ved denne utførelsesform illustreres av følgende lign-ing: In step 1 above, when a cyanamide salt is used, the cyanamide is present in the aqueous solution in anionic form. This ionization is in itself sufficient to give the desired pH. However, cyanamide as an aqueous solution can also be used in step 1 if a base is added to the solution to maintain the desired pH. Step 1 in this embodiment is illustrated by the following equation:
In this reaction, any basic alkali metal or alkaline earth metal compound can be used as a base. Alkali metal and alkaline earth metal hydroxides are preferred, although oxides and carbonates can be used.
Denne overføringsreaksjon oppsummeres av ligningen: The 2-benzimidazolcarbamic acid alkyl esters produced by the present process can be transferred to the esters shown in formula II. This transfer reaction is summarized by the equation:
hvor X, R og R^er som ovenfor angitt. where X, R and R^ are as indicated above.
In the first step of the present process, an appropriate alkyl chloroformate is reacted with either cyanamide or a salt of cyanamide in substantially neutral to basic aqueous media to form a salt of an alkyl cyanocarbamate. Readily available salts of technical grade are suitable for this procedure. Suitable starting materials are cyanamide alkaline earth or alkaline metal salts, especially the salts with magnesium, potassium, sodium or calcium. Commercially available calcium cyanamide is the preferred starting material. Cyanamid even in aqueous solution is also a preferred starting material.
M0 / NCN, kan anvendes, imidlertid foretrekkes dobbel-<s/m In the first step, when the alkyl chloroformate is reacted with a cyanamide salt, either a single salt, i.e. M(NHCN) m, or a double salt, i.e. M0 / NCN, can be used, however, double-<s/m is preferred
cyanamidsaltet for omsetning med alkylklorformiat.the cyanamide salt for reaction with alkyl chloroformate.
Det første trinn utføres i vann. The first step is carried out in water.
The concentrations of the starting materials in the reaction mixture are not critical, however, for economic reasons, usually high concentrations are preferred. The concentration of the cyanamide in the aqueous solution can vary from 5 to 50%, and as cyanamide is currently commercially available as a 50% aqueous solution, such solutions are preferred. With respect to the suspension or solution of the cyanamide salts, the concentration is limited only by the handling characteristics of the suspension.
When a cyanamide salt is used, it is preferred to add the alkyl chloroformate with mixing to the aqueous suspension or stirring containing the cyanamide salt. Reduced alkyl chloroformate consumption is obtained when the pH of the cyanamide salt suspension in water is first lowered from approx. pH 12 to pH 8 - 10 by adding mineral acid before the alkyl chloroformate is added. When a cyanamide-water solution is used, the alkyl chloroformate and base can be added simultaneously to the cyanamide solution. During this simultaneous addition, preferably the molar amount of base added at any one time should be equivalent or slightly in excess of the molar amount of chloroformate.
In the first step, the pH should be maintained at largely neutral or basic conditions, i.e. within the range from 6 to 13. As previously mentioned, when free cyanamide is used, this pH range can be maintained by adding base as needed. When an aqueous cyanamide solution is used, the preferred pH range is from 7 to 9. With cyanamide salts, however, it is preferred to maintain the pH between 8 and 10.
The temperature in this first reaction step is not critical within the range 0 - 105°C, in general the range 25 - 6o°C is preferred. If desired, this step can be performed at higher temperatures under pressure.
The reaction is fast, and the product is stable in the reaction medium, and the reaction time is thus not critical and the product can be used directly in the second step or stored for a time, depending on what is desired from an equipment point of view. in general, the reaction time will depend on the heat transfer rate and can be from 5 minutes to 2 hours.
When cyanamide salts of technical quality have been used for the first step, sometimes insoluble impurities and salts, e.g. calcium salts, present in the reaction mixture. It is then preferable to filter out these contaminants before proceeding to step 2.
In the second reaction step, o-phenylenediamine is added to the reaction product from step 1, or if cyanamide salts have been used, to the filtered reaction product, in an aqueous acidic medium to form the 2-benzimidazolcarbamic acid alkyl ester. The o-phenylenediamine can be added to the reaction product while the product is at the temperature it had during the first reaction step, 0 - 105°C.
In order to carry out the condensation and ring closure, in the second reaction step the pH should be kept in the range 1-5", preferably 2.5 - 4.5 by adding acid as needed. The pH can be kept at the desired value by adding any acid, e.g. formic acid, acetic acid, hydrochloric acid, sulfuric acid, phosphoric acid, hydroxyacetic acid, sulfamic acid and the like. Optionally, the o-phenylenediamine can be in the form of a mineral acid salt or can be added as a solution in water. When o-phenylenediamine salts are used, the amount of acids required to give the desired pH will be correspondingly lower.
Den annen reaksjon vil foregå ved temperaturer over 40°C,The second reaction will take place at temperatures above 40°C,
Hvis dette gjøres, kan temperaturen gå opp til 130°C. and it is preferred to heat during this step to accelerate the complete reaction. The heat input is important as the reaction in the second step would be too slow at low temperatures. The reaction mixture should be maintained at from 40°C to the reflux temperature, preferably 6o - 105°C. The second reaction step can also be carried out under pressure if desired. If this is done, the temperature can rise to 130°C.
During the heating step, the desired product will coalesce. The end of the precipitation is thus a sign that the reaction mixture has been heated for a sufficient time for the reaction to be complete. The time is not critical, and depends on temperature and pH. At a pH of 2.5 - 4.5 and a temperature between 70 and 105°C, the reaction time can thus be 5-30 minutes. When lower temperatures are used, the time will be longer, and if the pH is not kept in the range of 1-5, the reaction will be very slow or not take place at all.
The desired product can be recovered in any conventional manner, e.g. spray drying, filtering or centrifuging, or it can be transferred to another liquid medium by de-distillation of the water.
In the overall reaction, the reactants can be used in the molar equivalent ratios indicated in the following table:
The molar concentrations are not critical at the upper limit, but it will not be practical or economical to use larger amounts. It is obvious that the concentration in the second step will depend on the concentration in the first step.
Denne reaksjon illustreres av følgende ligninger: It has also been shown that the first step of this reaction has yielded a new compound, methylcyanocarbamate. This compound can be prepared by reacting methyl chloroformate with a cyanamide salt and then acidifying the resulting solution, or reacting the methyl chloroformate with cyanamide in the presence of a base, such as sodium hydroxide, and then acidifying the resulting solution. This reaction is illustrated by the following equations:
The methyl cyanocarbamate thus formed can be isolated by extraction with organic solvents if desired, however the above solution containing the methyl cyanocarbamate can be used in the second step of the present method. The product, methylcyanocarbamate, is therefore a useful compound in that it can be reacted with o-phenylenediamine to form 2-benzimidazole-
carbaminsyre-methylester.carbamic acid methyl ester.
As previously mentioned, the present method gives a better yield than the method according to Belgian patent 666,795 - Thus, according to the Belgian patent, in example 4 a crude yield of 13"7% and a yield of 7>3% purified product is obtained, in example 6 a crude yield of 24.4% with 11.3% purified product and in example 7 a yield of 1%. With the present method, yields of 82 - 93% and higher are obtained.
The following experiments show that carrying out the second step at a pH of 1-5 is important.
